STING regulates BcR signaling in normal and malignant B cells

STING is an endoplasmic reticulum(ER)-resident protein critical for sensing cytoplasmic DNA and promoting the production of type Ⅰ interferons;however,the role of STING in B cell receptor(BcR)signaling remains *** generated STING V154M knock-in mice and showed that B cells carrying constitutively activated STING specifically degraded membrane-bound IgM,Ig a and Igβ via SEL1L/HRD1-mediated ER-associated degradation(ERAD).B cells with activated STING were thus less capable of responding to BcR activation by phosphorylating Igα and Syk than those without activated *** immunized with T-independent antigens,STING V154M mice produced significantly fewer antigen-specific plasma cells and antibodies than immunized wild-type(WT)*** further generated B cell-specific STING^(KO) mice and showed that STING^(KO) B cells indeed responded to activation by transducing stronger BcR signals than their STING-proficient *** B cell-specific STING^(KO) mice were T-independently immunized,they produced significantly more antigen-specific plasma cells and antibodies than immunized STIN^(WT)*** both human and m ouse IGHV-unmutated malignant chronic lymphocytic leukemia(cLL)cells downregulated the expression of STING,we explored whether STING downregulation could contribute to the well-established robust BcR signaling phenotype in malignant cLL *** generated a STING-deficient cLL mouse model and showed that STING-deficient cLL cells were indeed more responsive to BcR activation than their STING-proficient *** results revealed a novel B cell-intrinsic role of STING in negatively regulating BcR signaling in both normal and malignant B cells.

Effects of Processing on Antioxidant capacity and Metabolizing Enzyme Inhibition of Tiger Nut Tubers

Tiger nut (cyperus esculentus), commonly referred to as chufa, is a “nut-like” tuber, native to tropical and Mediterranean regions. It is often consumed in the form of a beverage (horchata de chufa). Health benefits of tiger nut have been attributed to presence of phytochemicals and their antioxidant potential. Tiger nut is often processed in order to enhance sensory characteristics. The objective of study was to determine effects of processing on antioxidant content, potential and inhibition of metabolizing enzyme activities of tiger nuts. Total phenolic content (TPc), total flavonoid content (TFc), free radical scavenging activity by 1,1-diphenyl-2-picryhydrazyl (DPPH), trolox equivalent antioxidant capacity (TEAc), ferric reducing antioxidant power (FRAP) and inhibition of lipase and α-amylase were evaluated in raw tiger nuts extracted with water (TRWA) and methanol (TRWM) and roasted (110°c – 115°c for 10 mins) tiger nuts extracted with water (TRA) and methanol (TRM). Highest phenolic content was observed in TRWA (1186.68 GAE/100g) and TRWM had lowest (43.77 GAE/100g). TRWA also exhibited highest ferric reducing ability with a FRAP value of 169.83 mm Fe2+/100g). TRWM had a higher flavonoid content (220.68 cAE/100g) compared to all other extracts. At a concentration of 0.02 mg/ml, all extracts reached at least 50% inhibition of DPPH, while highest scavenging activity was observed in TRM (58%). Highest TEAc was observed in TRM with 55.66% of ABTS radical scavenged. TRA had a significantly higher ability to inhibit α-amylase and lipase compared to other extracts. Results suggest that processing had an effect on the antioxidant potential of tiger nut, and this tuber may benefit consumers by improving their antioxidant status.

comparison of the chloride channel activator lubiprostone and the oral laxative Polyethylene Glycol 3350 on mucosal barrier repair in ischemic-injured porcine intestine

AIM： To investigate the effects of lubiprostone and Polyethylene Glycol 3350 （PEG） on mucosal barrier repair in ischernic-injured porcine intestine. METHODS： Ileum from 6 piglets （approximately 15 kg body weight） was subjected to ischemic conditions by occluding the local rnesenteric circulation for 45 min in vivo. Ileal tissues from each pig were then harvested and mounted in Ussing chambers and bathed in oxygenated Ringer＇s solution in vitro. Intestinal barrier function was assessed by measuring transepithelial electrical resistance （TER） and mucosal-to-serosal fluxes of ^3H-rnannitol and ^14c-inulin. Statistical analyses of data collected over a 120-min time course included 2-way ANOVA for the effects of time and treatment on indices of barrier function. RESULTS： Application of 1μmol/L lubiprostone to the rnucosal surface of ischemic-injured ileum in vitro induced significant elevations in TER compared to nontreated tissue. Lubiprostone also reduced mucosal-toserosal fluxes of ^3H-rnannitol and ^14c-inulin. Alternatively, application of a polyethylene laxative （PEG, 20 rnmol/L） to the mucosal surface of ischernic tissues significantly increased flux of ^3H-rnannitol and ^14c-inulin. cONcLUSION： This experiment demonstrates that lubiprostone stimulates recovery of barrier function in ischemic intestinal tissues whereas the PEG laxative had deleterious effects on mucosal repair. These results suggest that, unlike osmotic laxatives, lubiprostone stimulates repair of the injured intestinal barrier.

Rattlesnakes use chemical stimuli in ambush site selection and for relocation of envenomated prey through atrike-induced chemosensory searching. Shifts in responsiveness to prey chemicals have been documented in many snakes, and often correlate with prey commonly taken as snakes increase in age and size as well as geographical locations of the species. For instance, neonate rattlesnakes that prey primarily on ectotherms responded most strongly to chemical cues of commonly taken lizard prey, whereas adult rattlesnakes that prey primarily on small mammals responded significantly to chemical cues of com- monly taken rodents. In the current study, 11 Prairie Rattlesnakes crotalus viridis viridis which were classified as large neonates based on measures of snout-vent length （SVL） and body mass, yet chronologically were at or near adulthood, were tested for their responsiveness to chemical extracts of natural and non-natural prey items. Although the snakes had eaten only neonate lab mice （Mus musculus）, they responded significantly more to chemical cues of natural prey items and particularly to chemical cues of prey normally taken by subadults （Peromyscus mice and Sceloporus lizard）. These results suggest that ontogenetic shifts in re- sponsiveness to natural prey chemical cues are innately programmed and are not based on body size or feeding experience in c. v. viridis. This does not imply, however, that growth and experience are without effects, especially with novel prey or rare prey that have experienced recent population expansion [current Zoology 59 （2）： 175-179, 2013].

Assessing disease activity using the pediatric crohn’s disease activity index:can we use subjective or objective parameters alone?

BAcKGROUND The pediatric crohn’s disease activity index(PcDAI)is used as a standard tool to assess disease activity in clinical trials for pediatric crohn’s *** To examine which items on the PcDAI drive assessment of disease activity,and how subgroups of subjective and objective items reflect change in disease state over *** Selective raw data from three prospectively collected datasets were combined,including 703 children with full PcDAI data at baseline,at 3-mo(Q1,n=670),and 1-year(Q4,n=474).change in individual PcDAI scores from baseline to Q1 and to Q4 were examined using the non-weighted *** Abdominal pain,well-being,weight,and stooling had the highest change scores over *** indicators including albumin,abdominal exam,and height velocity *** scores for well-being and abdominal exam did not explain significant variance at Q1 but were significant predictors at Q4(Pctive and objective subgroups of items predicted less variance(18%and 22%)on total PcDAI scores at Q1 and Q4 compared to the full PcDAI,or a composite scale(both 32%)containing significant *** Although subjective items on the PcDAI change the most over time,the full PcDAI or a smaller composite of items including a combination of subjective and objective components classifies disease activity better than a subgroup of either subjective or objective items *** on subjective or objective items as stand-alone proxies for disease activity measurement could result in misclassification of disease state.

Large electromechanical strain at high temperatures of novel <001> textured BiFeGaO3-BaTiO3 based ceramics

BiFeGaO3-BaTiO3(BFG-BT)based ceramics with a large piezoelectric coefficient are potential high performance lead-free piezoelectric *** this work,textured and random BFG-BT ceramics were realized by the solid state reaction method with and without BaTiO3(BT)*** ceramics were obtained by a reactive templated grain growth(RTGG)method leading to a high-temperature electromechanical strain of S=0.27%at 40 kV/cm and to an effective piezoelectric coefficient(d33*)up to 685 pm/V at 180℃.The easy movement of oriented domains enhanced the electromechanical strain under an applied electric field in textured sample(Lotgering factor f=66.3%).Structural investigations reveal that the proportion and degree of distortion of BFG-BT rhombohedral phase(R3c)reached its maximum in textured ceramics,resulting in large ferrodistortive displacements under electric *** addition,the dense nanodomains with low domain wall energies,inferred from the high-resolution transmission electron microscope(HR-TEM)observations,contribute to the extra displacement of the textured sample under an applied electric *** textured ceramics,the remnant polarization was stable(about 17μc/cm2)from room temperature to 180℃,contributing to the stable ferroelectric property at high *** the introduction of BT templates,high-density nanodomains were formed and the Burns temperature was enhanced in textured *** electromechanical strain,polarization and dielectric behavior were correlated to the textured or random forms of the BFG-BT based ceramics.

Modeling and multiobjective optimization of traction performance for autonomous wheeled mobile robot in rough terrain

Application of terrain-vehicle mechanics for determination and prediction of mobility performance of autonomous wheeled mobile robot （AWMR） in rough terrain is a new research area currently receiving much attention for both terrestrial and planetary missions due to its significant role in design, evaluation, optimization, and motion control of AWMRs. In this paper, decoupled closed form terramechanics considering important wheel-terrain parameters is applied to model and predict traction. Numerical analysis of traction performance in terms of drawbar pull, tractive efficiency, and driving torque is carried out for wheels of different radii, widths, and lug heights, under different wheel slips. Effects of normal forces on wheels are analyzed. Results presented in figures are discussed and used to draw some conclusions. Furthermore, a multiobjective optimization （MOO） method for achieving optimal mobility is presented. The MOO problem is formulated based on five independent variables in- eluding wheel radius r, width b, lug height h, wheel slip s, and wheel rotation angle 0 with three objectives to maximize drawbar pull and tractive efficiency while minimizing the dynamic traction ratio. Genetic algorithm in MATLAB is used to obtain opti- mized wheel design and traction control parameters such as drawbar pull, tractive efficiency, and dynamic traction ratio required for good mobility performance. comparison of MOO results with experimental results shows a good agreement. A method to apply the MOO results for online traction and mobility prediction and control is discussed.

BAcKGROUND The study investigates the connection between academic productivity and industry earnings in foot and ankle orthopedic surgery *** metrics like the H-index and Open Payments Database(OPD)data,it addresses a gap in understanding the relationship between scholarly achievements and financial outcomes,providing a basis for further exploration in this specialized medical *** To elucidate the trends between academic productivity and industry earnings across foot and ankle orthopedic surgery fellowship programs in the United *** This study is a retrospective analysis of the relationship between academic productivity and industry earnings of foot and ankle orthopedic surgery fellowships at an individual faculty and fellowship *** productivity was defined via H-index and recorded from the Scopus *** earnings were recorded from the *** Forty-eight foot and ankle orthopedic surgery fellowships(100%of fellowships)in the United States with a combined total of 165 physicians(95.9%of physicians)were *** individual physician(n=165)total life-time earnings reported on the OPD website was United States Dollar(USD)451430.30±1851084.89(range:USD 25.16-21269249.85;median:USD 27839.80).Mean physician(n=165)H-index as reported on Scopus is 14.24±12.39(range:0-63;median:11).There was a significant but weak correlation between individual physician H-index and individual physician total life-time earnings(Pcant and moderate positive correlation between combined fellowship H-index and total life-time earnings per fellowship(P=0.004,Spearman’s rho=0.409).cONcLUSION There is a significant and positive correlation between academic productivity and industry earnings at foot and ankle orthopedic surgery fellowships in the United *** observation is true on an individual physician level as well as on a fellowship level.

Dual Phase Renewable Fuel combustion in an Atmospheric Gas Turbine Burner

Expanding the fuel flexibility of continuous combustion systems to include multiphase fuel combustion offers additional support to combat the problem of energy security and,potentially,environmental *** this study,apart from establishing stability limits and measuring post-combustion emissions,flames generated from simultaneous combustion of biodiesel and syngas were examined using c_(2)*and cH*chemiluminescence imaging to capture changes in the reaction *** proportion of syngas in the fuel mix was varied from 0 to 30%content(by energy contribution)while maintaining a total power output of 15 *** overall equivalence ratio was held at 0.7 in cases other than for determining the flammability *** results indicate a reduction of stability limits as gas proportion in fuel blend ***,chemiluminescence imaging of the two targeted species suggest a general reduction in reaction rate as well as reaction zone area and length with increase in gas ratio in the dual phase ***,emissions performance in the context of NOxand cO was investigated as liquid-to-gas ratios were ***,the study demonstrates the feasibility,limitations and potential benefits of multiphase renewable fuel combustion in a swirl-stabilised burner.
