A new hemostatic agent composed of Zn^(2+)-enriched Ca^(2+)alginate activates vascular endothelial cells in vitro and promotes tissue repair in vivo

To control capillary bleeding, surgeons may use absorbable hemostatic agents, such as Surgicel® and TachoSil®. Due to their slow resorption, their persistence in situ can have a negative impact on tissue repair in the resected organ. To avoid complications and obtain a hemostatic agent that promotes tissue repair, a zinc-supplemented calcium alginate compress was developed: HEMO-IONIC®. This compress is non-absorbable and is therefore removed once hemostasis has been achieved. After demonstrating the hemostatic efficacy and stability of the blood clot obtained with HEMO-IONIC, the impact of Surgicel, TachoSil, and HEMO-IONIC on cell activation and tissue repair were compared (i) in vitro on endothelial cells, which are essential to tissue repair, and (ii) in vivo in a mouse skin excision model. In vitro, only HEMO-IONIC maintained the phenotypic and functional properties of endothelial cells and induced their migration. In comparison, Surgicel was found to be highly cytotoxic, and TachoSil inhibited endothelial cell migration. In vivo, only HEMO-IONIC increased angiogenesis, the recruitment of cells essential to tissue repair (macrophages, fibroblasts, and epithelial cells), and accelerated maturation of the extracellular matrix. These results demonstrate that a zinc-supplemented calcium alginate, HEMO-IONIC, applied for 10 min at the end of surgery and then removed has a long-term positive effect on all phases of tissue repair.

Atm deficient zebrafish model reveals conservation of the tumour suppressor function and a role in fertility

Biallelic loss-of-function variants in ATM (Ataxia Telangiectasia Mutated) cause Ataxia Telangiectasia (AT), a rare disorder associated with cerebellar degeneration and ataxia, cancer predisposition, infertility, growth retardation, etc. ATM is a phosphoinositide 3-kinase-related kinase (PIKK) with a role in DNA repair and maintenance of genome stability. Studying a multisystem genetic disease like AT requires animal models to ascertain its pathogenesis at the level of tissues, organs and the organism. Due to its small size, cheap maintenance, large progeny, rapid development and initial transparency, zebrafish (Danio rerio) is an increasingly popular vertebrate model organism, suitable for genetic modifications and large-scale in vivo therapeutic screens as embryos are chemically permeable to small compounds. Currently, no zebrafish model for AT exists.1 We generated atm knock-outs through CRIPSR-Cas9 mutagenesis. We show that atm conserved its function as a tumour suppressor gene and is involved in gametogenesis and fertility. Therefore, this mutant is of great value for further studies investigating the role of atm in reproduction and tumorigenesis.

Plant functional and phylogenetic turnover correlate with climate and land use in the Western Swiss Alps

Aims Understanding the relative importance of historical and environ-mental processes in the structure and composition of communities is one of the longest quests in ecological research.Increasingly,researchers are relying on the functional and phylogeneticβ-diversity of natural communities to provide concise explanations on the mechanistic basis of community assembly and the drivers of trait variation among species.The present study investigated how plant functional and phylogeneticβ-diversity change along key environmental and spatial gradients in the Western Swiss Alps.Methods Using the quadratic diversity measure based on six functional traits-specific leaf area,leaf dry matter content,plant height,leaf carbon content,leaf nitrogen content and leaf carbon to nitrogen content alongside a species-resolved phylogenetic tree-we relate variations in climate,spatial geographic,land use and soil gradients to plant functional and phylogenetic turnover in mountain commu-nities of the Western Swiss Alps.Important Findings Our study highlights two main points.First,climate and land-use factors play an important role in mountain plant community turnover.Second,the overlap between plant functional and phy-logenetic turnover along these gradients correlates with the low phylogenetic signal in traits,suggesting that in mountain land-scapes,trait lability is likely an important factor in driving plant community assembly.Overall,we demonstrate the importance of climate and land-use factors in plant functional and phyloge-netic community turnover and provide valuable complementary insights into understanding patterns ofβ-diversity along several ecological gradients.

Bioenergy,currently the largest renewable energy source in the EU(64%of the total renewable energy consumption),has sparked great interest to meet the32%renewable resources for the 2030 bioeconomy goal.The design of innovative cropping systems informed by bioeconomy imperatives requires the evaluate of the effects of introducing crops for bioenergy into conventional crop rotations.This study aimed to assess the impacts of changes in conventional cropping systems in mixed dairy cattle farms redesigned to introduce bioenergy crops either by increasing the biomass production through an increase of cover crops,while keeping main feed/food crops,or by substituting food crops with an increase of the crop rotation length.The assessment is based on the comparison between conventional and innovative systems oriented to feed and biogas production,with and without tillage,to evaluate their agri-environmental performances(biomass production,nitrogen fertilization autonomy,greenhouse gas emissions and biogas production).The result showed higher values in the biogas cropping system than in the conventional and feed ones for all indicators,biomass productivity(27%and20%higher,respectively),nitrogen fertilization autonomy(26%and 73%higher,respectively),methanogenic potential(77%and 41%higher,respectively)and greenhouse gas emissions(15%and 3%higher,respectively).There were no negative impacts of no-till compared to the tillage practice,for all tested variables.The biogas cropping system showed a better potential in terms of agri-environmental performance,although its greenhouse gas emissions were higher.Consequently,it would be appropriate to undertake a multicriteria assessment integrating agri-environmental,economic and social performances.

Objective:In patients with head and neck squamous cell carcinoma(HNSCC),cetuximab[a monoclonal antibody targeting epidermal growth factor receptor(EGFR)]has been shown to improve overall survival when combined with radiotherapy in the locally advanced setting or with chemotherapy in first-line recurrent and/or metastatic(R/M)setting,respectively.While biomarkers of resistance to cetuximab have been identified in metastatic colorectal cancer,no biomarkers of efficacy have been identified in HNSCC.Here,we aimed to identify biomarkers of cetuximab sensitivity/resistance in HNSCC.Methods:HNSCC patients treated with cetuximab at the Curie Institute,for whom complete clinicopathological data and formalin-fixed paraffin-embedded(FFPE)tumor tissue collected before cetuximab treatment were available,were included.Immunohistochemistry analyses of PTEN and EGFR were performed to assess protein expression levels.PIK3 CA and H/N/KRAS mutations were analyzed using high-resolution melting(HRM)and Sanger sequencing.We evaluated the predictive value of these alterations in terms of progression-free survival(PFS).Results:Hot spot activating PIK3 CA and KRAS/HRAS mutations were associated with poor PFS among HNSCC patients treated with cetuximab in the first-line R/M setting,but not among HNSCC patients treated with cetuximab in combination with radiotherapy.Loss of PTEN protein expression had a negative predictive value among HNSCC patients treated with cetuximab and radiotherapy.High EGFR expression did not predict cetuximab sensitivity in our patient population.Conclusions:Hot spot activating PIK3 CA and RAS mutations predicted cetuximab resistance among HNSCC patients in the firstline R/M setting,whereas loss of PTEN protein expression predicted resistance to cetuximab when combined to radiotherapy.

Survey of recent advances of in the field of π-conjugated heterocyclic azomethines as materials with tuneable properties

This account gives an overview of our recent work in the area of conjugated azomethines derived from 2-aminothiophenes.It will be presented that mild reaction conditions can be used to selectively prepare symmetric and unsymmetric conjugated azomethines.It further will be demonstrated that azomethines consisting of various 5-membered aryl heterocycles lead to chemically,reductively,hydrolytically,and oxidatively robust compounds.The optical and electrochemical properties of these materials can be tuned contingent on the degree of conjugation,type of aryl heterocycle,and by including various electronic groups.The end result is materials having colors spanning 250 nm across the visible spectrum.These colors further can be tuned via electrochemical or chemical doping.The resulting doped states have high color contrasts from their corresponding neutral states.The collective opto-electronic properties and the means to readily tune them,make thiophenoazomethine derivatives interesting materials for potential use in a gamut of applications.

Long term outcome and quality of life after pelvic exenteration for recurrent endometrial and cervical cancers

Introduction: Pelvic exenteration can cure recurrent gynecological malignancies. However, treatment morbidity is over 50% in radiated pelves. We evaluated the outcome, the morbidity and the quality of life after exenteration. Methods: Patients who underwent an exenteration for recurrent cervical or endometrial cancer in our institution between 1999 and 2011 were retrieved. Survival rates were calculated according to Kaplan-Meier method and compared using the logrank test. The EORTC QLQ-C30 and QLQ-CX24 questionnaires were sent to patients alive in January 2012. Results: We identified 25 patients: 17 (68%) had cervical cancer and 8 (32%) endometrial cancer. 92% of them had received radiotherapy previously. All patients had a central pelvic recurrence, within a median time of 30 months [4 - 384] from initial treatment. Early complications requiring a re-laparotomy occurred in 9 patients (36%). Late complications included 2 (8%) fistulas, 2 (8%) occlusion and 1 (4%) ureteral stenosis. Complete resection was obtained in 92% of patients. Disease Free and Overall survival rates were better in cervical rather than in endometrial cancer (median DFS in months 17 [2 - 145] vs 9.5 [3 - 21], p = 0.064, median OS in months 20 [2 - 145] vs 13 [4 - 42], p = 0.019). 69% of patients answered the quality of life questionnaires. Mean global quality of life score was 45 on a scale of 0 - 100, none of the patients had a sexual activity. Conclusions: Morbidity of exenteration remains high and quality of life is altered. Endometrial cancer is associated with a poorer prognosis. In those patients, exenteration should be put in balance with best supportive care.
