The mammalian carboxylesterase 1(ces1/cES1)family comprises several enzymes that hydrolyze many xenobiotic chemicals and endogenous *** investigate the pharmacological and physiological roles of ces1/cES1,we generated ces1 cluster knockout(ces1^(-/-))mice,and a hepatic human cES1 transgenic model in the ces1^(-/-)background(TgcES1).ces1^(-/-)mice displayed profoundly decreased conversion of the anticancer prodrug irinotecan to SN-38 in plasma and ***1 mice exhibited enhanced metabolism of irinotecan to SN-38 in liver and ***1 and hcES1 activity increased irinotecan toxicity,likely by enhancing the formation of pharmacodynamically active ***1^(-/-)mice also showed markedly increased capecitabine plasma exposure,which was moderately decreased in TgcES1 ***1^(-/-)mice were overweight with increased adipose tissue,white adipose tissue inflammation(in males),a higher lipid load in brown adipose tissue,and impaired blood glucose tolerance(in males).These phenotypes were mostly reversed in TgcES1 ***1 mice displayed increased triglyceride secretion from liver to plasma,together with higher triglyceride levels in the male *** results indicate that the carboxylesterase 1 family plays essential roles in drug and lipid metabolism and ***1^(-/-)and TgcES1 mice will provide excellent tools for further study of the in vivo functions of ces1/cES1 enzymes.

Although the frequency of metabolic risk factors for cirrhosis and hepatocellular carcinoma(Hcc)is increasing,chronic hepatitis B(cHB)and chronic hepatitis c(cHc)remain the most relevant risk factors for advanced liver disease *** addition to liver damage,hepatitis B virus(HBV)and hepatitis c virus(HcV)infections are associated with a myriad of extrahepatic manifestations including mixed cryoglobulinaemia,lymphoproliferative disorders,renal disease,insulin resistance,type 2 diabetes,sicca syndrome,rheumatoid arthritislike polyarthritis,and autoantibody ***,the list has grown to include *** of muscle mass or muscle function is a critical feature of malnutrition in cirrhotic patients and has been found in approximately 23.0%-60.0%of patients with advanced liver ***,among published studies,there is significant heterogeneity in the aetiologies of hepatic diseases and measurement methods used to determine *** particular,the interaction between sarcopenia,cHB and cHc has not been completely clarified in a real-world *** can result from a complex and multifaceted virus-host-environment interplay in individuals chronically infected with HBV or ***,in the present review,we provide an overview of the concept,prevalence,clinical relevance,and potential mechanisms of sarcopenia in patients with chronic viral hepatitis,with an emphasis on clinical outcomes,which have been associated with skeletal muscle loss in these patients.A comprehensive overview of sarcopenia in individuals chronically infected with HBV or HcV,independent of the stage of the liver disease,will reinforce the necessity of an integrated medical/nutritional/physical education approach in the daily clinical care of patients with cHB and cHc.

chemiluminescence detection was developed as an alternative to amperometric detection for glucose analysis in a portable, microfluidics-based continuous glucose monitoring system. Amperometric detection allows easy determination of hydrogen peroxide, a product of the glucose oxidase-catalyzed reaction of glucose with oxygen, by oxidation at a microelectrode. However, (micro)electrodes in direct contact with physiological sample are subject to electrode fouling, which leads to signal drift, decreased reproducibility and shortened detector lifetimes. Moreover, there are a few species present in the body (e.g. ascorbic acid, uric acid) which can undergo oxidation at the same applied potential as hydrogen peroxide. These species can thus interfere with the glucose measurement, reducing detection specificity. The rationale for exploring chemiluminescence as opposed to amperometric detection is thus to attempt to improve the lifetime and reproducibility of glucose analysis for monitoring purposes, while reducing interference caused by other chemicals in the body. The study reported here represents a first step in this direction, namely the realization of a microfluidic device with integrated silicon photodiode for chemiluminescence detection of glucose. This microflow device uses a chaotic mixing approach to perform enzymatic conversion of glucose, followed by reaction of the hydrogen peroxide produced with luminol to produce light at 425 nm. The chemiluminescence reaction is catalyzed by horseradish peroxidase in the presence of iodophenol. The performance of the fabricated chip was characterized to establish optimal reaction conditions with respect to sample and reagent flow rates, pH, and concentrations. A linear calibration curve was obtained for current response as a function of glucose concentration in the clinically relevant range between 2 and 10 mM, with a sensitivity of 39 pA/mM (R = 0.9963, one device, n = 3) and a limit of detection of 230 μM (S/N = 3).

Sap flow for beech(Fagus sylvatica L.)in a natural and a managed forest—effect of spatial heterogeneity

[目的/意义]标准竞争优势的形成实质上是动态能力发挥作用的结果。从过程和内容视角构建基于资源和能力的整合框架,不仅是应对环境不确定性和复杂性的必然选择,也是改善组织绩效的现实要求。[方法/过程]在明晰关键知识构成要素的基础之上,阐述了动态知识管理能力的特征和影响因素。由“知识传导—动态能力—企业成长”的逻辑关系入手,从流程、位置和路径三个层面构建了动态知识管理能力的整合分析框架,揭示了动态知识管理能力的形成机理,并进一步归纳了其提升路径。[结果/结论]研究发现,标准知识宽度、标准知识深度与知识整合机制有效解释了动态知识管理能力的提升机理,动态知识管理能力的提升路径主要包括基础层、转化层和应用层,以此为企业获取技术标准竞争优势提供了理论依据。
