The gastrointestinal tract of livestock and poultry is prone to challenge by feedborne antigens,pathogens,and other stress factors in the farm environment.Excessive physiological inflammation and oxidative stress that arises firstly disrupts the intestinal epithelial barrier followed by other components of the gastrointestinal tract.In the present review,the interrelationship between intestinal barrier inflammation and oxidative stress that contributes to the pathogenesis of inflammatory bowel disease was described.Further,the role of naturally existing immunomodulatory nutrients such as the omega-3 polyunsaturated fatty acids,citrus pectin,and milk-derived exosomes in preventing intestinal barrier inflammation was discussed.Based on the existing evidence,the possible molecular mechanism of these bioactive nutrients in the intestinal barrier was outlined for application in animal diets.

Vitamin E Bioavailability: Past and Present Insights

Over the past decades several studies have investigated the importance of an adequate vitamin E status to sustain both animal health and production in dairy cows. Cow milk is considered as a remarkable source of bioactive components promoting human health, which has renewed interest in the effects of vitamin E supplementation on its nutritional value, sensory quality and shelf life. Thus, defining relative bioavailability, utilisation and transfer into milk of different vitamin E formulations is particularly important to assess the adequate levels of supplementation for animal health and milk quality. In nature vitamin E is present under one isomeric form, RRR α-tocopherol;when α-tocopherol is synthesized chemically, a racemic mixture of 8 possible isomers of α-tocopherol in equimolar concentrations is produced (all-rac α-tocopherol). The different stereoisomers have different biopotencies in humans and livestock;the conversion factor between RRR and all-rac vitamin E was estimated by early studies on the basis of the rat foetal resorption bioassay, and then extended to other species. Recent advances on the distribution of vitamin E stereoisomers in plasma and tissues have highlighted the need to formulate new conversion factors in dairy cows as well as in humans. On account of this, the present article aims to consider past and recent data related to vitamin E in dairy cow nutrition.

Tobacco Seeds By-Product as Protein Source for Piglets

Tobacco seed cake is a by-product with interesting characteristics for animal nutrition, due to its high protein content. The focus of this study is to evaluate if tobacco seed cake, administered in feed, can affect principal serum metabolic parameters of weaned piglets in order to establish if it can be used as both a delivery system of edible vaccines and an alternative protein source in piglets diet. A total of 48 weaned piglets were divided in two homogeneous groups for weight, control (CG) and treatment (TG);TG and CG were fed ad libitum with two isoenergetic and isoproteic experimental diets (CP: 17.68% CG and 17.64% TG) differentiated for the inclusion of 4% of tobacco seed cake in the TG diet in replacement of wheat bran and soybean meal. Growth performances were evaluated and feed intake was measured weekly. Blood samples were collected on days 0, 20 and 43 to evaluate hematocrit and principal metabolic parameters, in order to assess the health status of the animals. The administration of tobacco did not impaire health status and growth performance of piglets. The use of bioenergy co-products from non-food crops may represent a good approach creating an integration between biofuel and food production, with consequent benefits for food security and environmental impact.

Escherichia coli diarrhoea is a multifactorial condition which usually occurs during the post-weaning and is responsible for economic losses in pig production. One approach, to evaluate if substitute of antibiotic in vivo is effective in controlling postweaning diarrhea in the gastrointestinal tract ecosystem, is to use an appropriate disease model. However, there are still many criticisms related to the incidence and the severity of the diarrhoea in the experimental conditions. The aim of the study was to set up an Escherichia coli challenge model in order to induce a significant percentage of diarrhoea in weaned piglets for the evaluation of innovative compounds in vivo. A total of 35 piglets, weaned at 33±2 days were randomized into 3 groups: control (CG), infected 1 (IG1) and infected 2 (IG2). One day after arrival piglets of IG1 and IG2 were orally inoculated with 3.7 × 108 CFU of Escherichia coli O149. All piglets were fed a high protein ration for 3 days. Daily health status and faeces were recorded by a point scale individually. Challenge strains in faecal samples were evaluated by polymerase chain reaction, serotyping and biochemical identification. Diarrhoea was observed in 96.67% (58.6% severe;41.4% mild) of all infected piglets and occurred on average 1.3 days after the challenge. The CG group presented one piglet with a transient mild diarrhoea. The E. coli challenge significantly affected the consistency and color of faeces (P E. coli O149, mainly hemolytic (88%), was isolated in 56% of faecal samples and the 70% of piglets with severe diarrhoea shed E. coli O149 in the faeces. Zootechnical parameters did not show significant differences. The experimental conditions described in this study allowed to effectively induce diarrhoea in weaned piglets. In conclusion a multifactorial approach (infectious, nutritional and management) is necessary to reproduce in vivo diarrhea in piglets.

Community batteries(CBs)are emerging to support and even enable energy communities and generally help consumers,especially space-constrained ones,to access potential techno-economic benefits from storage and support local grid decarbonization.However,the economic viability of CB projects is often uncertain.In this regard,typical feasibility studies assess CB value for behind-the-meter(BTM)operation or whole-sale market participation,i.e.,front-of-meter(FOM).This work proposes a novel techno-economic operational framework that allows systematic assessment of the different options and introduces a two-meter architecture that co-optimizes both BTM and FOM benefits.A real CB project application in Australia is used to demonstrate the significant two-meter co-optimization opportunities that could enhance the business case of CB and energy communities by multi-service provision and value stacking.

The literary review presented in the following paper aims to analyze the tracking tools used in different countries during the period of the COVID-19 pandemic. Tracking apps that have been adopted in many countries to collect data in a homogeneous and immediate way have made up for the difficulty of collecting data and standardizing evaluation criteria. However, the regulation on the protection of personal data in the health sector and the adoption of the new General Data Protection Regulation in European countries has placed a strong limitation on their use. This has not been the case in non-European countries, where monitoring methodologies have become widespread. The textual analysis presented is based on co-occurrence and multiple correspondence analysis to show the contact tracing methods adopted in different countries in the pandemic period by relating them to the issue of privacy. It also analyzed the possibility of applying Blockchain technology in applications for tracking contagions from COVID-19 and managing health data to provide a high level of security and transparency, including through anonymization, thus increasing user trust in using the apps.

Oxidative stress is one of the main mechanisms responsible for male infertility.Various conditions such as varicocele,obesity,advanced age,and lifestyle can lead to an increase in reactive oxygen species,causing an oxidative imbalance in the reproductiveenvironment.Spermatozoa are sensitive to reactive oxygen species and require energy to carry out their main function of fertilizingthe egg.Excessive reactive oxygen species can affect sperm metabolism,leading to immobility,impaired acrosome reaction,and celldeath,thereby impairing reproductive success.This double-blind randomized study evaluated the effect of supplementation withL-carnitine,acetyl-L-carnitine,vitamins,and other nutrients on semen quality in 104 infertile patients with or without varicocele,while also investigating the impact of factors such as obesity and advanced age on treatment.Sperm concentration significantlyincreased in the supplemented group compared to the placebo group(P=0.0186).Total sperm count also significantly increasedin the supplemented group(P=0.0117),as did sperm motility(P=0.0120).The treatment had a positive effect on patients upto 35 years of age in terms of sperm concentration(P=0.0352),while a body mass index(BMI)above 25 kg m−2 had a negativeeffect on sperm concentration(P=0.0110).Results were not showing a net benefit in stratifying patients in accordance withtheir BMI since sperm quality increase was not affected by this parameter.In conclusion,antioxidant supplementation may bebeneficial for infertile patients and has a more positive effect on younger patients with a normal weight.

The plant vacuole plays a fundamental role in cell homeostasis.The successful application of patch-clamp tech-nique on isolated vacuoles allows the determination of the functional characteristics of tonoplast ion channels and transporters.The parallel use of a sensor-based fluorescence approach capable of detecting changes in calcium and proton concentrations opens up new possibilities for investigation.In excised patch,the presence of fura-2 in the vacuolar solution reveals the direct permeation of calcium in plant TPC channels.In whole-vacuole,the activity of non-electrogenic NHX potassium proton antiporters can be measured by using the proton sensitive dye BCECF loaded in the vacuolar lumen by the patch pipette.Both vacuolar NHXs and CLCa(chloride/nitrate antiporter)are inhibited by the phosphoinositide PI(3,5)P2,suggesting a coordinated role of these proteins in salt accumulation.Increased knowledge in the molecular mechanisms of vacuolar ion channels and transporters has the potential to improve our understanding on how plants cope with a rapidly changing environment.

Currently,colorectal cancer(CRC)represents the third most common malignancy and the second most deadly cancer worldwide,with a higher incidence in developed countries.Like other solid tumors,CRC is a heterogeneous genomic disease in which various alterations,such as point mutations,genomic rearrangements,gene fusions or chromosomal copy number alterations,can contribute to the disease development.However,because of its orderly natural history,easily accessible onset location and high lifetime incidence,CRC is ideally suited for preventive intervention,but the many screening efforts of the last decades have been compromised by performance limitations and low penetrance of the standard screening tools.The advent of next-generation sequencing(NGS)has both facilitated the identification of previously unrecognized CRC features such as its relationship with gut microbial pathogens and revolutionized the speed and throughput of cataloguing CRC-related genomic alterations.Hence,in this review,we summarized the several diagnostic tools used for CRC screening in the past and the present,focusing on recent NGS approaches and their revolutionary role in the identification of novel genomic CRC characteristics,the advancement of understanding the CRC carcinogenesis and the screening of clinically actionable targets for personalized medicine.
