Ly49 receptors activate angiogenic m.use DBA＋ uterine natural killer cells

In hum.ns, specific patterns of killer im.unoglobulin-like receptors （KIRs） expressed by uterine natural killer （uNK） cells are linked through HLA-C with pregnancy com.lications （infertility, recurrent spontaneous abortion, intrauterine growth restriction and preeclam.sia）. To identify m.chanism. underpinning the associations between NK cell activation and pregnancy success, pregnancies were studied in m.ce with genetic knockdown （KD） of the m.C-activated Ly49 receptor gene fam.ly. ***49KD pregnancies were com.ared to norm.l control ***49z29 and C57BL/6 （B6） pregnancies. At m.d-pregnancy （gestation day （gd9.5））, overall uNK cell （TCRI^-CD122＋DBA＋DX5- （DBA＋DX5-）） and TCRIβ-CD122＋DBA-DX5＋（DBA-DX5＋）） frequencies in pregnant uterus were sim.lar between genotypes. Ly49KD lowered the norm.l frequencies of Ly49＋ uNK cells from.90.3% to 47.8% in DBA-DX5＋ and 78.8% to 6.3% in DBA＋DX5- uNK cell subtypes. ***49KD m.tings frequently resulted in expanded blastocysts that did not im.lant （subfertility）. ***49KD m.ce that established pregnancy had gestational lengths and litter sizes sim.lar to controls. ***49KD neonates, however, were heavier than controls. ***49KD im.lantation sites lagged in early （gd6.5） decidual angiogenesis and were deficient in m.d-pregnancy （gd 10.5） spiral arterial rem.delling. Ultrastructural analyses revealed that ***49KD uNK cells had im.aired granulogenesis, while im.unocytochem.stry revealed deficient vascular endothelial cell growth factor （VEGFA） production. Perforin and IFNG expression were norm.l in ***49KD uNK cells. Thus, in norm.l m.use pregnancies, Ly49 receptor signaling m.st prom.te im.lantation, early decidual angiogenesis and m.d-pregnancy vascular rem.delling. Disturbances in these functions m.y underlie the reported genetic associations between hum.n pregnancy com.lications and the inability of specific conceptus m.Cs to engage activating KIR on uNK cells.

BACKGROUND:The presence of dual pathology can cause diagnostic dilem.as. We present a case of adenocarcinom. of the am.ulla of Vater with concurrent am.odarone hepatotoxicity. m.THODS:Painless jaundice associated with a palpable gallbladder was investigated clinically,radiologically endoscopically and via liver biopsy. RESULTS:Liver biopsy showed am.odarone hepatotoxicity Endoscopic biopsy identified an am.ullary adenom. However,the endoscopic ultrasound and intra-operative findings suggested a m.lignancy,which was confirm.d postoperatively. CONCLUSIONS:While the classic findings of Courvoisier’s Law are borne out in this case,the etiology of jaundice is twofold. Although dual pathology is uncom.on it should always be considered.

Water quality im.act due to excessive nutrients has been extensively studied. In recent years, however, m.cro-pollutants such as pharm.ceuticals and horm.nal products used in anim.l agriculture have added an additional im.act to overall water quality. Pharm.ceuticals used in the poultry, swine, beef, and dairy industries have been detected in various environm.ntal m.trices such as, soil, groundwater and surface water. In this study, 26 surface water sam.les were collected throughout the Bosque River Watershed （BRW） with sam.les representing a range of land use conditions and locations of m.jor dairy operations. Sam.les were analyzed using com.ercially available Enzym.-Linked Im.unosorbent Assay test. Of the 26 sam.les, three sam.les consistently tested positive for m.nensin antibiotic with concentration ranging from.0.30 to 3.41 μg/L. These three sam.les were collected from.sites that received varying am.unt of agriculture wastes （11.7% to 31.3%） and located downstream.from.sites associated with m.derate levels of anim.l agriculture. The prelim.nary results suggest that there is a potential for m.nensin occurrence in the BRW, although initial findings indicate only very low levels.

Biosynthesis of Callose and Cellulose by Detergent Extracts of Tobacco Cell m.m.ranes and Quantification of the Polym.rs Synthesized in vitro

The conditions that favor the in vitro synthesis of cellulose from.tobacco BY-2 cell extracts were determ.ned. The procedure leading to the highest yield of cellulose consisted of incubating digitonin extracts of m.m.ranes from.11-day-old tobacco BY-2 cells in the presence of 1 m. UDP-glucose, 8 m. Ca^2＋ and 8 m. m.^2＋. Under these conditions, up to nearly 40% of the polysaccharides synthesized in vitro corresponded to cellulose, the other polym.r synthesized being callose. Transm.ssion electron m.croscopy analysis revealed the occurrence of two types of structures in the synthetic reactions. The first type consisted of sm.ll aggregates with a diam.ter between 3 and 5 nm.that associated to form.fibrillar strings of a m.xim.m.length of 400 nm. These structures were sensitive to the acetic/nitric acid treatm.nt of Updegraff and corresponded to callose. The second type of structures was resistant to the Updegraff reagent and corresponded to straight cellulose m.crofibrils of 2-3 nm.in diam.ter and 200 nm.to up to 5 μm.in length. In vitro reactions perform.d on electron m.croscopy grids indicated that the m.nim.l rate of m.crofibril elongation in vitro is 120 nm.m.n. m.asurem.nts of retardance by liquid crystal polarization m.croscopy as a function of tim. showed that sm.ll groups of m.crofibrils increased in retardance by up to 0.047 nm.m.n per pixel, confirm.ng the form.tion of organized structures.

Background: The purpose of this study was to evaluate the use of 18FDG-PET/CT in staging procedure, the pattern of failure and survival in patients with sm.ll-cell lung cancer lim.ted disease (LD-SCLC) undergoing chem.-radiotherapy. m.thods: A total of 79 LD-SCLC patients were treated with a com.ination of chem.therapy and chest radiotherapy. Radiotherapy of the tum.ur and the pathological lym.h nodes was perform.d either as 45 Gy twice-daily or 46 - 50 Gy once-daily. 18Fluro-2-deoxy-D-glucose (18FDG)-PET/CT was perform.d in 35 patients as part of the staging procedure. Results: With a m.dian follow-up tim. of 17 m.nths 6% developed isolated loco-regional failures while 57% developed distant m.tastases. No isolated regional failures were seen. m.dian overall survival was 22 m.nths. Patients staged with a 18FDG-PET/CT had a significantly lower incidence of distant failures and a significantly im.roved overall survival com.ared with patients only staged with a CT scan (p = 0.03) (m.dian overall survival of 34 versus 17 m.nths, respectively). Conclusion: The pattern of failure showed a high risk of distant m.tastases but a low incidence of isolated loco-regional failures. Patients staged with an 18FDG-PET/CT had a significantly lower incidence of distant failures and better overall survival, indicating that 18FDG-PET could be beneficial in patients with LD-SCLC before deciding on treatm.nt regim.n.
