BACKGROUND Peripheral blood stem cells(PBSC)are commonly cryopreserved awaiting clinical use for hematopoietic stem cell *** term cryopreservation is commonly defined as five years or longer,and limited data exists regarding how long PBSC can be cryopreserved and retain the ability to successfully *** programs,stem cell banks,and regulatory and accrediting agencies interested in product stability would benefit from such ***,we assessed recovery and colony forming ability of PBSC following long-term cryopreservation as well as their ability to engraft in NOD/SCID/IL-2 Rγnull(NSG)*** To investigate the in vivo engraftment potential of long-term cryopreserved PBSC *** PBSC units which were collected and frozen using validated clinical protocols were obtained for research use from the Cellular Therapy Laboratory at Indiana University *** units were thawed in the Cellular Therapy Laboratory using clinical standards of practice,and the pre-freeze and post-thaw characteristics of the units were *** function was assessed using standard colony-forming ***34-selected cells were transplanted into immunodeficient mice to assess stem cell *** Ten PBSC units with mean of 17 years in cryopreservation(range 13.6-18.3 years)demonstrated a mean total cell recovery of 88%±12%(range 68%-110%)and post-thaw viability of 69%±17%(range 34%-86%).BFU-e growth was shown in 9 of 10 units and CFU-GM growth in 7 of 10 units *** mice were transplanted with CD34-selected cells from four randomly chosen PBSC *** mice demonstrated long-term engraftment at 12 wk with mean34%±24%human CD45+cells,and differentiation with presence of human CD19+,CD3+and CD33+*** bone marrow from all mice demonstrated growth of erythroid and myeloid *** We demonstrated engraftment of clinically-collected and thawed PBSC following cryopreservation up to 18 years in NSG mice,sign

Improving Self-Supply of Urban Water and Sanitation Projects through Microfinancing in Nigeria

Most water and sanitation projects in poor neighbourhood of urban Nigeria are executed through self-supply option. This is defined as an incremental improvement of WASH products e.g. water and sanitation structures by a household or small group of households which are financed by their own effort and investment. It is indeed a concept which complements conventional water supply and sanitation funded by government. The aim of this paper is, therefore, to investigate the extent of the use of innovative financing especially group saving approach to improve water and sanitation projects in Awka town, Nigeria. Towards achieving the aim, data were collected through questionnaires which were designed and administered on the respondents between January and April, 2016 and from other secondary sources. Data were analyzed using Principal Component Analysis and Component scores to establish the extent of the use of this option in the 9 component quarters of the town. Result shows that some quarters have perfected using the self-supply option while in others, it is yet to be widely accepted. It is finally recommended among others that the urban water policy as well as urban water safety plan should be revised to include the option as a way of improving domestic water supply and sanitation in the town.

A Preliminary Assessment of the effects of Climate Variability on Agulu Lake, Anambra State, Nigeria

The paper assessed the effect of variability of climatic elements on Agulu lake, Anambra State, Nigeria. Data for the work were acquired from Landsat website (***) for 35 years. Monthly records of the elements were collected from the synoptic meteorological station at Amawbia in Awka urban area. The penman model was used in the estimation of open water vapour from the lake. The analysis was performed using the output of the classified satellite imagery which was digitized for the entire year and the area of the lake was calculated for all the years of study. Statistical analysis of satellite imagery was further employed to analyse trends and relationships. Result shows that the trend is positive and significant at 95% confidence level. Fluctuations in the value of temperature and other variables were removed by 5-year moving average. The Agulu lake surface area was found to have contracted from 0.6177 km2 in 1978 to 0.3583 km2 in 2013. Recommendations on how to ameliorate the problem were made.

Beta-Lactam Antibiotic Resistance among <i>enterobacter</i>spp. Isolated from Infection in Animals

Nosocomial infections are frequent complications of hospitalization, caused by opportunistic pathogens that gain access to hosts undergoing invasive procedures, such as surgery, intubation, and placement of deep vein lines. Nosocomial infections in animal hospitals can infect other animals, as well as be transmitted to human personnel. enterobacter is a genus of common gram-negative bacteria, which can be associated with antibiotic resistant hospital infections. Because of an outbreak in antibiotic resistance in the genus, we decided to investigate five years of enterobacter infections in the Large Animal Services of the Lois Bates Acheson Veterinary Teaching Hospital (LBAVTH) at Oregon State University. The demographics from 37 enterobacter-infected patients of the LBAVTH were obtained from charts and analyzed. The identified clusters of infections suggested possible patient-environment sources of infection. The environment of the hospital was sampled in an attempt to determine the source of infection. Although enterobacter was not isolated, three of the collected samples contained bacteria with resistance to third-generation cephalosporins. enterobacter isolates from six of the 37 patients were further analyzed for presence of specific eSBL resistance genes. All six of the isolates harbored multiple extended-spectrum beta-lactamase genes, i.e., CTX-M-15, TeM-80, SHV-2 and AmpC. In summary, enterobacter infection in the veterinary hospital was caused by beta-lactam-resistant strains, carrying eSBL-resistant genes. Veterinary hospital personnel should be aware of the potential for transmission, to both humans and animals, of eSBL-gene-containing bacteria.

From the early Paleozoic Platforms of Baltica and Laurentia to the Caledonide Orogen of Scandinavia and Greenland

The Caledonide Orogen in the Nordic countries is exposed in Norway, western Sweden, westernmost Finland, on Svalbard and in northeast Greenland. In the mountains of western Scandinavia, the structure is dominated by e-vergent thrusts with allochthons derived from the Baltoscandian platform and margin, from outboard oceanic (Iapetus) terranes and with the highest thrust sheets having Laurentian affinities. The other side of this bivergent orogen is well exposed in northeastern Greenland, where W-vergent thrust sheets emplace Laurentian continental margin assemblages onto the platform. Svalbard's Caledonides are disrupted by late Caledonian faults, but have close affinity with the Laurentian margin in Northeast Greenland. Only Svalbard's Southwestern terrane is foreign to this margin, showing affinity to the Pearya terrane of northern ellesmere Island in arctic Canada. Between the margins of western Scandinavia and eastern Greenland, the wide continental shelves, now covered by late Paleozoic and younger successions, are inferred to be underlain by the Caledonide hinterland, probably incorporating substantial Grenville-age basement. In northernmost Norway, the Ne-trending Caledonian thrust front truncates the NW-trending Neoproterozoic Timanide orogen of northwest Russia. Much of the central and eastern parts of the Barents Shelf are thought to be underlain by Caledonian-deformed Timanide basement. Caledonian orogeny in Norden resulted from. the closure of the Iapetus Ocean and Scandian collision of continent Baltica with Laurentia. Partial subduction of the Baltoscandian margin beneath Laurentia in the mid-late Silurian was followed by rapid exhumation of the highly metamorphosed hinterland in the early Devonian, and deposition of Old Red Sandstones in intramontane basins. Late Scandian collapse of the orogen occurred on major extensional detachments, with deformation persisting into the late Devonian.

The Impact of Urban Flooding on Surface Water Quality of Awka Town

Some physico-chemical and bacteriological parameters were analyzed for in the water samples of four different ponds located in Awka town both prior and after the rain to ascertain the impact of flood in these ponds and to establish whether the pollution was significant. Four samples were collected from each pond both before the rain and after making it a total of thirty-two samples. Twenty-seven parameters were analyzed for in each of the samples. The result revealed that twelve parameters exceeded WHO standard before the rain while fourteen exceeded after the rain. There was significant increment in the values of the analyzed parameters after the rain when compared with the value before the rain as revealed by the Anova statistical tool. It was discovered that there was a major increase in the bacteriological parameters after the rain which implied the serious health impact. It was recommended that users who utilized these ponds for squeezing bitter leaf, processing cassava and washing meats etc. should do so with the caution so as not to expose people to health problems.

采用细胞培养及自由基生化方法,研究正常和过氧化条件下,幼年仓鼠肾成纤维细胞(BHK-21/C13)及其多瘤病毒转化株(BHK-21/PyY)的抗氧化物和脂质过氧化物水平,发现:非转化株的抗氧化物水平,除谷胱甘肽S-转移酶(GST)和谷胱甘肽还原酶(GSHR)外,谷胱甘肽过氧化物酶(GSHPx)、还原型谷胱甘肽(GSH)、硒水平均高于转化株(BHK-21/PyY);培养基中加入维生素e,非转化株比转化株摄入更多的维生素e;而转化株的内源性丙二醛(MDA)的产生多于非转化株。在铁催化的过氧化条件下,除硒、GST和GSHR外,二株细胞内上述抗氧化物指标都显著下降,MDA产生显著增加。除GSHPx外,非转化株的这种上升和下降的变化幅度均大于转化株。提示:自由基代谢水平在正常及恶性细胞之间,至少在上述二株细胞之间有所不同;正常细胞对过氧化的敏感性高于恶性细胞。

The pharmaceutical and anticoagulant agent heparin,a member of the glycosaminoglycan family of carbohydrates,has previously been identified as a potent inhibitor of a key Alzheimer’s disease drug target,the primary neuronalβ-secretase,β-site amyloid precursor protein cleaving enzyme 1(BACe1).The anticoagulant activity of heparin has,however,precluded the repurposing of this widely used pharmaceutical as an Alzheimer’s disease ***,a glycosaminoglycan extract,composed predominantly of 4-sulfated chondroitin sulfate,has been isolated from Sardina pilchardus,which possess the ability to inhibit BACe1(IC50[half maximal inhibitory concentration]=4.8μg/mL),while displaying highly attenuated anticoagulant activities(activated partial thromboplastin time eC50[median effective concentration]=403.8μg/mL,prothrombin time eC50=1.3 mg/mL).The marine-derived,chondroitin sulfate extract destabilizes BACe1,determined via differential scanning fluorimetry(ΔTm–5°C),to a similar extent as heparin,suggesting that BACe1 inhibition by glycosaminoglycans may occur through a common mode of action,which may assist in the screening of glycan-based BACe1 inhibitors for Alzheimer’s disease.
