Enteropathic spondyloarthropathy:A common genetic background with inflammatory bowel disease?

The association between spondyloarthropathy and in flammatory bowel disease(IBD) is largely established, although prevalence is variable because of different population selection and diagnostic methodologies.Most studies indicate that as many as 10%15% of cases of IBD are complicated by ankylosing spondylitis(AS) or other forms of spondylarthritis(SpA).Of note, ileal inflammation resembling IBD has been reported in up to two thirds of cases of SpA, and it has been suggested that the presence of ileitis is associated with the chronic ity of articular complications.Although this observation is of interest to unravel the pathophysiology of the disease, systematic screening of patients with SpA by ileocolonos copy is not indicated in the absence of gut symptoms, as only a small proportion of patients with subclinical gut inflammation will develop overt IBD over time.The existence of familial clustering of both IBD and AS, the coexistence of both conditions in a patient, the evidence of an increased risk ratio among f irstand seconddegree relatives of affected AS or IBD patients and f inally, the increased crossrisk ratios between AS and IBD, strongly suggest a shared genetic background.So far, however, IL23R is the only identified susceptibility gene shared by both IBD and AS.Although functional studies are still needed to better understand its pathogenic role, great ef fort is being spent therapeutically targeting this pathway that may prove effective for both disorders.

AIM: To investigate gene variants in a large Italian inflammatory bowel disease (IBD) cohort, and to analyze the correlation of sub-phenotypes (including age at diagnosis) and epistatic interaction with other IBD genes. METHODS: Total of 763 patients with Crohn's disease (CD, 189 diagnosed at age < 19 years), 843 with ulcerative colitis (UC, 179 diagnosed <19 years), 749 healthy controls, and 546 healthy parents (273 trios) were included in the study. The rs2241880 [autophagy-related 16-like 1 (ATG16L1)], rs11209026 and rs7517847 [interleukin 23 receptor (IL23R)], rs2066844, rs2066845, rs2066847 (CARD15), rs1050152 (OCTN1), and rs2631367 (OCTN2) gene variants were genotyped. RESULTS: The frequency of G allele of ATG16L1 SNP (Ala197Thr) was increased in patients with CD compared with controls (59% vs 54% respectively) (OR = 1.25, CI = 1.08-1.45, P = 0.003), but not in UC (55%). The frequency of A and G (minor) alleles of Arg381Gln, rs11209026 and rs7517847 variants of IL23R were reduced significantly in CD (4%, OR = 0.62, CI = 0.45-0.87, P = 0.005; 28%, OR = 0.64, CI = 0.55-0.75, P < 0.01), compared with controls (6% and 38%, respectively). The A allele (but not G) was also reduced signifi cantly in UC (4%, OR = 0.69, CI = 0.5-0.94, P = 0.019). No association was demonstrated with sub-phenotypes and interaction with CARD15 , and OCTN1/2 genes, although both gene variants were associated with pediatric-onset disease. CONCLUSION: The present study confirms the association of IL23R polymorphisms with IBD, and ATG16L1 with CD, in both adult- and pediatric-onset subsets in our study population.

The change in the distribution of organisms in freshwater ecosystems due to natural or manmade processes raises the question of the impact of alien species on local communities.Although most studies indicate a negative effect,the positive one is more difficult to discern,espe-cially in multispecies systems,including hosts and parasites.The purpose of the study was to check whether the presence of an alien host,Potamopyrgus antipodarum,reduces the intensity of Echinoparyphium aconiatum metacercariae in a native host,Radix spp.We additionally tested the impact of water temperature and the biomass of the alien host on the dilution effect.We experimentally studied(1)the lifespan of echinostome cercariae in different temperatures,(2)the infectivity of cercariae toward the alien host and native host,and(3)the impact of different biomass of the alien host on the intensity of metacercariae in the native host.We found that cercarial survival and infectivity were tem-perature dependent.However,cercarial survival decreased with increasing temperature,contrary to cercarial infectivity.Echinostome cercariae entered the renal cavity of both the native host and alien host,and successfully transformed into metacercariae.The number of metacercariae in the native host decreased with the increasing biomass of the alien host.Our results indicate that lymnaeids may benefit from the co-occur-rence with P antipodarum,as the presence of additional hosts of different origins may reduce the prevalence of parasites in native communities.However,the scale of the dilution effect depends not only on the increased spectrum of susceptible hosts but also on the other variables of the environment,including water temperature and host density.

Cyclicity of the Lower Buntsandstein in the eastern part of the Central European Basin:implications for Early Triassic palaeogeography and for geochronological calibration

Lower Buntsandstein small—scale cycles recognized in the Central European Basin(CEB) are readily used for astrochronological calibration of the Early Triassic time scales,although they are not well studied sedimentologically.Three borehole sections from the eastern part of the CEB,forming a south-north transect perpendicular to the basin axis,were studied in terms to better understand the depositional history of the Lower Buntsandstein developed as the Baltic Formation in the studied Polish part of the basin.Eleven sedimentary facies were recognized,based on lithological and sedimentological investigations of 655.7 m of drill cores from the Otyń IG 1,Gorzów Wielkopolski IG 1 and Kamień Pomorski IG 1 boreholes.The facies can be grouped according to a lithological criterion into facies of predominantly siliciclastic lithology(including 9facies types) and facies of predominantly carbonate lithology(including 2 facies types).Facies analysis allowed to distinguish 8 facies associations of:1) alluvial fan,2) playa,3) sandy-muddy coastal plain,4)embayment and distal delta,5) lagoon,6) sand bars or shoals,7) ooidal shoals or bars,and 8) offshore.Deepening-upward(DC) and shallowing-upward(SC) sedimentary cycles were recognized in parts of the Baltic Formation,with DCs dominating in the southernmost located Otyń IG 1,and SCs dominating in Gorz ow Wielkopolski IG 1,located in the axial part of the basin.Symmetrical cycles are very rare.The statistical significance of the sedimentary cycles is relatively low according to Markov-chain analysis conducted with the phpSedistat software of Stanova et al.(2009).Analysis of small—scale cycles performed on well logs of investigated boreholes and additional boreholes in the vicinity of Otyń IG 1,implementing the earlier study of Becker(2005),showed that the correlation of well-log cycles(GR cycles) and sedimentary cycles is not obvious.GR cycles can be roughly correlated with lithological carbonate—siliciclastic cycles(c-s cycles),reflecting alternations of facies groups of predomina

WNT/β-catenin signaling in hepatocellular carcinoma:The aberrant activation,pathogenic roles,and therapeutic opportunities

Hepatocellular carcinoma(HCC)is a liver cancer,highly heterogeneous both at the histopathological and molecular levels.It arises from hepatocytes as the result of the accumulation of numerous genomic alterations in various signaling pathways,including canonical WNT/β-catenin,AKT/mTOR,MAPK pathways as well as signaling associated with telomere maintenance,p53/cell cycle regulation,epigenetic modifiers,and oxidative stress.The role of WNT/β-catenin signaling in liver homeostasis and regeneration is well established,whereas in development and progression of HCC is extensively studied.Herein,we review recent advances in our understanding of how WNT/β-catenin signaling facilitates the HCC development,acquisition of stemness features,metastasis,and resistance to treatment.We outline genetic and epigenetic alterations that lead to activated WNT/β-catenin signaling in HCC.We discuss the pivotal roles of CTNNB1 mutations,aberrantly expressed non-coding RNAs and complexity of crosstalk between WNT/β-catenin signaling and other signaling pathways as challenging or advantageous aspects of therapy development and molecular stratification of HCC patients for treatment.

Biodegradability of bioplastics in different aquatic environments:A systematic review

Bioplastics were first introduced as environmentally friendly materials,with properties similar to those of conventional plastics.A bioplastic is defined as biodegradable if it can be decomposed into carbon dioxide under aerobic degradation,or methane and CO_(2)under anaerobic conditions,inorganic compounds,and new cellular biomass,by the action of naturally occurring microorganisms.This definition however does not provide any information on the environmental conditions,timescale and extent at which decomposition processes should occur.With regard to the aquatic environment,recognized standards have been established to assess the ability of plastics to undergo biodegradation;however,these standards fail to provide clear targets to be met to allow labelling of a bioplastic as biodegradable.Moreover,these standards grant the user an extensive leeway in the choice of process parameters.For these reasons,the comparison of results deriving from different studies is challenging.The authors analysed and discussed the degree of biodegradability of a series of biodegradable bioplastics in aquatic environments(both fresh and salt water)using the results obtained in the laboratory and from on-site testing in the context of different research studies.Biochemical Oxygen Demand(BOD),CO_(2)evolution,surface erosion and weight loss were the main parameters used by researchers to describe the percentage of biodegradation.The results showed a large variability both in weight loss and BOD,even when evaluating the same type of bioplastics.This confirms the need for a reference range of values to be established with regard to parameters applied in defining the biodegradability of bioplastics.

In a recent article published in Cancer Discovery,Guirguis et al.demonstrated that enzymatic inhibitors of the m6 A methyltransferase METTL3 induced a formation of immunostimulatory doublestranded mRNAs(dsRNAs)in cancer cells,which triggered a profound tumor-cell-intrinsic interferon response leading to an enhanced antigen presentation and thereby increased killing of tumor cells by cytotoxic CD8+T-lymphocytes.The inhibition of METTL3 synergized with anti-PD1 immunotherapy,highlighting its potential for improving cancer treatment.

Dear Editor,The problem of age estimation in amphibians and reptiles with annual fluctuations of growth pattern has been considered to be mostly solved since the skeletochronological method was introduced(Kleinenberg and Smirina,1969).This method is based on counting the number of lines of arrested growth(LAGs)—cyclical growth marks that are usually formed annually and characterized by different optical aspects within the tubular bones.

Polymerization of 1,3-Butadiene with Catalysts Based on Cobalt Dichloride Complexes with Aminophosphines: Switching the Regioselectivity by Varying the MAO/Co Molar Ratio

Several novel cobalt dichloride complexes with amino-phosphine bidentate ligands were synthesized and characterized.For some of them single crystals were obtained and their molecular structure was determined by X-ray diffraction method.All the complexes were then used in combination with MAO for the polymerization of 1,3-butadiene,obtaining polymers with different structures(i.e.,predominantly 1,2 or cis-1,4)mainly depending on the type of ligand and on the MAO/Co molar ratio.The behavior of these novel catalysts was compared with that exhibited,in the polymerization of the same monomer,by the systems CoCl_(2)(PR3)2-MAO and CoCl_(2)(PRPh2)2-MAO(R=alkyl or cycloalkyl group),and by the systems based on cobalt dichloride complexes with various bi-and tridentate ligands(e.g.,diphosphines,bis-imines,pyridyl-imines,bis-iminopyridines).The comparison between the different systems allowed us to make some clarity about the actual and effective role played by the various types of ligands in the polymerization of conjugated dienes with catalytic systems CoCl_(2)(L)-MAO,in which L=mono-,bi-,or tri-dentate ligand.
