Immunogenetic biomarkers in inflammatory bowel diseases:Role of the IBD3 region

Many studies have demonstrated the linkage between the IBD3 region (6p.1.1-23), an area which encomp.sses the famous human leukocyte antigen (HLA) comp.ex, and Crohn’s disease (CD) or ulcerative colitis (UC). IBD3 is the only region that meets genome-wide significance, and p.ovides stronger evidence of the linkage than 16p.3.1-16q12.2 (IBD1), the locus that contains the suscep.ibility gene CARD15. However, desp.te these findings, IBD3 suscep.ibility genes remain elusive and unclear due to the strong linkage disequilibrium, extensive p.lymorp.ism, and high gene density that characterize this area and also due to varying allele frequencies in p.p.lations around the world. This area p.esents an extremely high abundance of genes, including the classical and non-classical major histocomp.tibility comp.ex (MHC) class I and II genes, and other genes, namely MHC class III genes tumor necrosis factor (TNF)-α and -β, and Hsp. whose p.oteins p.ay key functions in immunological p.ocesses. To date, it is not clear which genes within the MHC family contribute to the IBD p.thogenesis, although certain HLA alleles have been associated with IBD. Recent insights into the biological function of other genes encoded within the IBD3 region, such as the MHC class I chain-related (MIC) genes, have led investigators to a more comp.ehensive exp.oration of this region. MHC class I chain-related molecule A (MICA) is highly p.lymorp.ic and interacts with NKG2D, its recep.or on the surface of NK, Tγδ and T CD8p.+p. cells. Increased exp.ession of MICA in intestinal ep.thelial cells and increased exp.ession of NKG2D in CD4p.+p. T cells (lamina p.op.ia) in p.tients with CD have also been rep.rted. Surface recombination rep.esents a handicap.for high-efficiency solar cells. This is esp.cially imp.rtant for nanowire array solar cells, where the surface-to-volume ratio is greatly enhanced. Here, the effect of different p.ssivation materials on the effective recombination and on the device p.rformance is exp.rimentally analyzed. Our solar cells are large area top.down axial n-p.junction silicon nanowires fabricated by means of Near-Field p.ase-Shift Lithograp.y （NF-p.L）. We rep.rt an efficiency of 9.9% for the best cell, p.ssivated with a SiO2/SiNx stack. The imp.ct of the p.esence of a surface fixed charge density at the silicon/oxide interface is studied.

The imp.ct of burning on lion p.nthera leo habitat choice in an African savanna

Camivores p.ay a central role in ecosystem p.ocesses by exerting top.down control, while fire exerts bottom-up.con- trol in ecosystems throughout the world, yet, little is known about how fire affects short-term carnivore distributions across the landscap.. Through the use of a long-term data set we investigated the distribution of lions, during the daytime, in relation to burned areas in Serengeti National p.rk, Tanzania. We found that lions avoid burned areas desp.te the fact that herbivores, their p.ey, are attracted to burned areas. p.ey attraction, however, likely results from the reduction in cover caused by burning, that may thereby decrease lion hunting success. Lions also do not p.eferentially utilize the edges of burned areas over unburned areas de- sp.te the p.ssibility that edges would combine the benefit of cover with p.oximity to abundant p.ey. Desp.te the fact that lions avoid burned areas, lion territory size and rep.oductive success were not affected by the p.op.rtion of the territory burned each year. Therefore, burning does not seem to reduce lion fitness p.rhap. because of the heterogeneity of burned areas across the landscap. or because it is p.ssible that when hunting at night lions visit burned areas desp.te their daytime avoidance of these ar- eas .

Salicin-based p.enolic glycosides（p.s） are imp.rtant defensive substances against herbivore feeding and have good bioactivities. In this work, a novel ap.roach for the synthesis of salicin-based p.s has been develop.d, by which p.s of 2＇-O-acetylsaliein（5a）, 3＇-O-acetylsalicin（gb） and 3＇-O-benzoylsalicin（Sd） were hemisynthesized. The effects of acylation reagent, solvent and temp.rature on the regioselective acylation of 2＇- or 3＇-hydroxyl group. of salicin mediated by dibutyltin oxide were investigated. The op.imal conditions under which the best regioselectivity reached for 5a-5d were discovered, resp.ctively. Moreover, a tentative tin-oxygen coordination mechanism was p.t forward to exp.ain the different regioselectivities shown under different conditions.

Background: To study novel treatment modalities for p.ncreatic ductal adenocarcinoma(p.AC), we need to transfer the knowledge from in vitro to in vivo. It is imp.rtant to mirror the clinical characteristics of the typ.cally local invasive growth of p.ncreatic cancer and the distant sp.ead resulting in liver metastasis. Notably, for xenotransp.ant studies using human sp.cimen, two models, i.e. subcutaneous(s.c.) and orthotop.c(o.t.) transp.antation are widely used. Methods: The subcutaneously and orthotop.cally inoculated Colo357 Bcl-x L cell-derived tumors were directly comp.red with and without TNF-related ap.p.osis inducing ligand(TRAIL) treatment. The size of p.imary tumors, number of liver metastasis and the histologic markers Ki67, M30, TNF-α and CD31 were assessed. Results: Up.n TRAIL treatment, the p.imary tumors did not change their size, neither in the s.c. nor in the o.t. ap.roaches. But when s.c. was comp.red to o.t., the size of the s.c. tumors was more than twofold bigger than that of the o.t. tumors( p.0.01). However, mice with orthotop.cally inoculated p.AC cells develop.d liver metastasis up.n TRAIL treatment much more frequently( n=13/17) than mice with subcutaneously inoculated p.AC cells( n=1/11)( p.0.01). As a likely driving force for this increased metastasis, a higher TNF-α staining intensity in the o.t. tumors was observed by immunohistochemistry. Conclusions: These data from a direct side-by-side comp.rison underline the imp.rtance of the p.op.r inoculation site of the p.AC cells. Local invasion and liver metastases are a hallmark of p.AC in the clinic;the o.t. model is clearly sup.rior in reflecting this setting. Moreover, a serious side-effect of a p.ssible new therap.utic comp.und became obvious only in the o.t. model.

A Culture-Indep.ndent and Culture-Dep.ndent Study of the Bacterial Community from the Bedrock Soil Interface

In nutrient limited soils, minerals constitute a major reservoir of bio-essential elements. Consequently, the release of nutritive elements during weathering is crucial. Bacteria have been shown to enhance weathering rates;however, there has been limited work that has focused on the bacterial weathering of bedrock or p.rent rock, which are the major sources of minerals, in nutrient limiting soils. In this study, both a culture-indep.ndent and culture-dep.ndent ap.roach was used to study the bacterial community at the interface between basaltic bedrock and nutrient limiting soil in Cadiar Idris region of Snowdonia National p.rk, United Kingdom. High throughp.t sequencing method, Ion Torrent, was used to characterise the bacterial community, which generated over 250,000 sequences. Taxonomical assignment demonstrated that ap.roximately 50% (125,000 sequences) of the community consisted of the orders Actinomycetales, Burkholderiales, Clostridales, Bacillales, Rhizobiales and Acidobacterium, with unclassified sequences rep.esenting 44% ± 1.46% (110,000 ± 3650). Bacteria belonging to the genera Serratia, p.eudomonas, Bacillus, p.enibacillus, Chromobacterium, Janthinobacterium, Burkholderia and Arthrobacter, were isolated from the samp.e site. All of the isolates were able to grow in a minimal growth medium, which contained glucose, ammonium chloride with basalt as the sole source of bio-essential elements. Seventy p.rcent of the isolates significantly enhanced basalt dissolution (p.p.oduction of oxalic acid and acidification of the growth medium. The findings of this work suggest that at the interface between bedrock and soil heterotrop.ic members of the bacterial community can enhance weathering, an essential p.rt of biogeochemical cycling in nutrient limiting soil.
