N-methyladenosine(mA) is a chemical modification present in multiple RNA species and is added to mRNA by a multisubunit ‘writer’ complex composed of the METTL3/METTL14/WTAP heterotrimer and numerous additional adaptor proteins. Studies on mA reveal its comprehensive roles in almost every aspect of mRNA metabolism and mA has emerged as a widespread regulatory mechanism that controls gene expression in diverse physiological processes. Type I IFNs are important for host defence against viruses. To avoid excessive or dysregulated IFN responses, the host has developed sophisticated mechanisms at multiple-levels to maintain appropriate IFN responses and ensure the most favorable outcome. Some recent discoveries indicate that mA can affect the life cycles of numerous viruses as well as the cellular antiviral immune response. However, the specific mechanisms, especially whether and how mA modification regulate the production of type I IFNs and ISGs, are still largely unknown. Here, we reveal that WT1-associated protein(WTAP), one of the mA ‘writers’, is degraded via the ubiquitination-proteasome pathway upon activation of IFN-I signaling. By screening the key components involved in IFN-I signaling after knocking down WTAP, we find that degradation of WTAP can block antiviral responses by reducing the protein abundance of IFN-regulatory factor 3(IRF3) and interferon alpha/beta receptor subunit 1(IFNAR1). Further tests show that the mA levels of IRF3 and IFNAR1 mRNAs are reduced with the degradation of WTAP, leading to translational suppression of IRF3 and instability of IFNAR1 mRNAs. Thus, the WTAP-IRF3/IFNAR1 axis may serve as negative feedback pathway to fine-tune the activation of IFN-I signaling to protect the host from immunopathology of overreactive and harmful IFN-I production, which highlights the sophisticated regulatory mechanisms of antiviral responses at post-transcriptional level and the roles of mA in the antiviral responses by dictating the fate of mRNAs assoc

Polymorphisms of chemokine receptors and its ligand alleles influencing genetic suscepti-bity to HIV-1 infection in eight ethnic groups in Chinese mainland

Limited genetic information is available concerning the polymorphisms of HIV-1 resistant genes in indigenous Chinese populations. The aim of this study is to identify the allelic frequencies of the chemokine and chemokine receptor genes in the Chinese mainland. Genomic DNA samples extracted from whole blood of 2318 subjects were analyzed by using PCR or PCR/restriction fragment length polymorphism (RFLP) assays, and further confirmed by direct DNA sequencing. Higher frequencies of mutant CCR2-64I (19.15%-28.79%) and SDF1-3’A (19.10% -29.86%) alleles were found in subjects of 8 ethnic groups in the Chinese mainland. In contrast, the △32 mutation in CCRS gene occurs at a very low frequency (0.0016, n=1287) in Han population. A relatively high frequency of CCR5-wt/△32 heterozygotes was observed in Uygurian and Mongolian populations. No A32 mutation allele was detected in Tibetan and other 4 ethnic groups in Yunnan Province. There was no CCR5-m303 mutation in subjects of any ethnic group in the Chinese

Celastrol alleviates arthritis by modulating the inflammatory activities of neutrophils

目的构建预测急诊脓毒性休克患者28 d病死率的列线图模型,并进行预测效能验证。方法基于天津医科大学朱宪彝纪念医院、天津医科大学总医院和天津医科大学第二医院的急诊医学科病案机构数据库,收集2017年1月至2020年10月急诊收治的913例脓毒性休克患者的信息,包括人口基线资料和临床特征、实验室指标及主要结局(28 d病死率)。根据简单随机抽样法将患者分为训练集和验证集。将训练集单因素二元Logistic回归分析中所有重要的变量纳入多因素Logistic回归分析,分析急诊脓毒性休克患者28 d死亡的危险因素,并构建列线图模型。采用校准曲线和受试者工作特征曲线(ROC曲线)评估列线图模型的预测效能。结果最终纳入860例符合标准的脓毒性休克患者,其中训练集472例,验证集388例。训练集和验证集的28 d病死率分别为52.5%(248/472)、54.1%(210/388)。在训练集中,死亡组的年龄、呼吸频率(RR)、C-反应蛋白(CRP)、D-二聚体、白细胞计数(WBC)、中性粒细胞计数(NEU)、中性粒细胞/淋巴细胞比值(NLR)、单核细胞/淋巴细胞比值(MLR)、平均血小板体积(MPV)、血小板计数(PLT)水平均明显高于存活组,碱剩余(BE)、淋巴细胞计数(LYM)、血红蛋白(Hb)水平和慢性阻塞性肺疾病(COPD)比例均明显低于存活组(均P<0.05)。多因素Logistic回归分析显示,NLR〔优势比(OR)=0.0230,95%可信区间(95%CI)为-0.2044~0.1130〕、MPV(OR=0.1798,95%CI为-0.8776~0.1727)、Hb(OR=0.0078,95%CI为0.0103~0.0408)、降钙素原(PCT;OR=1.9570,95%CI为1.2430~3.0810)和D-二聚体(OR=0.0001,95%CI为-0.0004~0.0001)是急诊脓毒性休克患者28 d病死率的独立预测因子(均P<0.05)。基于上述变量建立列线图模型,ROC曲线显示,训练集和验证集中列线图模型预测脓毒性休克患者28 d病死率的ROC曲线下面积(AUC)分别为0.907(95%CI为0.864~0.940)、0.822(95%CI为0.781~0.863)。校准曲线显示,训练集及验证集的预测结果与观察结果之间均具有良好的一致性。结论基于NLR、MPV、Hb、PCT和D-二聚体构建的列线图模型对预测急诊脓毒性休克患者的28 d病死率具有重要的临床价值。

中国式现代化道路是在中国共产党领导下探索出来的新型现代化道路,“社会主义”和“中国国情”是蕴含其中的关键词。新中国成立以来,中国式现代化道路经历了起步探索、快速发展和深化拓展三个阶段。从其属性特征来说,中国式现代化道路是一条正确处理普遍性与特殊性关系的特色之路、正确处理继承性与发展性关系的拓新之路、正确处理长期性与阶段性关系的递进之路、正确处理自主性与借鉴性关系的开放之路。事实证明,中国式现代化道路是一条能够走得通、走得好的正确道路,其路径指向在于始终坚持科学理论指导、先进政党领导、人民利益至上、立足中国国情等。不断坚持和继承以上有益经验,中国式现代化道路必然越走越宽广。
