Activation of complement generates C5a which leads to signaling through C5aR 1. This is tightly controlled, including by the plasma proteins factor H （FH） and carboxypeptidase N. Here we studied a chronic serum sickness （CSS） model of glomerulonephritis （GN） in which there is an active humoral immune response, formation of glomerular immune complexes （ICs）, and resulting glomerular inflammation. The antibody response, glomerular IC deposition, the degree of GN, and consequent renal functional insufficiency in CSS were all worse in FH-/- mice compared to wild-type FH＋/＋ animals. This was ameliorated in the former by giving a C5aR1 antagonist for the final 3 weeks of the 5-week protocol. In contrast, blocking CP-mediated inactivation of C5a increased these disease measures. Thus, complement regulation by both plasma FH and CP to limit the quantity of active C5a is important in conditions where the humoral immune response is directed to a continuously present foreign antigen. Signaling through C5aR1 enhances the humoral immune response as well as the inflammatory response to ICs that have formed in glomeruli. Both effects are relevant even after disease has begun. Thus, pharmacological targeting of C5a in IC-mediated GN has potential clinical relevance.

To the Editor: Hypoxic hepatitis(HH), also known as ischemic hepatitis or shock liver, is a liver injury characterized by necrosis of centrilobular hepatocytes with a rapid increase in serum aminotransferase levels. The incidence rate of HH among patients in the intensive care unit(ICU) was found to be 0.9%-11.9% [1]. Occurrence of HH appears to have a significant impact on the clinical outcome.

Systemic lupus erythematosus(SLE)is an autoimmune disease that is a challenge to diagnose and *** is an urgent need for biomarkers to help define organ involvement,and more effective therapies.A unique population of T cells,the CD3^(+)CD4^(−)CD8^(−)(DNeg)cells,is significantly increased in lupus *** cases(53%)of pediatric SLE patients had elevated DNeg cells in their peripheral blood,which correlated with kidney function(R^(2)=0.54).Significant infiltration of DNeg cells was observed in both adult and pediatric lupus kidneys by *** the first time,this study provides direct evidence that DNeg cells facilitate kidney injury in preclinical 8-week-old MRL/lpr lupus *** lupus mice,the increase in DNeg cells tracked with worsening disease and correlated with kidney function(R^(2)=0.85).Our results show that DNeg cells per se can cause kidney dysfunction,increase in number with increase in disease pathology,and could serve as a potential biomarker.

AIM: To describe the clinicopathologic features of concurrent polyomavirus nephropathy(PVN) and endarteritis due to rejection in renal ***: We searched our electronic records database for cases with transplant kidney biopsies demonstrating features of both PVN and acute rejection(AR). PVN was defined by the presence of typical viral cytopathic effect on routine sections and positive polyomavirus SV40 large-T antigen immunohistochemistry. AR was identified by endarteritis(v1 by Banff criteria). All cases were subjected to chart review in order to determine clinical presentation, treatment course and outcomes. Outcomes were recorded with a length of follow-up of at least one year or time to nephrectomy. RESULTS: Of 94 renal allograft recipients who developed PVN over an 11-year period at our institution, we identified 7(7.4%) with viral cytopathic changes, SV40 large T antigen staining, and endarteritis in the same biopsy specimen, indicative of concurrent PVN and AR. Four arose after reduction of immunosuppression(IS)(for treatment of PVN in 3 and tuberculosis in 1), and 3 patients had no decrease of IS before developing simultaneous concurrent disease. Treatment consisted of reduced oral IS and leflunomide for PVN, and antirejection therapy. Three of 4 patients who developed endarteritis in the setting of reduced IS lost their grafts to rejection. All 3 patients with simultaneous PVN and endarteritis cleared viremia and were stable at 1 year of follow up. Patients with endarteritis and PVN arising in a background of reduced IS had more severe rejection and poorer ***: Concurrent PVN and endarteritis may be more frequent than is currently appreciated and may occur with or without prior reduction of IS.

The purposes of this study are to assess the COVID-19 pandemic’s impacts on the dairy industries in China and the United States and to derive policy recommendations for enhancing the diary industries’resilience to pandemics and other market ***,data from the two nations are used to analyze and compare the mechanisms through which the pandemic has affected their dairy industries and to discuss potential lessons from their *** findings suggest that this pandemic has heavily affected the dairy industries in both China and the United States through similar mechanisms,such as decreased farmgate milk prices,disruption and difficulties of moving milk within the supply chains,worker shortages,increased production costs,and lack of operating *** were also significant differences in the affecting mechanisms between the two nations,including transportation difficulties from widespread road closures and significant reduction in holiday sales of dairy products in China,and the shutdown of many dairy processors in the United States due to the closing of schools,restaurants,and *** government financial reliefs are highly needed to help many dairy farms and processors survive this pandemic in the short term,the dairy industries and governments need to work together to develop long-term strategies and policies to balance the industries’efficiency and flexibility,product specialization and diversification,supply chain integration and local food systems,and market mechanisms and policy regulations and interventions.

Background:There are two forms of system implementation of optical coherence tomography angiography(OCTA)in ophthalmic imaging,i.e.,spectral domain(SD-)and swept source OCTA(SS-OCTA).The purpose of this paper is to compare the SD-OCTA and SS-OCTA for elucidating structural and vascular features associated with central serous retinopathy(CSR),and to evaluate the effects of CSR on SD-and SS-OCTA’s imaging ***:Normal subjects and CSR patients were imaged by SD-and SS-OCTA using 3×3 mm and 6×6 mm scan *** signal strengths at the superficial retina,deep retina,Sattler’s layer and Haller’s layer were used to compare the ability of SD-and SS-OCTA to image structural *** addition,the ability to acquire angiograms were discussed by evaluating retinal vessel *** serous volume(CSV)was measured and it was correlated with difference in signal strengths(ΔS)between two OCTA ***:Seven normal eyes and seven diseased eyes were *** showed no significant differences between SD-and SS-OCT in detecting structural features of the retinal layer according to the paired ***,when imaging the Sattler’s layer for normal eyes,a significant difference is found between SD-and SS-OCT(p<0.0001 for 3×3 mm scan,and p=0.0002 for 6×6 mm);while for CSR eyes,the corresponding values were p<0.0001 and p=0.0003,*** Haller’s layer for normal eyes,the corresponding values were p=0.0004 and p=0.0014;and for CSR eyes,p=0.0004 and p<0.0001,respectively.A strong correlation betweenΔS and CSV was observed in the Sattler’s layer(3×3 mm–p=0.0031 and R^(2)=0.951;6×6 mm–p=0.0075 and R^(2)=0.911)and Haller’s layer(3×3 mm–p=0.0026 and R^(2)=0.955;6×6 mm–p=0.0013 and R^(2)=0.972).Conclusions:The results suggest no differences between SD-and SS-OCTA for imaging the retinal layers however,when imaging beyond retinal layers,SS-OCTA appears advantageous in detecting returning *** CSR cases,the CSV may have an impact on sub-C
