Rbfox1 controls alternative splicing of focal adhesion genes in cardiac muscle cells

Alternative splicing is one of the major cellular processes that determine the tissue-specific expression of protein ***,it remains challenging to identify physiologically relevant and tissue-selective proteins that are generated by alternative ***,we investigated the target spectrum of the splicing factor Rbfox1 in the cardiac muscle context in more *** using a combination of in silico target prediction and in-cell validation,we identified several focal adhesion proteins as alternative splicing targets of *** focused on the alternative splicing patterns of vinculin(metavinculin isoform)and paxillin(extended paxillin isoform)and identified both as potential Rbfox1 *** analyses suggested that both isoforms are promoted by Rbfox1 due to binding in the *** adhesions play an important role in the cardiac muscle context,since they mainly influence cell shape,cytoskeletal organization,and cell–matrix *** data confirmed that depletion of Rbfox1 changed cardiomyoblast morphology,cytoskeletal organization,and multinuclearity after differentiation,which might be due to changes in alternative splicing of focal adhesion ***,our results indicate that Rbfox1 promotes alternative splicing of focal adhesion genes in cardiac muscle cells,which might contribute to heart disease progression,where downregulation of Rbfox1 is frequently observed.

Laser powder bed fusion(LPBF)for the fabrication of dense components used for tooling applications,is highly *** stresses,which evolve in the additively manufactured part,are inherent to LPBF *** additional stress contribution in high-carbon steels arises from the austenite-to-martensite phase transformation,which may eventually lead to cracking or even *** an alternative to pre-heating the base plate,which is not striven by industry,lowering the martensite content which forms in the part,is essential for the fabrication of dense parts by LPBF of high-carbon tool steels which are then adapted to *** this study,a successful strategy demonstrates the processing of the Fe85Cr4Mo1V1W8C1(wt%)high-carbon steel by LPBF into dense parts(99.8%).The hierarchical microstructure consists of austenitic and martensitic grains separated by elemental segregations in which nanoscopic carbide particles form a network.A high density of microsegregation was observed at the molten pool boundary ultimately forming a *** LPBF-fabricated steel shows a yield strength,ultimate compressive stress,and total strain of 1210 MPa,3556 MPa,and 27.4%,*** mechanical and wear performance is rated against the industrially employed and highly wear-resistant 1.2379 tool steel taken as the *** its lower macro-hardness,the LPBF steel(58.6 HRC,0.0061 mm^(3) Nm^(-1))shows a higher wear resistance than the reference steel(62.6 HRC,0.0078 mm^(3) Nm^(-1)).This behavior results from the wear-induced formation of martensite in a microscale thick layer directly at the worn surface,as it was proven via high-energy X-ray diffraction mapping.

Methyltrioxorhenium (MTO) and its derivatives were utilized as catalysts for a broad variety of organic reactions after the discovery of an easy synthetic access way to this class of compounds. Among the numerous characterization techniques, 17O-NMR spectroscopy and vibrational spectroscopy were exemplary discussed. The immobilization of MTO and its derivatives is of significant current interest as is the synthesis of chiral derivatives.

Scanning probe microscopes (SPM) are limited in their speed of data acquisition by the mechanical stability of the scanner. Therefore many types of scanners have been developed to achieve a rigid setup while maintaining an acceptable image size. We have followed here a different path to accelerate data acquisition by improving the feedback loop to achieve the same SPM image quality in a shorter time. While the feedback loop in a scanning probe microscope typically starts to probe a new pixel starting from the previous position, we have reduced the total control time by using an improved starting point for the feedback loop at each pixel. By exploiting the information of the already scanned pixels a forecast for the new pixel is created. We have successfully used several simple methods for a prognosis in MATLAB simulations like one dimensional linear or cubic extrapolation and others. Only scanning tunnelling microscope data from real experiments were used to test the forecasts. A doubling of the speed was achieved in the most favourable cases.

Water reinjection into the formation is an indispensable operation in many energy engineering *** operation involves a complex hydromechanical(HM)coupling process and sometimes even causes unpredictable disasters,such as induced *** is acknowledged that the relative magnitude and direction of the principal stresses significantly influence the HM behaviors of rocks during ***,due to the limitations of current testing techniques,it is still difficult to comprehensively conduct laboratory injection tests under various stress conditions,such as in triaxial extension stress *** this end,a numerical study of HM changes in rocks during injection under different stress states is *** this model,the saturated rock is first loaded to the target stress state under drainage conditions,and then the stress state is maintained and water is injected from the top to simulate the formation injection *** attention is given to the difference in HM changes under triaxial compression and extension *** includes the differences in the pore pressure propagation,mean effective stress,volumetric strain,and stress-induced *** numerical results demonstrate that the differential stress will significantly affect the HM behaviors of rocks,but the degree of influence is different under the two triaxial stress *** HM changes caused by the triaxial compression stress states are generally greater than those of extension,but the differences decrease with increasing differential stress,indicating that the increase in the differential stress will weaken the impact of the stress state on the HM *** addition,the shear failure potential of fracture planes with various inclination angles is analyzed and summarized under different stress *** is recommended that engineers could design suitable injection schemes according to different tectonic stress fields versus fault occurrence to reduce the risk of
