Clinical Assessment of a Mesotherapy Formulation for Skin Rejuvenation in Healthy Volunteers

Mesotherapy is a mildly invasive procedure involving several subcutaneous drug injections, which are performed simultaneously. The concept is to inject a very small amount of active compounds (hyaluronic acid, vitamins, antioxidants, enzymes, humectants, …) and the aim is to stimulate the biosynthetic capacity of fibroblasts, increase collagen and elastin production and to give active essential molecules for a fibroblasts environment. Numerous studies suggest that the mesotherapy procedure improves skin hydration and mechanical properties;however, some results have not been statistically significant. In this study, we investigated an antiaging treatment as a complementary approach to a non-surgical rejuvenating procedure after 5 sessions of mesotherapy. Our main objective was to assess the improvement of face, neck and neckline skin quality, after intradermal microinjections of NCTF 135HA®. This mixture, manufactured by FILORGA Laboratories, includes a non-cross-linked hyaluronic acid, vitamins, antioxidants, mineral salts and enzymes. A statistically significant decrease in the depth of crow’s-feet wrinkles (43.3%) and in pore size (58.5%) was observed. Also, dermatological scores showed a significant decrease, and data suggest that skin tone was improved with a brighter tone. The injected mixture could then have an action on the skin pigmentation process with a photoprotective effect. Evolution of Dermis density was also investigated and an increase of dermal density and thickness were highlighted, showing an improvement of skin firmness with significant increases of dermis thickness (20%) and dermis density (24%), indicating a neocollagen activity.

Introduction: Skin aging is the result of many cellular dysfunctions over time particularly the fibroblasts and the keratinocytes. These dysfunctions could be decelerated by the preventive effects of some skin treatments such as Intradermal injections. NCTF 135HA has a polycomponent formulation designed to improve the efficacy of non-cross linked hyaluronic acid (as a micro-filler) on fibroblasts function. Although NCTF 135HA has been used by aesthetic practitioners since 20 years, we have analyzed all in vitro, ex vivo and in vivo studies during past 10 years in order to summarized its anti-aging effect. Methods: In these series of studies, the known effects of HA have been evaluated with this boosted formula. Collagen I synthesis, antiglycation effect and contractile forces developed by fibroblasts were studied. Fibroblasts and keratinocytes proliferations were evaluated in monolayer cells culture. The filling effect thanks to NCTF 135HA injection was ex vivo performed by fringe projection and the protective effect of NCTF 135HA against solar irradiation was performed ex vivo to study proliferation rate, elastin and collagen expression. Two clinical trials were performed on women to evaluate the anti-aging effect of NCTF 135HA injection. Hydration, firmness, radiance, wrinkles size, pore size, thickness and density of the dermis were analyzed. Results: Collagen I, anti-AGE products, cell proliferation and contractile forces were significantly increased with NCTF 135HA in culture medium. Ex vivo studies showed that NCTF 135HA increased, cells proliferation rate, elastin and collagen production. In addition, the filling effect of the NCTF 135HA is significantly superior to placebo. The first clinical trial shows that NCTF 135HA injection significantly increased tonicity, hydration and radiance of the skin and significantly decreased wrinkle score. 目的探究铜绿假单胞菌(PA)感染对肺血管内皮细胞功能的影响,并探讨其加重小鼠肺部炎症反应的可能机制。方法PAO1菌株感染人肺微血管内皮细胞HULEC-5a 2 h后,采用逆转录实时荧光定量PCR(RT-qPCR)检测自噬相关基因5(ATG5)、6-磷酸果糖-2-激酶/果糖-2,6-二磷酸酶3(PFKFB3)和钙黏蛋白5(CDH5)的mRNA表达,免疫荧光检测ATG5、PFKFB3和血管内皮钙黏蛋白(VE-cadherin)的蛋白表达。采用小干扰RNA(siRNA)分别敲减ATG5、PFKFB3后,RT-qPCR检测ATG5、PFKFB3和CDH5的mRNA表达,免疫荧光检测PFKFB3和VE-cadherin的蛋白表达,并采用乳酸测定试剂盒检测细胞内乳酸的水平。采用PAO1菌株感染小鼠后,肺组织切片病理染色观察小鼠肺部炎症,激光共聚焦显微镜观察并分析荧光标记的内皮细胞PFKFB3和VE-cadherin的表达量。结果与对照组比较,PAO1感染HULEC-5a细胞后,PFKFB3 mRNA和蛋白表达均增加(P均<0.05),CDH5 mRNA表达减少(P=0.023),VE-cadherin蛋白表达减少(P<0.001),且连续性被破坏,乳酸含量升高(P=0.017)。与PAO1感染HULEC-5a细胞比较,PAO1感染敲减PFKFB3的HULEC-5a细胞后,CDH5 mRNA表达增加(P=0.043),VE-cadherin荧光连续性得到恢复,乳酸含量降低(P=0.047);PAO1感染敲减ATG5的HULEC-5a细胞后,PFKFB3 mRNA(P=0.013)和蛋白表达(P=0.003)均增加,CDH5 mRNA(P=0.020)和VE-cadherin蛋白表达(P=0.001)均减少,乳酸含量升高(P=0.015)。PAO1感染小鼠后,病理HE染色结果显示,肺泡内明显的红细胞渗漏,炎症细胞浸润,肺泡间隔增宽和部分血管内皮细胞脱落。免疫荧光染色结果显示,与正常小鼠比较,PAO1感染小鼠内皮细胞PFKFB3表达增加,VE-cadherin荧光减弱,且不连续。结论PA可能通过AGT5调控PFKFB3通路,破坏肺血管内皮细胞功能,进而加重小鼠肺部炎症反应。

The strong societal demand to reduce pesticide use and adaptation to climate change challenges the capacities of phenotyping new varieties in the ***-throughput phenotyping is a way to obtain meaningful and reliable information on hundreds of genotypes in a limited *** evaluated traits related to growth in 209 genotypes from an interspecific grapevine biparental cross,between IJ119,a local genitor,and Divona,both in summer and in winter,using several methods:fresh pruning wood weight,exposed leaf area calculated from digital images,leaf chlorophyll concentration,and LiDAR-derived apparent *** high-density genetic information obtained by the genotyping by sequencing technology(GBS),we detected 6 regions of the grapevine genome[quantitative trait loci(QTL)]associated with the variations of the traits in the *** detection of statistically significant QTLs,as well as correlations(R^(2))with traditional methods above 0.46,shows that LiDAR technology is effective in characterizing the growth features of the *** calculated with LiDAR-derived total canopy and pruning wood volumes were high,above 0.66,and stable between growing *** variables provided genetic models explaining up to 47%of the phenotypic variance,which were better than models obtained with the exposed leaf area estimated from images and the destructive pruning weight *** results highlight the relevance of LiDAR-derived traits for characterizing genetically induced differences in grapevine growth and open new perspectives for high-throughput phenotyping of grapevines in the vineyard.

Background Malaria continues to cause burden in various parts of the ***,a Caribbean country,is among those aiming to eliminate malaria within a few *** surveys were conducted in Haiti during which we aimed to evaluate the performance of the simple and rapid procedure for ultra-rapid extraction-loop-mediated isothermal amplifcation(PURE-LAMP)method with dried blood spots as an alternative diagnostic method for malaria in the context of low to very low rates of *** Febrile and afebrile people were recruited from three administrative divisions within Haiti:Nippes,Sud and grand’Anse,during the summers of 2017(early August to early September)and 2018(late July to late August).Their blood samples were tested by microscopy,rapid diagnostic tests(RDT),PURE-LAMP and nested PCR to detect Plasmodium ***,specifcity,positive and negative predictive values and kappa statistics were estimated with the nested PCR results as the gold *** Among 1074 samples analyzed,a positive rate of 8.3%was calculated based on the nested PCR *** febrile participants,the rates in 2017 and 2018 were 14.6%and 1.4%,*** positives were detected among 172 afebrile participants in 2018 by PURE-LAMP and nested PCR,and all three were from the same *** was no afebrile participants recruited in *** PURE-LAMP,RDT and microscopy had respective sensitivities of 100%,85.4%and 49.4%.All of the testing methods had specifcities over 99%.Conclusions This study confrmed the high performance of the PURE-LAMP method to detect Plasmodium infection with dried blood spots and recommends its use in targeted mass screening and treatment activities in low endemic areas of malaria.
