Attenuation of dextran sodium sulphate induced colitis in matrix metalloproteinase-9 deficient mice

AIM: To study whether matrix metalloproteinase-9 (MMP-9) is a key factor in epithelial damage in the dextran sodium sulphate (DSS) model of colitis in ***: MMP-9-deficient and wild-type (wt) mice were given 5% DSS in drinking water for 5 d followed by recovery up to 7 d. On d 5 and 12 after induction of colitis, gelatinases, MMP-2 and MMP-9, were measured in homogenates of colonic tissue by zymography and Western blot, whereas tissue inhibitor of metalloproteinases (TIMPs) were measured by reverse zymography. The gelatinolytic activity was also determined in supernatants of polymorphonuclear leukocytes (PMN) isolated from mice blood. Moreover, intestinal epithelial cells were stimulated with TNF-α to study whether these cells were able to produce MMPs. Finally, colonic mucosal lesions were measured by microscopic examination. RESULTS: On d 5 of colitis, the activity of MMP-9 was increased in homogenates of colonic tissues (0.24 ± 0.1 vs 21.3 ± 6.4, P < 0.05) and PMN from peripheral blood in wt (0.5 ± 0.1 vs 10.4 ± 0.7, P < 0.05), but not in MMP-9-deficient animals. The MMP-9 activity was also up-regulated by TNF-α in epithelial intestinal cells (2.5 ± 0.5 vs 14.7 ± 3.0, P < 0.05). Although colitis also led to increase of TIMP-1 activity, the MMP-9/TIMP-1 balance remained elevated. Finally, in the MMP-9-deficient colitic mice both the extent and severity of intestinal epithelialinjury were significantly attenuated when compared with wt mice. CONCLUSION: We conclude that DSS induced colitis is markedly attenuated in animals lacking MMP-9. This suggests that intestinal injury induced by DSS is modu-lated by MMP-9 and that inhibition of this gelatinase may reduce inflammation.

Leprosy is an infectious disease caused by Mycobacterium leprae, transmitted from person to person through contact among susceptible untreated patients. It presents a broad clinical spectrum which is related to the host’s ability to mount a specific immune response. The lesions caused by the proliferation of Mycobacterium leprae (M. leprae) were significantly reduced in recent years with the early detection of new cases. Because they are less evident and/or study, maxillofacial injuries and the oral mucosa may reveal important details about the transmissibility and immunopathogenesis of leprosy. This article was based on a literature to verify an interrelation between oral manifestations in virchowian patients and immuno-pathological factors. Association between the infection of oral mucosa and some pathological findings as well as the participation of the local immune response in protection against the disease are research topics still not fully exploited.

MM： To investigate whether, under the influence of pol ypectomy, the incidence of adenoma decreases with age.METHODS： Consecutive patients with colonic adenomas identified at index colonoscopy were retrospectively selected if they had undergone three or more complete colonoscopies, at least 24 mo apart. Patients who had any first-degree relative with colorectal cancer were excluded. Data regarding number of adenomas at each colonoscopy, their location, size and histological classification were recorded. The monthly incidence density of adenomas after the index examination was estimated for the study population, by using the person-years method. Baseline adenomas were excluded from incidence calculations but their characteristics were correlated with recurrence at follow-up, using the X^42 test.RESULTS： One hundred and fifty-six patients were included （109 male, mean age at index colonoscopy 56.8 ± 10.3 years）, with follow-up that ranged from 48 to 232 mo. No significant correlations were observed between the number, the presence of villous component, or the size of adenomas at index colonoscopy and the presence of adenomas at subsequent colonoscopies （P = 0.49, 0.12 and 0.78, respectively）. The incidence of colonic adenomas was observed to decay from 1.4% person-months at the beginning of the study to values close to 0%, at 12 years after index colonoscopy.CONCLUSION： Our results suggest the sporadic formation of adenomas occurs within a discrete period and that, when these adenomas are removed, all neoplasia-prone clones may be extinguished.

Brazil has around 8.000 kilometers of coastal line,covering a diversity of ecological systems such as estuaries,lagoons,coral reefs,mangroves, rocky and sandy *** fishing is practiced in a variety of ecosystems that greatly influence the way fishing activities are *** currents,winds,tides,waves,coastal vegetation,fauna and flora and particularly ecological cycles are important elements that are taken into consideration by artisanal fishermen in order to organise their fishing *** have a deep traditional knowledge of these ecosystems and have developed different management *** traditional knowledge and management are at stake as many of these ecosystems are affected by increasing urbanization,industrialization and *** fishermen belong to different ***,in the southern coast of the country,there are artisanal fishermen,who are descendants of the Azorian migrants who developed techniques and systems suited to a jagged coastline,with many islands,bays and *** the southeastern region are the “caicaras”,descendants of the Portuguese colonisers,natives and blacks who combine small-scale fishing with *** the northeast the raR fishermen(jangadeiros)who use a specialised raft adapted to beaches without piers,known as the“jangada”.In the northern end are the“praieiros”fishers who developed a large variety of boats suited especially to a coast with fluctuating tide *** long time these 800.000 artisanat fishermen have been excluded from Government policies that have benefited mainly industrial *** recently,the Government has established a specific ministry to deal with fisheries and aquaculture,and artisanal fishermen got more visibility as food producers,as they are responsible for over 60% of total *** are also more socially organized,establishing their own social movements and institutions that are pressing for broader changes in public policies.

Cerium Separation from Light Rare Earth Concentrate by Liquid-Liquid Extraction

This work presents an investigation of solvent extraction parameters in order to obtain high purity cerium from a mixture containing other rare earths elements, as an alternative to oxidation and selective precipitation or dissolution. The study was carried out using a sample of sulfuric liquor obtained from the leaching of monazite rich in light rare earth elements (La, Ce, Pr, Nd) provided by INB (Indústrias Nucleares do Brasil S.A.). Experiments were carried out in chloridric, nitric and sulfuric media. The nitric and hydrochloric solutions were prepared by precipitation of the REE from the sulfuric liquor as rare earths oxalate, calcining the precipitate and dissolving it with nitric or hydrochloric acid. The separation of cerium was investigated in its trivalent and tetravalent forms. The parameters investigated were: type and concentration of extractant, type and amount of the oxidizing agent, liquor acidity, and volumetric ratio between organic and aqueous phases. The preference of the organic phase to extract cerium in its oxidized form (IV) over the other rare earths elements was confirmed, allowing the obtainment of a high purity cerium solution. The best results were achieved in nitric medium, with cerium in its tetravalent form, using cationic extractants (P507 or D2EHPA) and a mixture of potassium persulfate and silver chloride as oxidizing agents, yielding over 99% of cerium extraction with over 99% of purity.

Pacman Renormalization in Siegel Parameters of Bounded Type

A novel method of renormalization called Pacman renormalization allows us to study (unicritical) Siegel functions through Pacman-type functions. It has been used to investigate the Siegel parameters with combinatorially periodic rotation number in the main cardioid of the Mandelbrot set. It is already known that it can be defined a Pacman renormalization operator such that for Siegel pacmen, with combinatorially periodic rotation numbers, the operator is compact, analytic and has a unique fixed point, at which it is hyperbolic with one-dimensional unstable manifold. In this paper we observe that this Pacman renormalization operator is compact and analytic at any Siegel Pacman or Siegel map with combinatorially bounded rotation number. This allows us to define a renormalization operator on the hybrid classes of the standard Siegel pacmen to which we built its horseshoe where the operator is topologically semiconjugated to the left shift on the space of bi-infinite sequences of natural numbers bounded by some constant.

Ependymomas are a somewhat diverse category of glial tumors that often develop from the lining of the brain’s ventricles, or the spinal cord’s central canal. They make up 5% of all neuroepithelial tumors, 10% of paediatric brain tumors, and up to 33% of brain tumors in children under the age of three. Hydrocephalus is one of the complications, and it can be identified as progressive macrocephaly or increasing head circumference crossing percentiles, nausea, vomiting, poor appetite, irritability, and regression of developmental milestones.

The present study aimed to develop a protocol for somatic embryogenesis and encapsulation of coffee embryos(Coffea arabica L.),for the conservation of genotypes with characteristics of commercial *** embryos were induced from leaf explants in Murashige and Skoog medium(MS)supplemented with 1 mg·L^(−1) of 2,4-dichlorophenoxiacetic acid(2,4-D)combined with 2 mg·L^(−1) of benzyladenine(BA).Somatic embryos(SE)at the globular stage were encapsulated in a sodium alginate matrix;two treatments were tested:MS+5 mg·L^(−1) BA+1 mg·L^(−1) NAA+3%(w/v)alginate,and MS+7 mg·L^(−1) BA+5.7 mg·L^(−1) indoleacetic acid(IAA)+3%(w/v)*** was complexed with 100 mM calcium chloride(CaCl_(2)).Viability of the encapsulated SE was determined by staining with 0.01%fluorescein diacetate(FDA)after 0,15,30,and 45 days of storage at 4℃.Embryo viability was 100%in both treatments.
