Impact of Helicobacter pylori on the healing process of the gastric barrier

AIM To determine the impact of selected well defined Helicobacter pylori(H. pylori) antigens on gastric barrier cell *** In this study,using two cellular models of gastric epithelial cells and fibroblasts,we have focused on exploring the effects of well defined H. pylori soluble components such as glycine acid extract antigenic complex(GE),subunit A of urease(Ure A),cytotoxin associated gene A protein(Cag A) and lipopolysaccharide(LpS) on cell turnover by comparing the wound healing capacity of the cells in terms of their proliferative and metabolic activity as well as cell cycle distribution. Toxic effects of H. pylori components have been assessed in an association with damage to cell nuclei and inhibition of signal transducer and activator of transcription 3(STAT3) phosphorylation. RESULTS We showed that H. pylori GE,Cag A and Ure A promoted regeneration of epithelial cells and fibroblasts,which is necessary for effective tissue healing. However,in vivo increased proliferative activity of these cells may constitute an increased risk of gastric neoplasia. In contrast,H. pylori LpS showed a dose-dependent influence on the process of wound healing. At a low concentration(1 ng/m L) H. pylori LpS accelerated of healing epithelial cells,which was linked to significantly enhanced cell proliferation and MTT reduction as well as lack of alterations in cell cycle and downregulation of epidermal growth factor(EGF) production as well as cell nuclei destruction. By comparison,H. pylori LpS at a high concentration(25 ng/m L) inhibited the process of wound repair,which was related to diminished proliferative activity of the cells,cell cycle arrest,destruction of cell nuclei and downregulation of the EGF/STAT3 signalling *** In vivo H. pylori LpS driven effects might lead to the maintenance of chronic inflammatory response and pathological disorders on the level of the gastric mucosal barrier.

Evaluation of co-spoilage potential of pseudomonas fragi and pseudomonas lundensis isolated from chilled pork: in vitro and in situ

pseudomonas (p.) fragi and p. lundensis have been reported as key spoilage bacteria in aerobically-stored chilled meat. However， the co-spoilage effect of these bacteria has not been effectively evaluated. This study evaluated the spoilage potential of p. fragi， p. lundensis and p. fragi + p. lundensis in vitro and in situ at 4 ℃. The results showed that p. fragi + p. lundensis performed the highest growth rate and displayed larger decomposition zone diameters on Raw-pork juice agar (RJA) plates. p. fragi + p. lundensis inoculants exhibited the strongest proteolytic activity， which resulted in the highest values of trichloroacetic acid (TCA)-soluble peptides， total volatile basic nitrogen (TVB-N) and myofibril fragmentation index (MFI) in chilled pork. Moreover， the inoculated samples showed different pH and sensory changes. Notably， increased amounts of volatile organic compounds (VOCs)， such as octanal， nonanal， 2-nonanone， 1-propanol， 1-octanol， isopropyl acetate， and 2，6-dimethylpyazine， were observed in inoculated p. fragi + p. lundensis samples， hinting their potential use as spoilage markers for spoilage monitoring of co-cultures. This study would provide a deeper understanding of meat spoilage and serve as a reference for future studies to inhibit meat spoilage.

Academic productivity correlates with industry earnings in foot and ankle fellowship programs in the United States:A retrospective analysis

Effect of Dietary Components on the Shear Bond Strength of Orthodontics Brackets after Thermal Aging

Introduction: The stability of orthodontic brackets throughout orthodontic treatment plays a critical role in the treatment’s effectiveness. The present in vitro study was designed to assess the impact of various dietary components on the performance of orthodontic brackets. Methods: Metal orthodontic brackets were bonded to 66 extracted anterior teeth divided into groups based on the solution type: Milk, Gatorade, Cold Coffee, and a control group using water. Each group consisted of 20 teeth except for the control group, which included six teeth. The bracketed teeth were submerged in their respective solutions for 15 minutes three times daily at different intervals to mimic an in vivo environment and were stored in artificial saliva at room temperature (23?C). The specimens underwent artificial aging through 10,000 cycles of thermocycling (representing one clinical year) between 5?C and 55?C. Shade measurements were taken using a VITA Easy Shade device, capturing the classic shade and L*, a*, and b* values. Delta E values were calculated immediately post-bonding and after 7 days, 1 month, 1, and 2 clinical years. The shear bond strength of each bracket was measured using an ultra-tester machine. Results: After two clinical years, significant differences in ΔE color values were observed across all groups, with the most substantial change noted in teeth immersed in cold coffee. Brackets submerged in milk demonstrated lower shear bond strength than other solutions, whereas the control group exhibited the highest shear bond strength (p = 0.01). Conclusion: The study indicates that dietary components significantly influence tooth color stability and the shear bond strength of orthodontic brackets, underscoring the importance of considering these factors in orthodontic treatment planning.
