Polymorphisms in NF-κB,PXR,LXR,PPARγ and risk of inflammatory bowel disease

AIM:To investigate the contribution of polymorphisms in nuclear receptors to risk of inflammatory bowel disease(IBD) . METHODS:Genotypes of nuclear factor(NF) -κB(NFKB1) NFκB-94ins/del(rs28362491) ;peroxisome proliferatoractivated receptor(PPAR) -γ(PPARγ) PPARγPro12Ala(rs1801282) and C1431T(rs 3856806) ;pregnane X receptor(PXR) (NR1I2) PXR A-24381C(rs1523127) ,C8055T(2276707) ,and A7635G(rs 6785049) ;and liver X receptor(LXR) (NR1H2) LXR T-rs1405655-C and T-rs2695121-C were assessed in a Danish case-control study of 327 Crohn's disease patients,495 ulcerative colitis(UC) patients,and 779 healthy *** ratio(OR) and 95%CI were estimated by logistic regression models. RESULTS:The PXR A7635G variant,the PPARγPro12Ala and LXR T-rs2695121-C homozygous variant genotypes were associated with risk of UC(OR:1.31,95% CI:1.03-1.66,P=0.03,OR:2.30,95%CI:1.04-5.08,P=0.04,and OR:1.41,95%CI:1.00-1.98,P=0.05,respectively) compared to the corresponding homozygous wild-type *** never smokers,PXR A7635G and the LXR T-rs1405655-C and T-rs2695121-C variant genotypes were associated with risk of IBD(OR:1.41,95%CI:1.05-1.91,P=0.02,OR:1.63,95%CI:1.21-2.20,P=0.001,and OR:2.02,95%CI:1.36-2.99,P=0.0005,respectively) compared to the respective homozygous variant *** A7635G(rs6785049) variant genotype was associated with a higher risk of UC diagnosis before the age of 40 years and with a higher risk of extensive disease(OR:1.34,95%CI:1.03-1.75 and OR:2.49,95%CI:1.24-5.03,respectively) . CONCLUSION:Common PXR and LXR polymorphisms may contribute to risk of IBD,especially among never smokers.

Dear Editor,In the European region,which shares 22.8%of the global cancer burden for 10%of the global population,there were around 4.4 million new cancer cases and 1.9 million deaths from cancer in 2020[1].The reasons for the high cancer incidence rates are complex;however,diet and dietary components are among the main contributors to cancer risk[2].In modern-day living,a growing proportion of people include in their diets ultra-processed *** of food processing and home-prepared foods are so-called dietary advanced glycation endproducts(AGEs),which are reactive metabolites emerging during the breakdown of reducing *** production is preponderant in dry high-heat processes(e.g.,baking,roasting);hence foods such as cakes,crisps,crackers,cereal products,meat and meat-derived products represent a major source of dietary AGEs[3].

Combinatorial Complexity in a Transcriptionally Centered Signaling Hub in Arabidopsis

A subfamily of four Phytochrome （phy）-Interacting bHLH transcription Factors （PIFs） collectively promote skotomorphogenic development in dark-grown seedlings. This activity is reversed upon exposure to light, by photoacti- vated phy molecules that induce degradation of the PIFs, thereby triggering the transcriptional changes that drive a tran- sition to photomorphogenesis. The PIFs function both redundantly and partially differentially at the morphogenic level in this process, To identify the direct targets of PIF transcriptional regulation genome-wide, we analyzed the DNA-binding sites for all four PIFs by ChlP-seq analysis, and defined the genes transcriptionally regulated by each PIF, using RNA-seq analysis of pif mutants. Despite the absence of detectable differences in DNA-binding-motif recognition between the PIFs, the data show a spectrum of regulatory patterns, ranging from single PIF dominance to equal contributions by all four. Similarly, a broad array of promoter architectures was found, ranging from single PIF-binding sites, containing single sequence motifs, through multiple PIF-binding sites, each containing one or more motifs, with each site occupied prefer- entially by one to multiple PIFs. Quantitative analysis of the promoter occupancy and expression level induced by each PIF revealed an intriguing pattern. Although there is no robust correlation broadly across the target-gene population, examination of individual genes that are shared targets of multiple PIFs shows a gradation in correlation from strongly positive, through uncorrelated, to negative. This finding suggests a dual-layered mechanism of transcriptional regulation, comprising both a continuum of binding-site occupancy by each PIF and a superimposed layer of local regulation that acts differentially on each PIF, to modulate its intrinsic transcriptional activation capacity at each site, in a quantitative pattern that varies between the individual PIFs from gene to gene. This paper presents an application of the sparse Bayesian learning(SBL)algorithm to linear inverse problems with a high order total variation(HOTV)sparsity *** the problem of sparse signal recovery,SBL often produces more accurate estimates than maximum a posteriori estimates,including those that useℓ1 ***,rather than a single signal estimate,SBL yields a full posterior density estimate which can be used for uncertainty ***,SBL is only immediately applicable to problems having a direct sparsity prior,or to those that can be formed via *** paper demonstrates how a problem with an HOTV sparsity prior can be formulated via synthesis,and then utilizes *** expands the class of problems available to Bayesian learning to include,e.g.,inverse problems dealing with the recovery of piecewise smooth functions or signals from *** examples are provided to demonstrate how this new technique is effectively employed.

Green roof technology and implementation are taking root in North America at an accelerating *** recogni-tion of the benefits of green roofs and increasing interest in green infrastructure are leading to expansion of green roof tech-nologies that have been in use for decades in Europe and *** some regions have adopted the use of green roofs on a large scale,other areas are warming up to the concept more ***-scale implementation of green roofs has not yet occurred in Indiana,but a number of exemplary projects have been constructed,and there are signs that interest in the technology is increasing in the *** purpose of this article is to provide an overview of green roof technology,analyze selected green roofs in Indiana,explore trends in the state,and address issues for future development of green roof technol-ogy in the region.A variety of green roofs were investigated throughout the *** were held with individuals involved in each project to obtain technical and logistical details of green roof design,installation,and performance.

Cellulose Synthases and Synthesis in Arabidopsis

Plant cell walls are complex structures composed of high-molecular-weight polysaccharides, proteins, and lignins. Among the wall polysaccharides, cellulose, a hydrogen-bonded β-1,4-1inked glucan microfibril, is the main load-bearing wall component and a key precursor for industrial applications. Cellulose is synthesized by large multi-meric cellulose synthase （CesA） complexes, tracking along cortical microtubules at the plasma membrane. The only known components of these complexes are the cellulose synthase proteins. Recent studies have identified tentative interaction partners for the CesAs and shown that the migratory patterns of the CesA complexes depend on phosphorylation status. These advances may become good platforms for expanding our knowledge about cellulose synthesis in the near future. In addition, our current understanding of cellulose chain polymerization in the context of the CesA complex is discussed.

Science Fiction as a Manifestation of Culture in America

Culture is broadly viewed as all the human activities that define us as a species. ″Sturgeon’s Law″ (that 90% of everything is inferior) is introduced to defend the study of fiction, and science fiction in particular, in higher education as a way of understanding ourselves better, and perhaps improving the world through the increased understanding of tomorrow’s leaders. Several ways of reading particular works of science fiction are examined, like providing historical contexts for readers, close reading and structural analysis, and application of theories based on particular ideologies or from other disciplines in the social and political sciences (such as feminist theory, Marxist theory, psychological theory, etc.). A conclusion is drawn that for literary analysis, a hybrid methodology, one devised individually for each text, works best.
