All living organisms share similar reactions within their central metabolism to provide precursors for all essential building blocksand reducing power. To identify whether alternative metabolic routes of glycolysis can operate in E. coli, we complementarilyemployed in silico design, rational engineering, and adaptive laboratory evolution. First, we used a genome-scale model andidentified two potential pathways within the metabolic network of this organism replacing canonical Embden-Meyerhof-Parnas(EMP) glycolysis to convert phosphosugars into organic acids. One of these glycolytic routes proceeds via methylglyoxal andthe other via serine biosynthesis and degradation. Then, we implemented both pathways in E. coli strains harboring defectiveEMP glycolysis. Surprisingly, the pathway via methylglyoxal seemed to immediately operate in a triosephosphate isomerasedeletion strain cultivated on glycerol. By contrast, in a phosphoglycerate kinase deletion strain, the overexpression ofmethylglyoxal synthase was necessary to restore growth of the strain. Furthermore, we engineered the “serine shunt” whichconverts 3-phosphoglycerate via serine biosynthesis and degradation to pyruvate, bypassing an enolase deletion. Finally, toexplore which of these alternatives would emerge by natural selection, we performed an adaptive laboratory evolution studyusing an enolase deletion strain. Our experiments suggest that the evolved mutants use the serine shunt. Our study reveals theflexible repurposing of metabolic pathways to create new metabolite links and rewire central metabolism.

Background: In caribbean space stroke incidence is 50% higher than in Europe. This study aims to describe characteristics of intracerebral hemorrhage in Guadeloupe. Methods: This was a retrospective study of hospitalized patients with a no-traumatic intracerebral hemorrhage during one year from 2013 to 2014. Survivors were contacted by telephone at the end of the study and;if accept, vital status and Rankin scale were assessed. Results: In one year, 1418 patients were hospitalized for an acute stroke. 112 had no-traumatic intracerebral hemorrhage (7.9%). The sex ratio F/h was 0.72. Mean age of the population was at 63.0 ± 14.7 years. hypertension and diabetes were the leading risk factors notice with respectively 62 and 25 patients. Intracerebral hemorrhage distribution was: deep 48.2%, lobar 30.4%, sub-tentorial 17.9%, meningeal 3.5%. Severity signs distribution were mass effect 79%, ventricular contamination 43% and cerebral herniation 37%. Mean during of in-hospital stay was 10.6 ± 10.3 days and in-hospital mortality was 22 patients (19.6%). A year and a half after emergency 5 additional death (4.5%) between 35 patients survivors was noticed with median Rankin scale 1.5. Conclusion: Intracerebral hemorrhage is less frequent in African American as compared to Caucasian;the epidemiological excess of strokes in this population is related to ischemic strokes.

Background: The adipose tissue mainly consists of adipocytes but also contains non-adipose cells. Among them, progenitor cells represent a local pool of immature cells that, in vitro, can undergo various lineage differentiation processes. These cells are thought to contribute to normal homeostasis of the adipose tissue through adipogenesis but also to the growth of the adipose tissue under chronic energy overload. The aim of the present study is to evaluate in vitro the capacity of a Celosia cristata extract to impact the adipogenic potential of native human adipose tissue progenitor cells, i.e. commitment and differentiation towards adipogenic lineage. Methods: Native adipose tissue progenitor cells were isolated by immunoselection/depletion approaches from human subcutaneous adipose tissues. Two distinct cell culture conditions were used to assess the effect of Celosia cristata extract on commitment and differenciation of progenitor cells. Cells were cultured either in differentiation medium for 10 days in the presence/absence of Celosia cristata extracts to study the impact on differentiation or first cultured in a commitment-inducing medium, with or without Celosia cristata extract, for 48 h and then cultured 10 days in differentiation medium to assess the impact on commitment. In both experimental series, the fate of progenitor cells was studied by quantification of lipids and by determining the expression of key genes involved in adipogenesis. Results: Data show that Celosia cristata extract reduces lipid content of progenitor cells undergoing differentiation. This reduction correlates with a reduced expression of C/EBPα. When progenitor cells are placed in commitment-inducing conditions, Celosia cristata extract induces a more potent reduction of lipid content. This reduction correlates with a decrease in the expression levels of master genes involved in adipogenesis: the genes of transcription factors PPARγ2 and C/EBPα as well as marker genes coding for LPL

采用湿热预处理(195℃,15min)与同步糖化发酵对玉米秸秆制备酒精进行了研究。结果表明:玉米秸秆经过湿热预处理后,86.5%纤维素保留在滤饼中,而大部分半纤维素被溶解。在底物质量体积含量50g/L,温度30℃,ph值5.5,摇床转速130r/min条件下,树干毕赤酵母(Pichia stipitis)利用预处理后的玉米秸秆经过192h同步糖化发酵,酒精浓度达到了12.12g/L,对应的酒精产量和生产效率分别为0.34g/g(葡萄糖+木糖)和0.065g/(L·h)。该项研究为工业化生产打下了基础。

Soybean seed protein, oil, fatty acids, and mineral composition as influenced by soybean-corn rotation

Effects of crop rotation on soybean (Glycine max (L) Merr.) seed composition have not been well investigated. Therefore, the objective of this study was to investigate the effects of soybean-corn (Zea mays L.) rotations on seed protein, oil, and fatty acids composition on soybean. Soybeans were grown at Stoneville, MS, from 2005 to 2008 in five different scheduled cropping sequences. In 2007, following three years of rotation with corn, seed oleic acid percentage was significantly higher in any crop rotation than continuous soybean. The increase of oleic fatty acid ranged from 61 to 68% in 2007, and from 27 to 51% in 2008, depending on the rotation. The increase of oleic acid was accompanied by significant increases in seed concentrations of phosphorus (P), iron (Fe), and boron (B). In 2007, the increase of P ranged from 60 to 75%, Fe from 70 to 72%, and B from 34 to 69%. In 2008, the increase of P ranged from 82 to 106%, Fe from 32 to 84%, and B from 62 to 77%. Continuous soybean had higher linoleic:oleic ratio and linoleic: palmitic + stearic + oleic ratio, indicating that relative quantity of linoleic acid decreased in rotated crops. The total production of protein, oil, stearic and oleic fatty acids was the lowest in continuous soybean. The total production of palmitic acid was inconsistent across years. The results show that soybean- corn rotation affects seed composition by consistently increasing seed oleic fatty acid, P, Fe, and B concentrations. higher oleic acid, unsaturated fatty acid, is desirable for oil stability and long-shelf storage. The mechanisms of how these nutrients are involved are not yet understood.

AIM: To study the benef icial effects of triterpene α,β-amyrin and the underlying mechanisms in an experimental pancreatitis model. METhODS: Acute pancreatitis was induced in five groups of rats (n = 8) by L-arginine (2 × 2.5 g/kg, intraperitoneal, 1 h apart) and 1 h later, they received a single oral dose of α,β-amyrin (10, 30 and 100 mg/kg),methylprednisolone (30 mg/kg) and vehicle (3% Tween 80). A saline (0.9% NaCl) treated group served as a normal control. Efficacy was assessed at 24 h by determination of serum levels of amylase, lipase and proinflammatory cytokines [tumor necrosis factor (TNF)-α and interleukin (IL)-6], pancreatic myeloperoxidase (MPO) activity, lipid peroxidation [thiobarbituric acid reactive substances (TBARS)], nitrate/nitrite levels, and the wet weight/body weight ratio. Tissue histology and the immunoreactivity for TNF-α and inducible nitric oxide synthetase (iNOS) were performed. RESULTS: α,β-amyrin and methylprednisolone treatments significantly (P he L-arginine-induced increases in pancreatic wet weight/body weight ratio, and decreased the serum levels of amylase and lipase, and TNF-α and IL-6, as compared to the vehicle control. Also, pancreatic levels of MPO activity, TBARS, and nitrate/nitrite were signifi cantly lower. histological f indings and TNF-α and iNOS immunostaining further confirmed the amelioration of pancreatic injury by α,β-amyrin. CONCLUSION: α,β-amyrin has the potential to combat acute pancreatitis by acting as an anti-in? ammatory and antioxidant agent.

In a sustainable development context, the monitoring systems are essential to study the building energy performances. With the recent technology advances, these systems can be based on wireless sensor networks, where the energy efficiency is the main design challenge. To this end, most of the studies focus on low power Medium Access Control (MAC) protocols to reduce the overall energy consumption of a network. Nevertheless, the performances assessment of these protocols is generally not performed in a realistic way, and does not take into account the performances of the other layers of the OSI model. In this paper, we propose a cross-layer methodology to assess the real performances of a MAC protocol by taking into account the traffic volume, the synchronization losses and more particularly the physical layer performances through a Bit Error Rate (BER) criterion. The simulation results demonstrate clearly the physical layer impact on a sensor lifetime. Finally, the proposal of an energy efficient MAC protocol for a wireless sensor network dedicated to an application of building monitoring is proposed.

Objectives: To evaluate the long-term outcome of adolescents with anorexia nervosa and attempt to define prognostic factors for outcome. Methods: Transversal, descriptive epidemiological study with a strict methodology: minimum interval of 4 years since first admission, evaluation using clearly defined diagnostic criteria (EAT-40 self-evaluation, hSCL, global patient-driven self-evaluation and hetero evaluation: clinical interview and LECE). The 198 subjects are former adolescent anorexics treated at the hôpital d’Enfants (Children’s hospital) in Nancy (France). Results: Clinical evaluation of the 144 subjects traced showed positive outcome for 68.1%, intermediate outcome for 23.2% and poor outcome for 8.7%. 5 factors for a good prognosis were found (middle socioeconomic class of parents, no prior treatment, hospitalization > 1 month,
