Ferroptosis drives immune-mediated neurodegeneration in multiple sclerosis

One of the leading issues in multiple sclerosis(MS)research is the lack of understanding regarding the triggers of neurodegeneration.While demyelination is a major feature in MS,the degeneration of gray matter neurons contributes to major changes in patient function,but the mechanisms driving this neuronal loss are unknown.In exciting research published by Luoqian et al.[1]in the December issue of Cellular and Molecular Immunology,new insights have shown an important contribution of ferroptosis to T-cell-mediated neurodegeneration in the common animal model of MS,‘experimental autoimmune encephalomyelitis’(EAE).Although a relationship between ferroptosis and T-cell activation has been reported in other disorders,this is the first discovery of a link between abnormal iron homeostasis and T-cell infiltration in MS.In addition,the authors found that unlike other studies on ferroptosis-T-cell interactions[2,3],ferroptotic activation of T cells in EAE,and potentially MS,involved the infiltration and activation of CD4+T cells,rather than CD8+cells.Moreover,in this study,ferroptosis was a key driver of T-cell activation rather than the opposite interaction where CD8+cells drive ferroptosis[3].The research also mapped a key role for Acsl4 in the early stages of ferroptosis-T-cell-mediated neurodegeneration.Acsl4 converts free long-chain fatty acids into fatty acyl-CoA esters and has been identified as a central contributor to ferroptotic processes[4].This study has mapped a major new pathway potentially leading to neurodegeneration in MS,and this opens the possibility of new therapeutic approaches to treat MS based on iron targeting as the authors describe in their final experiments.

Utilizing supported single atoms as catalysts presents an opportunity to reduce the usage of critical raw materials such as platinum,which are essential for electrochemical reactions such as hydrogen oxidation reaction(HOr).Herein,we describe the synthesis of a Pt single electrocatalyst inside single-walled carbon nanotubes(SWCNTs)via a redox reaction.Characterizations via electron microscopy,X-ray photoelectron microscopy,and X-ray absorption spectroscopy show the single-atom nature of the Pt.The electrochemical behavior of the sample to hydrogen and oxygen was investigated using the advanced floating electrode technique,which minimizes mass transport limitations and gives a thorough insight into the activity of the electrocatalyst.The single-atom samples showed higher HOr activity than state-of-the-art 30%Pt/C while almost no oxygen reduction reaction activity in the proton exchange membrane fuel cell operating range.The selective activity toward HOr arose as the main fingerprint of the catalyst confinement in the SWCNTs.

Big data analytics has been widely adopted by large companies to achieve measurable benefits including increased profitability,customer demand forecasting,cheaper development of products,and improved stock control.Small and medium sized enterprises(SMEs)are the backbone of the global economy,comprising of 90%of businesses worldwide.However,only 10%SMEs have adopted big data analytics despite the competitive advantage they could achieve.Previous research has analysed the barriers to adoption and a strategic framework has been developed to help SMEs adopt big data analytics.The framework was converted into a scoring tool which has been applied to multiple case studies of SMEs in the UK.This paper documents the process of evaluating the framework based on the structured feedback from a focus group composed of experienced practitioners.The results of the evaluation are presented with a discussion on the results,and the paper concludes with recommendations to improve the scoring tool based on the proposed framework.The research demonstrates that this positioning tool is beneficial for SMEs to achieve competitive advantages by increasing the application of business intelligence and big data analytics.

As big data becomes an apparent challenge to handle when building a business intelligence(BI)system,there is a motivation to handle this challenging issue in higher education institutions(HEIs).Monitoring quality in HEIs encompasses handling huge amounts of data coming from different sources.This paper reviews big data and analyses the cases from the literature regarding quality assurance(QA)in HEIs.It also outlines a framework that can address the big data challenge in HEIs to handle QA monitoring using BI dashboards and a prototype dashboard is presented in this paper.The dashboard was developed using a utilisation tool to monitor QA in HEIs to provide visual representations of big data.The prototype dashboard enables stakeholders to monitor compliance with QA standards while addressing the big data challenge associated with the substantial volume of data managed by HEIs’QA systems.This paper also outlines how the developed system integrates big data from social media into the monitoring dashboard.

Parkinson’s disease(PD,OMIM#168600)is a common neurodegenerative disorder with a global prevalence of approximately 8.5 million.PD is characterized by four cardinal motor symptoms:bradykinesia,rigidity,resting tremor,and subsequently by postural instability.It usually involves non-motor symptoms such as rapid eye movement sleep disorder,dementia,anosmia,and autonomic dysfunction.The gene glucocerebrosidase 1(GBA1),which encodes the lysosomal enzyme glucocerebrosidase(GCase)(IUBMB:EC 3.***.1.45),shows strong linkage with PD;variants of GBA1 are the commonest genetic association with PD(Sidransky et al.,2009).Several mechanisms may underlie the relationship between GBA1 mutations/variants and the molecular pathology of PD(Figure 1A and B).

The spatiotemporal distributions of microbes in soil by different methods could affect the efficacy of the microbes to reduce the soil hydraulic conductivity.In this study,the specimens of bio-mediated sands were prepared using three different methods,i.***.injecting,mixing,and pouring a given microbial so-lution onto compacted sand specimens.The hydraulic conductivity was measured by constant-head tests,while any soil microstructural changes due to addition of the microbes were observed by scan-ning electron microscope(SEM)and mercury intrusion porosimetry(MIP)tests.The amount of dextran concentration produced by microbes in each type of specimen was quantified by a refractometer.results show that dextran production increased exponentially after 5-7 d of microbial settling with the supply of culture medium.The injection and mixing methods resulted in a similar amount and uniform dis-tribution of dextran in the specimens.The pouring method,however,produced a nonuniform distri-bution,with a higher concentration near the specimen surface.As the supply of culture medium discontinued,the dextran content near the surface produced by the pouring method decreased dramatically due to high competition for nutrients with foreign colonies.Average dextran concentration was negatively and correlated with hydraulic conductivity of bio-mediated soils exponentially,due to the clogging of large soil pores by dextran.The hydraulic conductivity of the injection and mixing cases did not change significantly when the supply of culture medium was absent.

Traumatic brain injury(TBI) impacts 69 million individuals globally each year and is a leading cause of death and disability(Dewan et al.,2019).The majority of moderate-to-severe TBI survivors endure long-lasting disturbances in motor,cognitive,and affect that negatively impacts their life.Although a plethora of research on pharmacological interventions for TBI has been conducted,none has translated to the clinic,thus advocating for the evaluation of nonpharmacological therapeutic approaches that may increase translational success.

Pulverizing is an essential unit operation in co-firing biomass with coal.Pulverizers are only compatible with pellet forms of fibrous biomass materials and crush them down to their original forming particle sizes.That is why the data on the size distribution of the particles forming a biomass pellet is crucial to achieving optimum combustion conditions.The current study determines the internal particle size distribution of pellets after wet disintegration,following Iso 17830 standard,and aims to suggest improvements to the mentioned standard based on new measured evidence.Experiments were carried out on white wood pellets(no bark)and brown wood pellets containing bark at four water temperatures:20,40,60,and 95℃,with or without stirring.The particle size distribution of the pre-pelletizer wood particles was also measured and compared with particles in the formed pellets.Ambient water temperature of 20℃ was found to be adequate for the complete disintegration of pellets,and no mechanical stirring was required.About 30% of particles in the disintegrated pellets were 0.5-1.0 mm.Pelletization changes the particle size distribution to smaller particles.The disintegrated bark pellets contained more fines than white pellets.

Brain functional impairment after stroke is common;however,the molecular mechanisms of post-stroke recovery remain unclear.It is well-recognized that age is the most important independent predictor of poor outcomes after stroke as older patients show poorer functional outcomes following stroke.Mounting evidence suggests that axonal regeneration and angiogenesis,the major forms of brain plasticity responsible for post-stroke recovery,diminished with advanced age.Previous studies suggest that ras-related C3 botulinum toxin substrate(rac)1 enhances stroke recovery as activation of rac1 improved behavior recovery in a young mice stroke model.Here,we investigated the role of rac1 signaling in long-term functional recovery and brain plasticity in an aged(male,18 to 22 months old C57BL/6J)brain after ischemic stroke.We found that as mice aged,rac1 expression declined in the brain.Delayed overexpression of rac1,using lentivirus encoding rac1 injected day 1 after ischemic stroke,promoted cognitive(assessed using novel object recognition test)and sensorimotor(assessed using adhesive removal tests)recovery on days 14–28.This was accompanied by the increase of neurite and proliferative endothelial cells in the periinfarct zone assessed by immunostaining.In a reverse approach,pharmacological inhibition of rac1 by intraperitoneal injection of rac1 inhibitor NSC23766 for 14 successive days after ischemic stroke worsened the outcome with the reduction of neurite and proliferative endothelial cells.Furthermore,rac1 inhibition reduced the activation of p21-activated kinase 1,the protein level of brain-derived neurotrophic factor,and increased the protein level of glial fibrillary acidic protein in the ischemic brain on day 28 after stroke.Our work provided insight into the mechanisms behind the diminished plasticity after cerebral ischemia in aged brains and identified rac1 as a potential therapeutic target for improving functional recovery in the older adults after stroke.
