Activation of the Ig la I promoter by the transcription factor Ets-1 triggers Ig lal-cal germline transcription in epithelial cancer cells

Immunoglobulins （Igs） are known to be synthesized and secreted only by B lymphocytes. class switch recombination （cSR） is a key event that enables B cells to express Igs, and one of the crucial steps for cSR initiation is the germline transcription of Iggenes. Surprisingly, recent studies have demonstrated that the Iggenes are also expressed in some epithelial cancer cells; however, the mechanisms underlying how cancer cells initiate cSR and express Igs are still unknown. In this study, we confirmed that the Ig la I promoter in cancer cell lines was activated by the Ets- 1 transcription factor, and the activity of the Ig la I promoter and ig lal-c^l germiine transcription were attenuated after knockdown of Ets-1 by specific small interfering RNAs （siRNA）. Furthermore, the expression of Ets-1 and Iga heavy chain in cancer cells was dose dependently upregulated by TGF-I^I. These results indicate that activation of the Ig lal promoter by the transcriotion factor Ets-1 is a critical Dathwav and orovides a novel mechanism for le exoression in non-B cell cancers.

Anti-hyperglycemic effects of aqueous Lenzites betulina extracts from the Philippines on the blood glucose levels of the IcR mice(Mus musculus)

Objective: To examine the anti-hyperglycemic effects of aqueous Lenzites betulina(L. betulina) extracts on normoglycemic glucose-loaded ***: Different doses of aqueous extract from L. betulina were administered to 45 IcR mice(Mus musculus) to determine whether there was an effect of L. betulina extracts on the blood glucose level of the IcR mice. Aqueous extracts of L. betulina were orally gavaged to mice using oral glucose tolerance test. A total of five groups were used to determine the effect of the fungi on blood glucose of the mice. Group A(positive control)was given 16.7 mg/kg glimepiride; Group B(negative control) was given distilled water;Group c(low dosage) was given 200 mg/kg aqueous extract; Group D(mid dosage) was given 400 mg/kg aqueous extract and Group E(high dosage) was given 800 mg/kg aqueous extract. Baseline blood glucose value was firstly acquired before induction of hyperglycemia through D-glucose, after which another check on blood glucose was made after 0.5 h. Immediately, after the acquisition of hyperglycemic blood glucose level, the individual administration of treatments were done. After that, three blood collections were done spanning 3 h with 1 h ***: The low dose(200 mg/kg) and the mid dose(400 mg/kg) of L. betulina extracts were significantly different(P ctive baseline values throughout the whole experiment with the latter surpassing its baseline value during the 3rd hour. On the other hand, the high dose(800 mg/kg) during the 1st hour after administration was not significantly different(P > 0.05) from its corresponding baseline value, acting faster than the positive control(glimepiride), which only became significantly different(P ct is able to produce hypoglycemic effects on the mice with all doses, which are able to normalize blood glucose levels at varying times.

食品的微生物检测涉及到样品前处理和后续的均质。本实验对Stomacher、Pulsifier、Bagmixer和Smasher共4种拍击式均质机进行了四方面的比较:10种食品的活菌计数;距离5英尺远,人工和分贝计比较设备操作过程中的噪音强度;设备使用后清洗的难易程度;操作性。具体操作是从10种食品各取25g,加入装有224.5mL 0.1%的蛋白胨水的样本袋中,再加入0.5mL大肠杆菌菌液。每种食品都用拍击式均质机均质60s。以拍击式均质机Stomacher作为参照,确定设备使用后清洗的难易程度及操作性。结果表明:4种拍击式均质机活菌计数的结果相似,Smasher和Bagmixer的噪音强度最低,Stomacher的噪音强度其次,Pulsifier的噪音强度最高。Smasher使用后清洗和操作最方便,依次分别是Bagmixer、Stomacher和Pulsifier。

The Department of Veterans Affairs (VA) manages the largest healthcare system under a single management structure in the United States. Providing access to high quality healthcare to the VA’s nearly 9 million enrollees is a mission priority for the Veterans Health Administration (VHA), the arm of the VA that oversees all medical facilities and operations. Geographic Information Systems (GIS) tools enable analysts to construct data-driven recommendations to policy makers for providing the best and most timely healthcare to those individuals who have honorably served their country. This paper illustrates how GIS is being used by the VHA and provides the example of acute stroke care access for Veterans in one Veterans Integrated Service Network (VISN) within the VHA.

BAckGROUND Sleep disturbance is a core feature of post-traumatic stress disorder(PTSD).Given the relationship between sleep disturbance and PTSD,there has been a relative paucity of studies examining the potential therapeutic impact of using pharmacotherapy to target sleep disturbance in patients with ***(ESZ)is a non-benzodiazepine y-aminobutyric acid-A receptor agonist indicated for the treatment of sleep and may affect sleep in patients with *** To evaluate the efficacy of ESZ vs placebo(PBO)for patients with PTSD and *** The study was a 12-wk,double blind,randomized controlled trial with 3 mg of ESZ(n=13)or PBO(n=12).RESULTS Patients in both arms experienced significant improvement in PTSD symptoms as assessed by the clinician-Administered PTSD Scale for DSM-IV(cAPS):ESZ(t11=-3.12,P=0.005)and PBO(t11=-3.5,P=0.002)and by self-report with the Short PTSD Rating Interview(ESZ t11=-3.38,P=0.003 and PBO t11=-4.48,P=0.0005).There were no significant differences between treatments on the cAPS(t22=-0.13,P=0.70)or the Short PTSD Rating Interview(t22=-0.58,P=0.56).Similarly,both treated groups improved on sleep measures as assessed by the Pittsburgh Sleep Quality Index with PTSD Addendum(PSQI)and on total sleep time(TST)and sleep latency assessed by actigraphy with no significant differences between groups(PSQI t22=-0.24,P=0.81;total sleep time t10=0.13,P=0.90 and sleep latency t10=0.68,P=0.50).There was a significant correlation between improvement in sleep and overall improvement in PTSD as measured by change scores on the PSQI and cAPS,r(8)=0.79,P=0.01 for ESZ treated subjects,but not for those treated with PBO r(9)=0.16,P=*** events of ESZ were consistent with the known profile of the medication including dysgeusia(30%,mild),sedation(20%,mild)and headache(20%,moderate to severe).cONcLUSION Results do not support the hypothesis of a specific positive effect of ESZ compared to PBO for measures of PTSD and associated sleep disturban

The microstructural evolution behaviors of 18cr-ODS steel under electron beam irradiation were in-situ investigated in a high resolution transmission electron microscopy(HRTEM).Fe3O4 particle was induced by electron beam irradiation through a series of dynamic procedures including lattice relaxation of the steel matrix,cr and Ni segregations,combination of iron and oxygen atoms,epitaxial growth as well as separation of Fe3O4 *** size of oxides increased with increasing electron irradiation fluence,and it reached up to about 6 nm at the fluence of 6.1×1023e/cm2.

Estrogen is implicated as playing an important role in aging and tumorigenesis of estrogen responsive tissues;however the mechanisms underlying the mitogenic actions of estrogen are not fully *** we report that estrogen deficiency in mice caused by targeted disruption of the aromatase gene results in a signi-ficant inhibition of telomerase maintenance of telomeres in mouse ovaries in a tissue-specific *** inhibition entails a significant shortening of telomeres and compromised proliferation in the follicular granulosa cell compartment of *** expression analysis showed decreased levels of proto-oncogene c-Myc and the telomerase catalytic subunit,telomerase reverse transcriptase(TERT),in response to estrogen *** replacement therapy led to increases in TERT gene expression,telomerase activity,telomere length and ovarian tissue growth,thereby reinstating ovary development to normal in four *** data demonstrate for the first time that telomere maintenance is the primary mechanism mediating the mitogenic effect of estrogen on ovarian granulosa cell proliferation by upregulating the genes of c-Myc and TERT in *** deficiency or over-activity may cause ovarian tissue aging or tumorigenesis,respectively,through estrogen regulation of telomere remodeling.

A comparative study of machine learning in predicting the mechanical properties of the deposited AA6061 alloys via additive friction stir deposition

Additive friction stir deposition(AFSD)provides strong flexibility and better performance in component design,which is controlled by the process *** is an essential and difficult task to tune those *** recent exploration of machine learning(ML)exhibits great potential to obtain a suitable balance between productivity and set *** this study,ML techniques,including support vector machine(SVM),random forest(RF)and artificial neural network(ann),are applied to predict the mechanical properties of the AFSD-based AA6061 *** for the stable parameters(temperature,force and torque)in situ monitored by the self-developed process-aware kit during the AFSD process and the other factors(rotation speed,traverse speed,feed rate and layer thickness)are also set as input *** output variables are microhardness and ultimate tensile strength(UTS).Prediction results show that the ann model performs the best prediction accuracy with the highest R2(0.9998)and the lowest mean absolute error(MAE,0.0050)and root mean square error(RMSE,0.0063).Furthermore,analysis suggests that the feed rate(24.8%/24.1%)and layer thickness(25.6%/26.6%)indicate a higher contribution that affects the mechanical properties.
