Drp1-dependent remodeling of mitochondrial morphology triggered by EBV-LmP1 increases cisplatin resistance

Immunoglobulins （Igs） are known to be synthesized and secreted only by B lymphocytes. Class switch recombination （CSR） is a key event that enables B cells to express Igs, and one of the crucial steps for CSR initiation is the germline transcription of Iggenes. Surprisingly, recent studies have demonstrated that the Iggenes are also expressed in some epithelial cancer cells; however, the mechanisms underlying how cancer cells initiate CSR and express Igs are still unknown. In this study, we confirmed that the Ig la I promoter in cancer cell lines was activated by the Ets- 1 transcription factor, and the activity of the Ig la I promoter and ig lal-C^l germiine transcription were attenuated after knockdown of Ets-1 by specific small interfering RNAs （siRNA）. Furthermore, the expression of Ets-1 and Iga heavy chain in cancer cells was dose dependently upregulated by TGF-I^I. These results indicate that activation of the Ig lal promoter by the transcriotion factor Ets-1 is a critical Dathwav and orovides a novel mechanism for le exoression in non-B cell cancers.

micropropagation protocols to minimize hyperhydricity were optimized for medicinal Scutellaria barbata and Scutellaria racemosa. Six cytokinins and eight different carbon sources at two different incubation periods of 14 and 21 days were studied for adventitious shoot bud induction using nodal explants. In S. barbata, 5 μm meta-Topolin and 0.1 μm NAA supplemented shoot induction medium produced four shoots each after 14 and 21 day incubation. Observation of S. racemosa nodal explants recorded four and five shoots after 14 and 21 day incubation. In both species, control explants (no plant growth regulators in the medium) consistently resulted in the bud break with two shoots in both 14 and 21 day incubation. The effect of carbon source on shoot regeneration was studied by supplementing eight different sugars at 0.1 m concentration to the optimized shoot induction medium (5 μm meta-Topolin and 0.1 μm NAA). S. barbata nodal explants cultured on shoot induction medium supplemented with fructose and glucose for 14 days produced 10 and nine adventitious shoots respectively;and after 21 day incubation adventitious shoot count reached 19 in glucose supplemented medium. S. racemosa explants in the same experiment produced five shoots in maltose and four shoots in sorbitol supplemented medium after 14 day incubation;whereas after 21 day incubation, sucrose and maltose produced five shoots;fructose, glucose, and sorbitol produced four shoots. Regenerated plants were successfully acclimatized and Scanning Electron microscopy of the leaf surface revealed differences in stomatal behavior and cuticle deposition between in vitro and acclimatized plants. The antioxidant assay conducted on both Scutellaria species showed considerable total polyphenol content, TEAC activity and flavonoid content in fresh and dried leaf samples attributing to their medicinal potential.

Background: Procalcitonin, a precursor protein of the hormone calcitonin, is a sensitive marker for sepsis in human medicine, which is used for diagnosis of bacterial pneumonia in adults and neonates to initiate antibiotic therapy. Objectives: In this study, procalcitonin was evaluated as a potential biomarker for Rhodococcus equi associated pneumonia. methods: In four foals procalcitonin was measured at four time-points (day 0 before antibiotic therapy, day 1, 3 and 5/6 during therapy) in plasma using an equine specific ELISA. Inclusion criteria for the study were a positive sepsis score, ultrasonographic evidence of pulmonary abscesses in addition >8 cm, a positive microbiology out of tracheobronchial secretion and positive response to antibiotic treatment (azithromycin 10 mg/kg BDW 24q PO and rifampicin 10 mg/kg BDW 12q PO) within a weak including improvement of clinical status and reduction of ultrasonographic score Rhodococcus equi.

The microstructural architecture of remodeled bone in the peri-implant region of screw implants plays a vital role in the distribution of strain energy and implant *** present a study in which screw implants made from titanium,polyetheretherketone and biodegradable magnesium-gadolinium alloys were implanted into rat tibia and subjected to a push-out test four,eight and twelve weeks after *** were 4 mm in length and with an m2 *** loading experiment was accompanied by simultaneous three-dimensional imaging using synchrotron-radiation microcomputed tomography at 5μm *** deformation and strains were tracked by applying optical flow-based digital volume correlation to the recorded image *** stabilities measured for screws of biodegradable alloys were comparable to pins whereas non-degradable biomaterials experienced additional mechanical ***-implant bone morphology and strain transfer from the loaded implant site depended heavily on the biomaterial *** implants stimulated rapid callus formation displaying a consistent monomodal strain profile whereas the bone volume fraction in the vicinity of magnesium-gadolinium alloys exhibited a minimum close to the interface of the implant and less ordered strain *** in our data suggest that implant stability benefits from disparate bone morphological properties depending on the biomaterial *** leaves the choice of biomaterial as situational depending on local tissue properties.

Polar transport of the phytohormone auxin and the establishment of localized auxin maxima regulate em- bryonic development, stem cell maintenance, root and shoot architecture, and tropic growth responses. The past decade has been marked by dramatic progress in efforts to elucidate the complex mechanisms by which auxin transport regulates plant growth. As the understanding of auxin transport regulation has been increasingly elaborated, it has become clear that this process is involved in almost all plant growth and environmental responses in some way. However, we still lack information about some basic aspects of this fundamental regulatory mechanism. In this review, we present what we know （or what we think we know） and what we do not know about seven auxin-regulated processes. We discuss the role of auxin transport in gravitropism in primary and lateral roots, phototropism, shoot branching, leaf expansion, and venation. We also discuss the auxin reflux/fountain model at the root tip, flavonoid modulation of auxin transport processes, and outstanding aspects of post-translational regulation of auxin transporters. This discussion is not meant to be exhaustive, but highlights areas in which generally held assumptions require more substantive validation.

Epidemiological studies have clearly established that human papillomavirus (HPV) infection is the major risk factor for cervical cancer. most cervical cancers and pre-cancers are HPV- positive. Not all pre-cancers progress to cancer; a significant number regress. The immunological basis for either spontaneous or treatment-mediated recovery from HPV-associated CIN is not clear. Currently, prophylactic vaccines are successfully inducing antibody responses in HPV negative patients. The rapeutic vaccines for HPV-positive patients with disease are needed. There is a need to understand the immunologic basis for the Cell-mediated Immune (CmI) response and for histological regression to help the formulation of therapeutic vaccines. material and methods. Four groups of women were identified for this cross-sectional study of CmI. Group 1 consisted of six women without cytological or histological diagnosis of CIN and with an HPV negative test (CIN(- )/HPV (- )). Group 2 included 31 women with a new histological diagnosis of CIN and HPV positive test (CIN(+ )/HPV(+ )). Groups 3 and 4 were selected from women who had undergone ablative or excisional treatment for CIN at the colposcopy clinic at least 6 months before the study. The women in groups 3 and 4 were (CIN(+ )/HPV(+ )) before CIN treatment. Group 3 consisted of 22 women without evidence of recurrence of CIN (Recur (- )), and group 4 included 10 with histological diagnosis of recurrent CIN (Recur(+ )). In particular, we investigated CmI responses to synthetic peptides from the E6 and E7 oncoproteins of HPV- 16. Results. Compared to patients with disease recurrence (Recur(+ ), n = 10), the majority of individuals who remained recurrence-free post-treatment (Recur(- ), n = 22) exhibited significant proliferative responses to synthetic peptides from the E6 (P = 0.001) and the E7 (P = < 0.001). In particular, significant responses were observed with the E6 peptide Q15L (aa 43- 57, P = 0.006) and the E7 peptide Q19D (aa 44- 62
