流感病毒、新冠病毒、乙肝病毒、登革病毒等给人们生命健康带来严重危害的同时,也给全球经济带来巨大的冲击.因此,抗病毒天然免疫一直是免疫学及微生物学领域的研究热点.宿主免疫系统通过复杂的信号网络来感知这些病原体并启动针对感染的免疫反应,在清除病毒感染的同时修复受损组织,维持机体免疫稳态.目前,越来越多的研究表明,包括转录后加工和修饰在内的表观遗传修饰在帮助机体启动抗病毒免疫并维持免疫稳态的同时,促进了病毒与宿主的协同演化.本文重点关注转录后加工和修饰,包括选择性剪接、选择性多聚腺苷化、RNA修饰和编辑等,如何在RNA层面调节宿主抗病毒免疫功能的发挥及病毒基因的表达.基于这些研究进展,本文探讨了病毒如何修改或利用转录后加工和修饰过程来逃避宿主免疫监测,以及转录后加工和修饰如何塑造宿主-病毒相互作用并介导二者之间的协同演化.最后,讨论了转录后加工修饰在病毒感染及其他免疫相关性疾病的诊疗以及mRNA疫苗开发中的应用前景.

Objective:To identify potential serum protein candidates involved in linking the traditional Chinese medicine(TCM)-defined qi deficiency constitution(QDC)to Pi-qi-deficiency syndrome(PQDS)of chronic superficial gastritis(CSG).Methods:Using participants with the TCM-defined balanced constitution as a control population,labelfree quantitative proteomics was adopted to identify differentially expressed proteins(DEPs)in serum samples from two case populations:case population 1(participants with QDC)and case population 2(patients with PQDS of CSG).The DEPs discovered in both case populations were analyzed to identify common DEPs as potential candidates for proteins involved in the link between QDC and *** on Kyoto Encyclopedia of Genes and Genomes pathway(KEGG)and Gene Ontology(GO)enrichment analysis and analysis of proteineprotein interaction networks,we evaluated the possible functions of these potential serum ***:We discovered 24 and 28 proteins that were differentially expressed in case populations 1 and 2,respectively,compared with the control *** clustering analysis showed that the expression profile of DEPs of individuals from the same population clustered well,while those from different populations were ***,GO analysis revealed the 10 DEPs that were common to both case populations to be mainly associated with negative regulation of cellular metabolic and immune system processes while KEGG analysis indicated these proteins to be associated with complement and coagulation cascades and peroxisome proliferator-activated receptor ***,serum levels of C4b-binding protein beta chain,glycosylphosphatidylinositol-specific phospholipase D1 and MS-F1 light chain variable region proteins were notably higher in the two case populations compared with the control,particularly in the case of CSG with ***:The results presented here provide new insights into the molecular mechanisms underlying de

Animals exploit different germline-encoded proteins with various domain structures to detect the signature molecules of pathogenic *** molecules are known as pathogen-associated molecular patterns(PAMPs),and the host proteins that react with PAMPs are called pattern recognition proteins(PRPs).Here,we present a novel type of protein domain structure capable of binding to bacterial peptidoglycan(PGN)and the minimal PGN motif muramyl dipeptide(MDP).This domain is designated as apextrin C-terminal domain(ApeC),and its presence was confirmed in several invertebrate phyla and *** apextrin-like proteins(ALP1 and ALP2)were identified in a basal chordate,the Japanese amphioxus Branchiostoma japonicum(bj).BjALPl is a mucosal effector secreted into the gut lumen to agglutinate the gram-positive bacterium *** via PGN *** of secreted bjALPl by anti-bjALP1 monoclonal antibodies caused serious damage to the gut epithelium and rapid death of the animals after bacterial infection BjALP2 is an intracellular PGN sensor mat binds to TRAF6and prevents TRAF6 from self-ubiquitination and hence from NF-kB *** was found to compete with TRAF6 for bjALP2,which released TRAF6 to activate the NF-κB *** and bjALP2 therefore play distinct and complementary functions in amphioxus gut mucosal *** conclusion,discovery of the ApeC domain and the functional analyses of amphioxus ALP1 and ALP2 allowed us to define a previously undocumented type of PRP that is represented across different animal phyla.

The ability to detect and respond to changes in the extracellular environment is a basic necessity for survival of all organisms. Consequently, diverse bio-rhythms are generated by hundreds of cellular oscillators that somehow manage to operate synchronously under various fluctuated environments. It remains, however, to be exploited how behaviors of a noisy microscopic molecule affect the macroscopic behaviors in the weak coupling or even without coupling between cells. Using a multi-cell system, we show that a common noisy signaling molecule can induce inphase synchronization across an ensemble of independent gene oscillators in Escherichia coli, leading to a collective response or rhythm. Such a mechanism of achieving biological rhythms would be exploited by realistic living cells to sense the extracellular signal. In addition, the model system considered here provides a quantitative example of phase transition from non-synchronization to synchronization across a population of biological oscillators.
