Comparative effects of plant growth regulators on leaf and stem explants of Labisia pumila var. alata

Objective: Labisia pumila var. alata, commonly known as 'Kacip Fatimah' or 'Selusuh Fatimah' in Southeast Asia, is traditionally used by members of the Malay community because of its post-partum medicinal properties. Its various pharmaceutical applications cause an excessive harvesting and lead to serious shortage in natural habitat. Thus, this in vitro propagation study investigated the effects of different plant growth regulators (PGRs) on in vitro leaf and stem explants of L. pumila. Methods: The capabilities of callus, shoot, and root formation were evaluated by culturing both explants on Murashige and Skoog (MS) medium supplemented with various PGRs at the concentrations of 0, 1, 3, 5, and 7 mg/L. Results: Medium supplemented with 3 mg/L indole-3-butyric acid (IBA) showed the optimal callogenesis from both leaf and stem explants with (72.34±19.55)% and (70.40±14.14)% efficacy, respectively. IBA was also found to be the most efficient PGR for root induction. A total of (50.00±7.07)% and (77.78±16.47)% of root formation were obtained from the in vitro stem and leaf explants after being cultured for (26.5±5.0) and (30.0±8.5) d in the medium supplemented with 1 and 3 mg/L of IBA, respectively. Shoot formation was only observed in stem explant, with the maximum percentage of formation ((100.00±0.00)%) that was obtained in 1 mg/L zeatin after (11.0±2.8) d of culture. Conclusions: Callus, roots, and shoots can be induced from in vitro leaf and stem explants of L. pumila through the manipulation of types and concentrations of PGRs.

Anti-neuroinflammatory responses in lipopolysaccharide-stimulated BV2 microglial cells upon treatment with methanol extract of Panax ginseng root

OBJECTIVE:To investigate the anti-neuroinflammatory properties of Panax ginseng(***)root by measuring the levels of nitric oxide(NO),tumour necrosis factor-alpha(TNF-α),interleukin-6(IL-6),and interleukin-10(IL-10)in lipopolysaccharide(LPS)-stimulated BV2 microglia ***:Maximal non-toxic dose(MNTD)of methanol extract of *** root culture on BV2 microglia cells was first determined via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay,followed by treatment and LPS stimulation of cells,and the measurement of NO using Griess assay and TNF-α,IL-6,and IL-10 using ELISA ***:The MNTD of *** root extract was determined to be(587±57)µg/*** that,NO and IL-6 levels were found to be insignificantly reduced by 6.88%and 0.14%respectively in stimulated cells upon treatment with *** with½MNTD yielded similar insignificant result,with only a reduction of 3.58%and 0.08%in NO and IL-6 levels ***,TNF-αand IL-10 levels were significantly downregulated by 15.64%and 34.96%respectively upon treatment with *** root extract at ***:Methanol extract of *** root culture did not show any significant anti-inflammatory effects on NO and IL-6 levels,but might potentially possess both anti-neuroinflammatory and pro-neuroinflammatory properties through the downregulation of TNF-αand IL-10 respectively.

Morphological and biochemical responses of Oryza sativa L.(cultivar MR219) to ion beam irradiation

Objective: Heavy ion beam, which has emerged as a new mutagen in the mutation breeding of crops and ornamental plants, is expected to result in the induction of novel mutations. This study investigates the morphological and biochemical responses of Oryza sativa toward different doses of carbon ion beam irradiation. Methods: In this study, the dry seeds of O. sativa were irradiated at 0, 20, 40, 60, 80, 100, and 120 Gy, followed by in-vitro germination under controlled conditions. Morphological and biochemical studies were conducted to investigate the morphological and physiological responses of O. sativa towards ion beam irradiation. Results: The study demonstrated that low doses(10 Gy) of ion beam have a stimulating effect on the height, root length, and fresh weight of the plantlets but not on the number of leaves. Meanwhile, doses higher than 10 Gy caused reductions in all the morphological parameters studied as compared to the control samples. The highest total soluble protein content [(2.11±0.47) mg/g FW] was observed in plantlets irradiated at 20 Gy. All irradiated plantlets were found to have 0.85% to 58.32% higher specific activity of peroxidase as compared to the control samples. The present study also revealed that low doses of ion beam(10 and 20 Gy) had negligible effect on the total chlorophyll content of O. sativa plantlets while 40 Gy had a stimulating effect on the chlorophyll content. Plantlets irradiated between 40 to 120 Gy were shown to be 0.38% to 9.98% higher in total soluble nitrogen content which, however, was not significantly different from the control samples. Conclusions: Carbon ion beam irradiation administered at low to moderate doses of 10 to 40 Gy may induce O. sativa mutants with superior characteristics.

Anti-adipogenic effects of extracts of Ficus deitoidea var. deltoidea and var. angustifolia on 3T3-L1 adipocytes

Objective： This study examined the anti-adipogenic effects of extracts of Ficus deltoidea var. deltoidia and var. angustifolia, a natural slimming aid, on 3T3-L1 adipocytes. Methods： Methanol and water extracts of leaves of the F. deltoidea varieties were analyzed to determine their total flavonoid content （TFC） and total phenolic content （TPC）, respectively. The study was initiated by determining the maximum non-toxic dose （MNTD） of the methanol and water extracts for 3T3-L1 preadipocytes. Possible anti-adipogenic effects were then examined by treating 2-d post confluent 3T3-L1 preadipocytes with either methanol extract or water extract at MNTD and half MNTD （1/2MNTD）, after which the preadipocytces were induced to form mature adipocytes. Visualisation and quantification of lipid content in mature adipocytes were carried out through oil red O staining and measurement of optical density （OD） at 520 nm, respec- tively. Results： The TFCs of the methanol extracts were 1.36 and 1.97 g quercetin equivalents （QE）/100 g dry weight （DW）, while the TPCs of the water extracts were 5.61 and 2.73 g gallic acid equivalents （GAE）/100 g DW for var. deltoidea and var. angustilofia, respectively. The MNTDs determined for methanol and water extracts were （300.0±28.3） and （225.0±21.2） μg/ml, respectively, for var. deltoidea, while much lower MNTDs [（60.0±2.0） IJg/ml for methanol extracts and （8.0＋1.0） μg/ml for water extracts] were recorded for var. angustifolia. Studies revealed that the methanol extracts of both varieties and the water extracts of var. angustifolia at either MNTD or 1/2MNTD significantly inhibited the maturation of preadipocytes. Conclusions： The inhibition of the formation of mature adipocytes indicated that leaf extracts of F. deltoidea could have potential anti-obesity effects.

Background and aim:Conventional hepatitis C treatment using pegylated interferon(PEG-IFN)and ribavirin is associated with significant side ***28B polymorphism can predict response to treatment,with CC genotype having a better *** gene deficiency protects against clinically significant anaemia induced by *** purpose of this study was to determine IL28B polymorphismand ITPA variation among hepatitis C genotype 1 patients who have undergone therapy with PEG-IFN and ribavirin and their association with sustained viral response(SVR).Methods:All hepatitis C genotype 1 patients who had been treated with PEG-IFN and ribavirin over the past 10 years were identified by available medical records and were contacted by letter of invitation to participate in the *** samples for IL28B and ITPA genotyping were *** records were reviewed for verification of treatment response,development of anaemia and if treatment reduction was required during the ***:A total of 61 patients with hepatitis C genotype 1 were treated with PEG-IFN and ribavirin,of whom 42 agreed to participate in the *** age was 45.6±12.9 years at time of treatment,and 83.3%of patients ***-three(78.6%)had IL28B CC genotype,of whom 25(75.8%)obtained SVR compared with only 3 of 9(33.3%)non C/C genotype patients who achieved SVR(P=0.041).Eleven(26.1%)patients had ITPA AC genotype,and 30(71.4%)had CC *** was no statistically significant difference between ITPA AC and CC genotypes in predicting clinically significant anaemia(45.5%vs 63.3%,P=0.302).Even among patients who developed anaemia,70.8%stillmanaged to achieve *** reduction also had no impact on ***:Hepatitis C genotype 1 patients should be informed of the response rate for treatment with PEG-IFN and ribavirin in a population with favourable IL28B genotype before consideration of newer therapeutic options.

Hydroxychavicol,a polyphenol from Piper betle leaf extract,induces cell cycle arrest and apoptosis in TP53-resistant HT-29 colon cancer cells

This study aims to elucidate the antiproliferative mechanism of hydroxychavicol(HC).Its effects on cell cycle,apoptosis,and the expression of c-Jun N-terminal kinase(JNK)and P38 mitogen-activated protein kinase(MAPK)in HT-29 colon cancer cells were *** was isolated from Piper betle leaf(PBL)and verified by high-performance liquid chromatography(HPLC),nuclear magnetic resonance(NMR),and gas chromatography-mass spectrometry(GC-MS).The cytotoxic effects of the standard drug 5-fluorouracil(5-FU),PBL water extract,and HC on HT-29 cells were measured after 24,48,and 72 h of *** cycle and apoptosis modulation by 5-FU and HC treatments were investigated up to 30 *** in phosphorylated JNK(pJNK)and P38(pP38)MAPK expression were observed up to 18 *** half maximal inhibitory concentration(IC_(50))values of HC(30μg/mL)and PBL water extract(380μg/mL)were achieved at 24 h,whereas the IC_(50)of 5-FU(50μmol/L)was obtained at 72 *** cycle arrest at the G0/G1 phase in HC-treated cells was observed from12 h *** apoptotic cell death in HC-treated cells compared to 5-FU-treated cells(P<0.05)was *** expression of pJNK and pP38 MAPK was observed at 12 h in HC-treated cells,but not in 5-FU-treated HT-29 cells(P<0.05).It is concluded that HC induces cell cycle arrest and apoptosis of HT-29 cells,with these actions possibly mediated by JNK and P38 MAPK.
