The tumor suppressor p53 is a transcription factor with a powerful antitumor activity that is controlled by its negative regulator murine double minute 2(MDM2,also termed HDM2 in humans)through a feedback *** the same time,TP53 is the most frequently mutated gene in human *** p53 proteins lose wild-type p53 tumor suppression functions but acquire new oncogenic properties,among which are deregulating cell proliferation,increasing chemoresistance,disrupting tissue architecture,and promoting migration,invasion and metastasis as well as several other pro-oncogenic *** oncogenic p53 mutation Y220C creates an extended surface crevice in the DNA-binding domain destabilizing p53 and causing its denaturation and *** cavity accommodates stabilizing small molecules that have therapeutic *** development of suitable small-molecule stabilizers is one of the therapeutic strategies for reactivating the Y220C mutant *** this review,we summarize approaches that target p53-Y220C,including reactivating this mutation with small molecules that bind Y220C to the hydrophobic pocket and developing immunotherapies as the goal for the near future,which target tumor cells that express the p53-Y220C neoantigen.

Predicting cardiovascular events in out-of-hospital patients presenting with atypical chest pain and complete left bundle branch block: role of CTA and echocardiographic Global Longitudinal Strain

Background Out-of-hospital patients presenting with atypical chest pain and complete left bundle branch block(LBBB)have to be stratified for the presence of coronary artery disease and the risk of developing heart failure(HF).We investigated the pro-gnostic role of coronary CT-angiography(CTA)and echocardiographic global longitudinal strain(GLS)in those patients in a mid-term *** Out-of-hospital patients with LBBB underwent echocardiography and a 64-slice CT angiography were evaluated *** of HF or a cardiovascular death were the events *** Seventy-eight patients(32 female;mean age:66.0±10.4 years were *** a follow-up of 33 months(IQr:17-77),one patient(1.5%)experienced a cardiovascular death,14 patients(17.9%)required urgent outpatient visits due to acute de-compensated HF(12 hospitalizations).Echocardiography showed a slightly reduced left ventricular ejection fraction(LVEF)(50.0%±9.8%)and GLS within the normal range(-16.2%±4.1%).CTA analysis showed coronary stenosis>50%in 28 patients(35.9%).A high Agatston score(>100)was observed in 29.5%.Notably,25 patients(32.1%)were diagnosed with left main coron-ary artery disease and 15 patients(16.7%)underwent revascularization during the follow *** associations were ob-served between events and LVEF(P=0.001),diastolic dysfunction grade≥2(P=0.02),GLS(Pronary stenos-is(P=0.04)and Agatston score(P=0.05).Multivariate analysis confirmed the relationships with LVEF(r2=0.89,Pr2=3.30,P=0.04),GLS(r2=1.43,Pre(r2=1.01,P=0.05).Conclusions In patients with complete LBBB,CTA and GLS identified those at a high risk of development HF.

recently, H_(2) has attracted increasing attention as green energy carrier holding the possibility to replace fossil fuel-based energy sources and thereby reduce CO_(2) emissions. Green hydrogen can be generated by water electrolysis using renewable energies like wind and solar power. When it is combusted, only water forms as by-product. However, the efficiency of water electrolysis is hampered by the anodic oxygen evolution reaction(OEr) because of the slow kinetics which leads to a high overpotential. Therefore, many catalysts have been developed for OEr to facilitate the kinetics and reduce the overpotential. In addition to electrocatalytic activity, the stability of the catalysts is imperative for industrial application and has been intensively studied. In this review, we cover recent findings on the stability and deactivation mechanisms of OEr catalysts. We discuss the correlation between OEr activity and stability, methodologies and experimental techniques to study the stability and deactivation as well as the deactivation mechanisms, together with factors influencing stability. Furthermore, strategies for stabilizing and regenerating OEr catalysts as well as methods to predict stability are summarized. Finally, the review highlights emerging methodologies yet to be explored and future directions of stability studies and the design of highly stable OEr catalysts.

Dear Editor,Afferent synapses of cochlear inner hair cells(IHCs)employ a unique molecular machinery(see extended background in Supplementary Materials).Otoferlin is a key player in this machinery and its defects cause human auditory synaptopathy(Moser and Starr,2016).Otoferlin,a tail-anchored(Vogl et al.,2016)multi-C_(2)-domain protein(Fig.1Ai)specific to hair cells(roux et al.,2006),is a member of the ferlin protein family involved in membrane trafficking and repair that are of major disease relevance(Pangršičet al.,2012),also see Supplementary *** is distributed broadly within IHCs(Fig.2Ai-Aiii;Pangrsic et al.,2010;roux et al.,2006).

Mitochondrial disorders are phenotypically varied, with serious clinical repercussions. Among them, there is the deficiency of combined oxidative phosphorylation of type 20, which occurs due to a defect in the VArS2 gene. This article presents a case of a 2-year-old female with progressive myoclonic epilepsy and psychomotor regression, with refractoriness to multiple anticonvulsants. The diagnosis was only made after the examination was carried out. Therefore, this article highlights the aspects of this rare disease and the importance of the exome for the diagnosis of rare conditions.

Poly(phenylene oxide)/renewable polyamide 11 (PPO/PA11 20/80) blends toughened with glycidylmethacrylate grafted ethylene-n-octene copolymer (GEOC) were prepared in a co-rotating twin-screwextruder. The reaction between GEOC and PPO/PA11 blend was analyzed by gel content tests. Themorphology of PPO/PA11/GEOC blends was observed by scanning electron microscope. The SEMresults showed that PPO formed the continuous phase, though it is a minority component of blends. Withincreasing GEOC content from 5 to 15 wt% the morphology of the blends transformed from droplet-matrix toco-continuous structure, in which both PA11 and PPO phases are continuous. The blend with co-continuousmorphology had better mechanical properties than those with droplet-matrix morphology. The impactstrength of the PPO/PA11/GEOC blends was much higher compared to the one without GEOC as well asPA11 due to the compatibilizing effect, which was also proved by DSC analysis, rheological behavior (MFr,DMTA) and tensile properties.

BACKGrOUND Brucellosis is the most common zoonosis worldwide and is endemic in the Middle East,Africa,Asia,and Latin ***,it is uncommon in Central Europe,and periprosthetic infections caused by Brucella are therefore *** to the low prevalence and nonspecific clinical presentation of the disease,accurate diagnosis can be challenging;no gold standard currently exists for treating *** SUMMArY Here,we present a 68-year-old Afghan woman living in Austria with a periprosthetic knee infection caused by Brucella *** interval from total knee arthroplasty to septic loosening was five years.A profound medical history and examinations suggested that the patient had been suffering from unrecognized chronic osteoarticular brucellosis prior to total knee *** was successfully treated by two-stage revision surgery and combined antibiotic therapy over three *** Clinicians should consider brucellosis as a possible cause of chronic arthralgia and periprosthetic infection in patients originating from countries with a high brucellosis burden.
