Secondary sclerosing cholangitis in a young COVID-19 patient resulting in death:A case report

bACKGROUND With the emergence of severe acute respiratory syndrome coronavirus 2(SARSCoV-2) in late 2019 in Wuhan, China, liver injury in patients with coronavirus disease 2019(COVID-19) due to SARS-CoV-2 infection has been regularly reported in the literature. There are a growing number of publications describing the occurrence of secondary sclerosing cholangitis(SSC) after SARS-CoV-2 infection in various cases. We present a case of sudden onset SSC in a critically ill patient(SSC-CIP) following COVID-19 infection who was previously *** SUMMARY A 33-year old female patient was admitted to our University Hospital due to increasing shortness of breath. A prior rapid antigen test showed a positive result for SARS-CoV-2. The patient had no known preexisting conditions. With rapidly increasing severe hypoxemia she required endotracheal intubation and developed the need for veno-venous extracorporeal membrane oxygenation in a setting of acute respiratory distress syndrome. During the patient’s 154-d stay in the intensive care unit and other hospital wards she underwent hemodialysis and extended polypharmaceutical treatment. With increasing liver enzymes and the development of signs of cholangiopathy on magnetic resonance cholangiopancreatography(MRCP) as well as endoscopic retrograde cholangiopancreatography(ERCP), the clinical setting was suggestive of SSC. At an interdisciplinary meeting, the possibility of orthotopic liver transplantation and additional kidney transplantation was discussed due to the constant need for hemodialysis. Following a deterioration in her general health and impaired respiratory function with a reduced chance of successful surgery and rehabilitation, the plan for transplantation was discarded. How do neurons age?A focused review on the aging of the microtubular cytoskeleton

Aging is the leading risk factor for Alzheimer’s disease and other neurodegenerative diseases. We now understand that a breakdown in the neuronal cytoskeleton, mainly underpinned by protein modifications leading to the destabilization of microtubules, is central to the pathogenesis of Alzheimer’s disease. This is accompanied by morphological defects across the somatodendritic compartment, axon, and synapse. However, knowledge of what occurs to the microtubule cytoskeleton and morphology of the neuron during physiological aging is comparatively poor. Several recent studies have suggested that there is an age-related increase in the phosphorylation of the key microtubule stabilizing protein tau, a modification, which is known to destabilize the cytoskeleton in Alzheimer’s disease. This indicates that the cytoskeleton and potentially other neuronal structures reliant on the cytoskeleton become functionally compromised during normal physiological aging. The current literature shows age-related reductions in synaptic spine density and shifts in synaptic spine conformation which might explain age-related synaptic functional deficits. However, knowledge of what occurs to the microtubular and actin cytoskeleton, with increasing age is extremely limited. When considering the somatodendritic compartment, a regression in dendrites and loss of dendritic length and volume is reported whilst a reduction in soma volume/size is often seen. However, research into cytoskeletal change is limited to a handful of studies demonstrating reductions in and mislocalizations of microtubule-associated proteins with just one study directly exploring the integrity of the microtubules. In the axon, an increase in axonal diameter and age-related appearance of swellings is reported but like the dendrites, just one study investigates the microtubules directly with others reporting loss or mislocalization of microtubule-associated proteins. Towards an enhanced understanding of the particle size effect on conversion/alloying lithium-ion anodes

Conversion/alloying materials(CAMs)represent a potential alternative to graphite as a Li-ion anode active material,especially for high-power *** far,however,essentially all studies on CAMs have been dealing with nano-sized particles,leaving the question of how the performance(and the de-/lithiation mechanism in general)is affected by the particle ***,we comparatively investigate four different samples of Zn_(0.9)Co_(0.1)O with a particle size ranging from about 30 nm to a few *** results show that electrodes made of larger particles are more susceptible to fading due to particle displacement and particle *** results also show that the conversion-type reaction in particular is affected by an increasing particle size,becoming less reversible due to the formation of relatively large transition metal(TM)and alloying metal nanograins upon lithiation,thus hindering an efficient electron transport within the initial particle,while the alloying contribution remains essentially *** generality of these findings is confirmed by also investigating Sn_(0.9)Fe_(0.1)O_(2) as a second CAM with a substantially greater contribution of the alloying reaction and employing Fe instead of Co as a TM dopant.

Effects of P301L-TAU on post-translational modifications of microtubules in human iPSC-derived cortical neurons and TAU transgenic mice

Modelling smear effect of vertical drains using a diameter reduction method

Vertical drains are used to accelerate consolidation of clays in ground improvement *** zones exist around these drains,where permeability is reduced due to soil disturbance caused by the installation *** solution is widely used in practice to consider the effects of drain discharge capacity and smear on the consolidation *** this study,a computationally efficient diameter reduction method(DRM)obtained from the Hansbo solution is proposed to consider the smear effect without the need to model the smear zone *** by analytical and numerical results,a diameter reduction factor is analytically derived to reduce the diameter of the drain,while achieving similar solutions of pore pressure dissipation profile as the classical full model of the smear zone and *** the DRM,the excess pore pressure u obtained from the reduced drain in the original un-disturbed soil zone is accurate enough for practical applications in numerical *** performance of DRM is independent of soil material *** also show equally accurate performance of DRM under conditions of multi-layered soils and coupled radial-vertical groundwater flow.

tRNA^(Glu)-derived fragments from embryonic extracellular vesicles modulate bovine embryo hatching

Transfer RNA-derived small RNAs(tsRNAs)have been shown to be involved in early embryo development and repression of endogenous retroelements in embryos and stem ***,it is unknown whether tsRNAs also regulate embryo *** this study,we mined the sequencing data of a previous experiment in which we demonstrated that the microRNA(miRNA)cargo of preimplantation embryonic extracellular vesicles(EVs)influences embryo *** thus profiled the tsRNA cargo of EVs secreted by blastocysts and *** majority of tsRNAs was identified as tRNA halves originating from the 5'ends of *** the 148 differentially expressed tsRNAs,the 19 nt tRNA fragment(tRF)tDR-14:32-Glu-CTC-1 was found to be significantly up-regulated in EVs derived from ***-qPCR assays confirmed its significant up-regulation in non-blastocyst embryos and their conditioned medium compared to the blastocyst group(Pbition of tDR-14:32-Glu-CTC-1 by supplementing antagomirs to the conditioned medium improved embryo hatching(Pbryos treated with tDR-14:32-Glu-CTC-1 antagomirs further showed differential expression of genes that are associated with embryo hatching and *** summary,tDR-14:32-Glu-CTC-1 is up-regulated in non-blastocyst embryos and their secretions,and inhibition of tDR-14:32-Glu-CTC-1 promotes embryo hatching,while influencing embryo implantation-related genes and *** results indicate that embryonic EVs containing specific tRFs may regulate preimplantation embryo development.
