Study of the mechanisms regulating human umbilical artery contractility

We studied the involvement of different types of Ca2+ channels, cyclic nucleotides and different kinases in the regulation of human umbilical artery (HUA) contractility. The elucidation of the precise mechanisms regulating the contractility of this artery could be very important to reveal potential therapeutic targets to treat HUA disorders such as preeclampsia. The relevancy of different types of Ca2+ channels on the regulation of HUA tonus was analyzed. Among the different Ca2+ channel inhibitors used, only the L-type calcium channels (LTCC) inhibition induced relaxation of HUA in Ca2+ containing medium. The inhibition of T-type calcium channels (TTCC) or TRP channels did not significantly affect HUA contractility. In presence of Ca2+, the intracellular increase of a cyclic nucleotide (cAMP or cGMP) induces relaxation of HUA, which was almost complete in histamine-con- tracted HUA, and lower effect was observed in arteries contracted by KCl and serotonin (5-HT). Inhibition of PKA and PKG weakly reduced the relaxations induced by the increase of cAMP and cGMP respectively, suggesting that the relaxation induced by these nucleotides is not totally mediated by the activation of their respective kinases and that other mechanisms are involved. In calcium containing solution, PP2A inhibition produces relaxation of contracted HUA. In KCl contracted arteries, the OA and nifedipine relaxant effects are similar and not additive, suggesting that PP2A could activate LTCC. Besides, the increase of cyclic nucleotides significantly increased the OA effect, suggesting that the effect of PP2A inhibition is independent of the cyclic nucleotide pathways. The contractions induced by KCl, histamine and 5-HT in presence of Ca2+ were not significantly affected by ROCK, ERK1/2 or p38MAPK inhibitors. Performance of masonry enclosure walls:lessons learned from recent earthquakes

This paper discusses the issue of performance requirements and construction criteria for masonry enclosure and infill walls. Vertical building enclosures in European countries are very often constituted by non-load-bearing masonry walls, using horizontally perforated clay bricks. These walls are generally supported and confined by a reinforced concrete frame structure of columns and beams/slabs. Since these walls are commonly considered to be nonstructural elements and their influence on the structural response is ignored, their consideration in the design of structures as well as their connection to the adjacent structural elements is frequently negligent or insufficiently detailed. As a consequence, nonstructural elements, as for wall enclosures, are relatively sensitive to drift and acceleration demands when buildings are subjected to seismic actions. Many international standards and technical documents stress the need for design acceptability criteria for nonstructural elements, however they do not specifically indicate how to prevent collapse and severe cracking, and how to enhance the overall stability in the case of moderate to high seismic loading. Furthermore, a review of appropriate measures to improve enclosure wall performance and both in-plane and out-of-plane integrity under seismic actions is addressed.

INTRODUCTION Seed dispersal is a key process in the life cycle of most plants,with ecological and evolutionary consequences that go beyond vegetation dynamics and cascade to entire ecosystem functioning and stability(Traveset et al.2014).Seed dispersal largely constrains plant recruitment and the distribution of future plant cohorts,thus playing a critical role in the restoration of disturbed sites(Cain et al.2000;Higgins et al.2003).It can also threaten plant diversity by facilitating plant invasions(Vavra et al.2007;Heleno 2020).

Prehospital critical care drug-therapy and 30-day mortality in patients with acute respiratory disease

BACKGROUND: Prehospital medication is a reality, and the role of these therapies must be explored to assess their validity, especially for acute respiratory diseases, which are usually associated with increased morbidity. The aim of this study was to examine the association of prehospital medication use with mortality in patients with acute respiratory ***: A prospective, multicenter, emergency medical service(EMS) delivery cohort study was carried out in adults with unselected respiratory diseases managed by EMS who were transferred to the emergency department. From January 1, 2019, to October 31, 2023, six advanced life support units, thirty-eight basic life support units, and four hospitals in Spain participated in the study. Demographic data, vital signs, use of mechanical ventilation, prehospital respiratory diagnosis, and prehospital medication were collected. The primary outcome was 30-day in-hospital ***: A total of 961 patients were included, with a mortality rate of 17.5%(168 patients). Age, an increasing number of comorbidities, the use of invasive mechanical ventilation(IMV), the use of major analgesics, hypnotics, and bicarbonate were risk factors. In contrast, elevated systolic blood pressure and Glasgow Coma Scale scores were found to be protective factors against mortality. The predictive capacity of the model reached an area under the curve(AUC) of 0.857(95% confidence interval [95% CI] 0.827–0.888).CONCLUSION: Our data revealed that IMV, major analgesics, hypnotics and bicarbonate administration were associated with elevated mortality. Adding prehospital drug therapy information to demographic variables and vital signs could improve EMS decision-making, allowing a better characterization of patients at risk of clinical worsening.

Fish constitutes the main protein source for the Amazonian ***,the impact of different anthropogenic activities on trace element and metal accumulation in fish and their risks for human health at a regional scale remain largely *** we assessed exposure levels of 10 trace elements andmetals(Cr,Mn,Fe,Ni,Cu,Zn,As,Cd,Pb,and Hg)in 56 samples belonging to 11 different species of fish from the Brazilian *** studied the relationship between exposure levels,fish origin,and fish feeding habits,and assessed toxicological and carcinogenic risks for the Amazonian *** significant correlation was found between sampling site and exposure levels to the studied elements,but a significant difference was found between the accumulation of some metals and the position of the fish species in the food *** concentrations of Cr and Hg in fish flesh were found to exceed the Brazilian limits for human *** study shows that current fish consumption patterns can lead to estimated daily intakes of Hg,As and Cr that exceed the oral reference dose,thus posing a toxicological ***,carcinogenic risks may be expected due to the continued exposure to Cr and *** results of this study show that the consumption of wild caught fish in the Amazon region should be ***,continued monitoring of trace element and metal contamination in fish and on the health of the Amazonian population is recommended,particularly for riverine and indigenous communities.
