Due to the functional importance of bovine milk protein polymorphisms, their correct discrimination is of great interest both from a scientific and practical point of view. Nowadays a large number of commercial platforms are available for semiautomated or fully automated SNP geno-typing. However, in some cases the use of simple and rather cheap methods is an effective tool to be implemented within one’s own laboratory for the routine analysis of a specific SNP. The present paper describes two simple tests based on the bidirectional allele-specific polymerase chain reaction (BAS-PCR) developed for the identification of β-casein (CSN2) B and I genetic variants. The practical application of the two methods on a panel of 84 Italian Brown bulls and 100 Italian Friesian cows is also discussed, including the biological significance of the two genetic variants and the importance of taking their occurrence into account when linkage analyses are performed on milk functional properties. A combined system for analysing milk protein variants by isoelectrofocusing (IEF) and the BAS-PCR assay developed for CSN2*I is described.

In the recent years, the number of endangered animals, both referred to livestock and wild species, has grown enormously. The “livestock” term refers to animals domesticated for producing commodities for man such as food, fiber and draught. Livestock biodiversity is integral to our culture, history, environment, and economy. Thousands of livestock breeds have evolved over time to suit particular environments and farming systems. Conservation and analyses of these genetic resources rely on demographic characterization and correct breeding schemes. In addition, molecular genetic studies allow to identify and monitor the genetic diversity within and across breeds and to reconstruct their evolution history. The conservation of livestock variability is also a crucial element in order to preserve and valorise specific nutritional and nutraceutical properties of animal products. Efficient ex situ and in situ conservation strategies, as well as the creation of bio-banks and specific biotechnological and bioinformatics tools for genetic analyses and digital preservation, are obligatory requirements in order to implement an appropriate action for the conservation of animal biodiversity. The main issues concerning different species are summarised, with particular reference to the livestock biodiversity still existing. Some examples of ex situ conservation strategies, which mainly refer to cryoconservation of semen, ova, embryos or tissues, developed in Italy, are presented, and the different actions in defense of Animal Genetic Resources (AnGR) developed within the European Community are illustrated. Interestingly, the same strategies for biological and digital analyses and preservation of livestock biodiversity can be exported to wild endangered animals in order to plan a correct conservation and repopulation of the species. The Canavese Zone(CZ)in the Western Alps represents the remnant of the distal passive margin of the Adria microplate,which was stretched and thinned during the Jurassic opening of the Alpine *** detailed geological mapping,stratigraphic and structural analyses,we document that the continental break-up of Pangea and tectonic dismemberment of the Adria distal margin,up to mantle rocks exhumation and oceanization,did not simply result from the syn-rift Jurassic extension but was strongly favored by older structu ral inheritances(the Proto-Canavese Shear Zone),which controlled earlier lithospheric *** findings allowed to redefine in detail(i)the tectono-stratigraphic setting of the Variscan metamorphic basement and the Late Carbonife rous to Early Cretaceous CZ succession,(ii)the role played by inherited Late Carboniferous to Early Triassic structures and(iii)the significance of the CZ in the geodynamic evolution of the Alpine *** large amount of extensional displacement and crustal thinning occurred during different pulses of Late Carbonife rous-Early Triassic strike-slip tectonics is wellconsistent with the role played by long-lived regional-scale wrench faults(e.g.,the East-Variscan Shear Zone),suggesting a re-discussion of models of mantle exhumation driven by low-angle detachment faults as unique efficient mechanism in stretching and thinning continental crust.

The Canavese Intracontinental Suture Zone(CISZ) within the Inner Western Alps represents the remnant of a long-lived minor subduction zone involving a narrow, thinned continental crust/oceanic lithosphere seaway between two continental domains of the Adria microplate(i.e., the Sesia Zone and the IvreaVerbano Zone). As opposed to many suture zones, the CISZ mostly escaped pervasive tectonic deformation and metamorphism, thus preserving the original stratigraphy and allowing the relationships between tectonics and sedimentation to be defined. Through detailed geological mapping(1:5000 scale),structural analysis, stratigraphic and petrographic observations, we document evidences for the late Paleozoic to late Cenozoic tectonic evolution of the CISZ, showing that it played a significant role in the context of the tectonic evolution of the Inner Western Alps region from the early to late Permian Pangea segmentation, to the Jurassic Tethyan rifting, and up to the subduction and collisional stages,forming the Western Alps. The site of localization/formation of the CISZ was not accidental but associated with the re-use of structures inherited from regional-scale wrench tectonics related to the segmentation of Pangea, and from the subsequent extensional tectonics related to the Mesozoic rifting, as documented by crosscutting relationships between stratigraphic unconformities and tectonic features. Our findings document that evidences derived from stratigraphy, facies indicators, and relationships between tectonics and sedimentation in the shallow crustal portions of suture zones, such in the CISZ, are important to better constrain the tectonic history of those metamorphic orogenic belts around the world in which evolutionary details are commonly complicated by high-strain deformation and metamorphic transformations.

Background: Innate immune responses induced by in vitro stimulation of primary mammary epithelial cells(MEC) using Gram-negative lipopolysaccharide(LPS) and Gram-positive lipoteichoic acid(LTA) bacterial cell wall components are well-characterized in bovine species. The objective of the current study was to characterize the downstream regulation of the inflammatory response induced by Toll-like receptors in primary goat MEC(pgMEC). We performed quantitative real-time RT-PCR(qPCR) to measure mRNA levels of 9 genes involved in transcriptional regulation or antibacterial activity: Toll-like receptor 2(TLR2), Toll-like receptor 4(TLR4), prostaglandin-endoperoxide synthase 2(PTGS2), interferon induced protein with tetratricopeptide repeats 3(IFIT3), interferon regulatory factor 3(IRF3), myeloid differentiation primary response 88(MYD88), nuclear factor of kappa light polypeptide gene enhancer in B-cells 1(NFKB1), Toll interacting protein(TOLLIP), and lactoferrin(LTF). Furthermore,we analyzed 7 cytokines involved in Toll-like receptor signaling pathways: C-C motif chemokine ligand 2(CCL2), C-C motif chemokine ligand 5(CCL5), C-X-C motif chemokine ligand 6(CXCL6), interleukin 8(CXCL8), interleukin 1 beta(IL1 B), interleukin 6(IL6), and tumor necrosis factor alpha(TNF).Results: Stimulation of pgMEC with LPS for 3 h led to an increase in expression of CCL2, CXCL6, IL6, CXCL8,PTGS2, IFIT3, MYD88, NFKB1, and TLR4(P < 0.05). Except for IL6, and PTGS2, the same genes had greater expression than controls at 6 h post-LPS(P < 0.05). Expression of CCL5, PTGS2, IFIT3, NFKB1, TLR4, and TOLLIP was greater than controls after 3 h of incubation with LTA(P < 0.05). Compared to controls, stimulation with LTA for 6 h led to greater expression of PTGS2, IFIT3, NFKB1, and TOLLIP(P < 0.05) whereas the expression of CXCL6, CXCL8, and TLR4 was lower(P < 0.05).
