Characterization of modified mesoporous silica nanoparticles as vectors for siRNA delivery

Gene therapy using siRNA molecules is nowadays considered as a promising approach. For successful therapy, development of a stable and reliable vector for siRNA is crucial. Non-viral and non-organic vectors like mesoporous silica nanoparticles(MSN) are associated with lack of most viral vector drawbacks, such as toxicity, immunogenicity, but also generally a low nucleic acid carrying capacity. To overcome this hurdle, we here modified the pore walls of MSNs with surface-hyperbranching polymerized poly(ethyleneimine)(hbPEI), which provides an abundance of amino-groups for loading of a larger amount of siRNA molecules via electrostatic adsorption. After loading, the particles were covered with a second layer of pre-polymerized PEI to provide better protection of siRNA inside the pores, more effective cellular uptake and endosomal escape. To test the transfection efficiency of PEI covered si RNA/MSNs, MdA-MB 231 breast cancer cells stably expressing GFP were used. We demonstrate that PEI-coated si RNA/MSN complexes provide more effective delivery of si RNAs compared to unmodified MSNs. Thus, it can be concluded that appropriately surface-modified MSNs can be considered as prospective vectors for therapeutic siRNA delivery.

1Introduction Hydrogen/air combustion is a problem both in respect of its fundamental science and in the use of hydrogen as a possible substitute for traditional hydrocarbon *** is now a generally agreed approach to the use of hydrogen as an addition to traditional fuels(He et al.,2017)but the use of hydrogen as a main fuel has been studied much less because of the substantially unresolved problems for effective hydrogen production(Liu et al.,2020).It is worth noting that the use of hydrogen as a main fuel may be primarily applied not only for internal combustion engines needed for transport(Balat,2008),but also for those serving as drives for electrogenerators within stationary power stations(Balat,2008;Ivanov et al.,2018).

Gastric cancer currently ranks fourth in cancer-related mortality worldwide. In the western world, it is most often diagnosed at an advanced stage, after becoming metastatic at distant sites. Patients with advanced disease(locally advanced or metastatic) have a somber prognosis, with a median overall survival of 10-12 mo, and palliative chemotherapy is the mainstay of treatment. In recent years, novel approaches using inhibition of human epidermal growth factor receptor 2(HER2) have demonstrated significant improvements in progression-free and overall survival, compared with chemotherapy alone, in first-line treatment of patients with overexpression of HER2. In addition, both second-line chemotherapy and treatment with the vascular endothelial growth factor receptor-inhibitor ramucirumab demonstrated significant benefits in terms of overall survival, compared with best supportive care, in randomized studies. Moreover, ramucirumab in combination with chemotherapy demonstrated further significant benefits in terms of progression-free and overall survival, compared with chemotherapy alone, in second-line treatment for patients with metastatic gastric cancer. A recently published molecular classification of gastric cancer is expected to improve patient stratification and selection for clinical trials and provide a roadmap for future drug development. Nevertheless, despite these developments the prognosis of patients with advanced gastric cancer remains poor. In this review we discuss current standards of care and outline major topics of drug development in gastric cancer.

An approximate solution of the d-dimensional Schrodinger equation with the modified Posehl-Teller potential is obtained with an approximation of the centrifugal term. Solution to the corresponding hyper-radial equation is given using the conventional Nikiforov-Uvarov method. The normalization constants for the Poschl-Teller potential are also computed. The expectation values of 〈r^-2 and 〈V（r）〉 are also obtained using the Feynman- Hellmann theorem.

The experimental research, presented in the study, focuses on track tests with the aim of highlighting changes in lap times after manipulative treatment of drainage of the glymphatic system and stimulation of the sympathetic nervous system. Introduction: The experimental research, presented in this study, focuses on analyzing the potential effects of a manipulative treatment on the performance of a professional driver. The main objective is to evaluate the change in lap times after the application of the treatment, trying to understand whether it can actually positively influence the driver’s performance. The study stands an important opportunity to extend knowledge, regarding the use of manipulative therapies in the context of optimized driving skills. The results obtained could provide useful insights and contribute to improving the performance of professional drivers by offering new perspectives and strategies to improve their performance. Leveraging a rigorous scientific approach and a sample of highly skilled drivers, the research aims to provide concrete evidence on the effectiveness of manipulative treatment in driving skills. Monitoring lap times before and after the intervention also capture any temporary or long-term effects of the treatment, ensuring a thorough and reliable analysis of the results. Materials and methods: 15 professional drivers, aged 18 to 36 years, with at least 10 years of experience as drivers, participated in this study. The test consisted of analyzing lap times before and after treatment.

The development of the giant magnetostrictive rare earth-iron materials is reported from their discovery in the early 1970's to their present use in transducer and sensor applications. Fifteen broad market areas, ranging from miniature devices in the medical industry to high powder sonar and seismic sources have been identified. A world-wide customer base in these high technology fields has been established and the superior properties of these materials are responsible for the rapid growth of the magnetostrictive transducer industries. The critical design criteria that lead to efficient devices are reported and the performance of several of these products is presented. The key patents associated with these giant magnetostrictive materials and devices using these materials are reviewed. The present and future of these technologies in reference to commercialization, market growth and the impact on the many transduction industries are discussed. (Author abstract) 9 Refs.

Ovarian carcinoma is one of the most common causes for cancer death in women;lack of early diagnosis and acquired resistance to platinum-based chemotherapy account for its poor prognosis and high mortality *** with other cancer types,ovarian cancer is characterized by dysregulated signaling pathways and protein synthesis,which together contribute to rapid cellular growth and *** mechanistic/mammalian target of rapamycin(mTOR)pathway represents the core of different signaling pathways regulating a number of essential steps in the cell,among which protein synthesis and the eukaryotic initiation factor 4E(eIF4E),the mRNA cap binding protein,is one of its downstream ***4E is a limiting factor in translation initiation and its overexpression is a hallmark in many *** its action is regulated by a number of factors that compete for the same binding site,eIF4E is an ideal target for developing novel antineoplastic *** inhibitors targeting the mTOR signaling pathway have been designed thus far,however most of these molecules show poor stability and high toxicity in *** minireview explores the possibility of targeting mTOR and eIF4E proteins,thus impacting on translation initiation in ovarian cancer,describing the most promising experimental strategies and specific inhibitors that have been shown to have an effect on other kinds of cancers.
