Therapeutic advances in neural regeneration for Huntington’s disease

Huntington’s disease is a neurodegenerative disease caused by the expansion mutation of a cytosine-adenine-guanine triplet in the exon 1 of the HTT gene which is responsible for the production of the huntingtin (Htt) protein. In physiological conditions, Htt is involved in many cellular processes such as cell signaling, transcriptional regulation, energy metabolism regulation, DNA maintenance, axonal trafficking, and antiapoptotic activity. When the genetic alteration is present, the production of a mutant version of Htt (mHtt) occurs, which is characterized by a plethora of pathogenic activities that, finally, lead to cell death. Among all the cells in which mHtt exerts its dangerous activity, the GABAergic Medium Spiny Neurons seem to be the most affected by the mHtt-induced excitotoxicity both in the cortex and in the striatum. However, as the neurodegeneration proceeds ahead the neuronal loss grows also in other brain areas such as the cerebellum, hypothalamus, thalamus, subthalamic nucleus, globus pallidus, and substantia nigra, determining the variety of symptoms that characterize Huntington’s disease. From a clinical point of view, Huntington’s disease is characterized by a wide spectrum of symptoms spanning from motor impairment to cognitive disorders and dementia. Huntington’s disease shows a prevalence of around 3.92 cases every 100,000 worldwide and an incidence of 0.48 new cases every 100,000/year. To date, there is no available cure for Huntington’s disease. Several treatments have been developed so far, aiming to reduce the severity of one or more symptoms to slow down the inexorable decline caused by the disease. In this context, the search for reliable strategies to target the different aspects of Huntington’s disease become of the utmost interest. Non-alcoholic fatty liver disease (NAFLD), a further expression of metabolic syndrome, strictly linked to obesity and diabetes mellitus, is characterized by insulin resistance (IR), elevated serum levels of free fatty acids and fatty infi ltration of the liver, which is known as hepatic steatosis. Hepatocyte apoptosis is a key feature of this disease and correlates with its severity. Free-fatty-acidinduced toxicity represents one of mechanisms for the pathogenesis of NAFLD and hormones, growth factors and adipokines influence also play a key role. This review highlights the various pathways that contribute to the development of hepatic steatosis. Circulating concentrations of inflammatory cytokines are reckoned to be the most important factor in causing and maintaining IR. Low-grade chronic inflammation is fundamental in the progression of NAFLD toward higher risk cirrhotic states.

Fine-scale analysis of heavily invaded Italian freshwater fish assemblages

Inland waters are highly vulnerable to the introduction and spread of non-native species,due to heavy human use of aquatic ecosystems and the natural linkages among streams and *** is particularly noticeable in freshwater fish *** better evaluate how these communities are affected by non-native species introductions,we conducted a fine-scale analysis of the changes in Italian freshwater fish assemblages after species *** this analysis,we collected information on fish species present in 44 *** present Italian freshwater fish fauna is composed of 48 native and 41 established introduced species,while a further 15 introduced species have been reported but are not yet considered *** changes in the fish assemblages mostly took place in the past 2 centuries and have increased recently,with nearly 60%of the species introduced in the past 3 *** number of species introduced per basin ranged from 0 to 35(mean 10.85±7.77 species/basin),and in 10 basins the number of species introduced is now equal to or even higher than the number of native *** the past,introduced species mainly originated from America,but over the past three decades,an increase of introductions from other parts of Europe and Asia has been *** results show that basins already rich in native species tend to become even richer as a consequence of the establishment of introduced *** confirms the trend toward a biotic homogenization of ecosystems even at a local scale,due to an increase in the human-mediated spread of generalist species.

Phenotypic divergences induced by different residence time in invasive common ragweeds

EFFECTS OF RIVER NETWORK WORKS AND SOIL CONSERVATION MEASURES ON RESERVOIR SILTATION

Knowledge of the morphological dynamics of a water course is essential for management of reservoir siltation. With an example of sedimentation in a reservoir in Basilicata, Italy, this paper demonstrates the effect on reservoir siltation of the hydraulic works, which are aimed to reduce sediment transport along the fluvial network and to prevent part of the sediment discharge from reaching the lake. The effect depends on the river type and on the the geological features of river basin slopes. The paper also shows how mass erosion can significantly contribute to development of reservoir siltation. Finally, preliminary results are provided about the time needed for river training works to be effective.

Economic and Qualitative Traits of Italian Alps Saffron

Saffron, obtained from the flower stigmas of Crocus sativus L., is one of the most expensive food spices. The introduction of saffron in alpine areas could help to broaden and diversify the activities of mountain multifunctional farms, with a positive impact on economy and land management. According to ISO 3632(2010/2011), saffron can be classified into three categories of quality(I, II, III) depending on the concentration of the three main metabolites responsible for its characteristic colour, flavor and aroma: Crocin, Picrocrocin and Safranal. This study represents the first investigation of the quality of saffron produced in the Italian Alps evaluated with spectrophotometry, HPLC, solid-phase microextraction(SPME), and gas chromatographic analysis combined with mass spectrometry(GC/MS). The experiments used Crocus sativus stigmas produced in 2012-2013 in different areas of the Central Italian Alps were located at an altitude between 720 and 1200 m a.s.l.. Results obtained were compared to commercial saffron. The analyses confirmed that all samples can be classified in the first quality category according to the ISO classification. This high quality is also confirmed by HPLC analysis. Moreover, the SPME-GC/MS analysis identified some differences in the aromatic profile of saffron samples, in particular regarding safranal concentration. A preliminary assessment of the economic viability of high quality saffron production for local markets was also performed. Our study provides valid information regarding the quality and economic sustainability of saffron production in the alpine area confirming this crop as a good candidate for a new source of income for multifunctional farms in mountain areas.
