Predicting the effectiveness of protected areas of Natura 2000 under climate change

Background:Protected areas(PAs)are aimed to hold the environmental conditions that facilitate species and ecosystems to *** can become climatically unsuitable and unable to sustain their current number of species under climate *** Natura 2000(N2K)is the largest coordinated conservation tool assigned to maintain the long-term survival of Europe’s most significant species and *** attempting to understand the effectiveness of PAs in the face of climate change scenarios,we tested two hypotheses:(1)PAs in the Alpine and the Boreal biogeographical regions will experience more newly emerged climate conditions(hotter and drier)compared to the climate representation of other biogeographical regions under future climate in Europe and(2)PAs in the Mediterranean and the Continental biogeographical regions will face more consistency in climate conditions due to less area of disappearing and novel climate in ***:Current climate data(1960–1990)and projections for 2050 and 2070 of PAs of N2K were extracted from WorldClim global climate *** components analysis(PCA)was performed to construct climate space for the PAs across the biogeographical regions based on 19 climatic variables assessed at 5-km *** 10.1 was used to map the location of the novel and disappearing ***:PAs in the Alpine region will experience more novel climate conditions in the future compared to other biogeographical *** future projections showed that 17.70%of the PAs in the Alpine region will experience novel climate by *** climate consistency was observed in the PAs in the Continental region compared to the other biogeographical *** results showed that about 176 km2 of the selected PAs in the Continental region will face new emerging climate,while about 110 km2 will disappear under RCP 8.5 *** prediction also revealed that in the Mediterranean region 08 PAs will experience novel climate and 786 km2 areas

Intelligent manufacturing system of impeller for computer numerical control(CNC) programming based on KBE

To solve the problem of advanced digital manufacturing technology in the practical application, a knowledge engineering technology was introduced into the computer numerical control(CNC) programming. The knowledge acquisition, knowledge representation and reasoning used in CNC programming were researched. The CNC programming system functional architecture of impeller parts based on knowledge based engineering(KBE) was constructed. The structural model of the general knowledge-based system(KBS) was also constructed. The KBS of CNC programming system was established through synthesizing database technology and knowledge base theory. And in the context of corporate needs, based on the knowledge-driven manufacturing platform(i.e. UG CAD/CAM), VC++6.0 and UG/Open, the KBS and UG CAD/CAM were integrated seamlessly and the intelligent CNC programming KBE system for the impeller parts was developed by integrating KBE and UG CAD/CAM system. A method to establish standard process templates was proposed, so as to develop the intelligent CNC programming system in which CNC machining process and process parameters were standardized by using this KBE system. For the impeller parts processing, the method applied in the development of the prototype system is proven to be viable, feasible and practical.

Surveying the Oligomeric State of Arabidopsis thaliana Chloroplasts

Blue native-PAGE （BN-PAGE） resolves protein complexes in their native state. When combined with immu- noblotting, it can be used to identify the presence of high molecular weight complexes at high resolution for any protein, given a suitable antibody. To identify proteins in high molecular weight complexes on a large scale and to bypass the requirement for specific antibodies, we applied a tandem mass spectrometry （MS/MS） approach to BN-PAGE-resolved chloroplasts. Fractionation of the gel into six bands allowed iden- tification and label-free quantification of 1000 chloroplast proteins with native molecular weight separation. Significantly, this approach achieves a depth of identification comparable with traditional shotgun proteo- mic analyses of chloroplasts, indicating much of the known chloroplast proteome is amenable to MS/MS identification under our fractionation scheme. By limiting the number of fractionation bands to six, we facil- itate scaled-up comparative analyses, as we demonstrate with the reticulata chloroplast mutant displaying a reticulated leaf phenotype. Our comparative proteomics approach identified a candidate interacting protein of RETICULATA as well as effects on lipid remodeling proteins, amino acid metabolic enzymes, and plastid division machinery. We additionally highlight selected proteins from each sub-compartment of the chloroplast that provide novel insight on known or hypothesized protein complexes to further illus- trate the utility of this approach. Our results demonstrate the high sensitivity and reproducibility of this technique, which is anticipated to be widely adaptable to other sub-cellular compartments.

Structural Health Monitoring(SHM)is the process of collecting,interpreting and analysing data from structures in order to determine its health status and the remaining life *** materials have been extensively use in recent years in several industries with the aim at reducing the total weight of structures while improving their mechanical ***,composite materials are prone to develop damage when subjected to low to medium impacts(i.e.1-10 m/s and 11-30 m/s respectively).Hence,the need to use SHM techniques to detect damage at the incipient initiation in composite materials is of high *** the availability of several SHM methods for the damage identification in composite structures,no single technique has proven suitable for all *** must be noted that the amount of techniques available nowadays is too extensive to be comprehensively reviewed in a single ***,the focus will be on techniques that can serve as a starting point for studies focusing on damage detection,localisation,assessment and prognosis on certain kinds of ***,the line of thought behind the search and the structure of this review is a result of objectives beyond the scope of the paper ***,it was considered that,once the above was understood,an updated synopsis such as this could also be useful for other researchers in the same field.

Enhanced migration of tissue inhibitor of metalloproteinase overexpressing hepatoma cells is attributed to gelatinases: Relevance to intracellular signaling pathways

AIM: To study the effect of gelatinases (especially MMP-9) on migration of tissue inhibitor of metalloproteinase (TIMP-1) overexpressing hepatoma cells. METHODS: Wild type HepG2 cells, cells stably transfected with TIMP-1 and TIMP-1 antagonist (MMP-9-H401A, a catalytically inactive matrix metalloproteinase (MMP) which still binds and neutralizes TIMP-1) were incubated in Boyden chambers either with or without Galardin (a synthetic inhibitor of MMP-1, -2, -3, -8, -9) or a specific inhibitor of gelatinases. RESULTS: Compared to wild type HepG2 cells, the cells overexpressing TIMP-1 showed 115% migration (P<0.05) and the cells overexpressing MMP-9-H401A showed 62% migration (P<0.01). Galardin reduced cell migration dose dependently in all cases. The gelatinase inhibitor reduced migration in TIMP-1 overexpressing cells predominantly. Furthermore, we examined intracellular signal transduction pathways of TIMP-1-dependent HepG2 cells. TIMP-1 deactivates cell signaling pathways of MMP-2 and MMP-9 involving p38 mitogen-activated protein kinase. Specific blockade of the ERK pathway suppresses gelatinase expression either in the presence or absence of TIMP-1. CONCLUSION: Overexpressing functional TIMP-1-enhanced migration of HepG2-TIMP-l cells depends on enhanced MMP-activity, especially MMP-9.

Magnetic resonance(MR)imaging based volumetry is recognized as an important technique for studying the *** this review,two principle volumetric methods using high resolution MR images were introduced,namely the Cavalieri method and the voxel based morphometry(VBM).The Cavalieri method represents a manual technique that allows the volume of brain structures to be estimated efficiently with no systematic error or sampling bias,whereby the VBM represents an automated image analysis which involves the use of statistical parametric mapping of the MR imaging *** methods have been refined and applied extensively in recent neuro- science *** present paper aims to describe the development of methodologies and also to update the knowledge of their applica- tions in studying the normal and diseased brain.

Semiautomated 3D Root Segmentation and Evaluation Based on X-Ray CT Imagery

Polymorphisms in NF-κB,PXR,LXR,PPARγ and risk of inflammatory bowel disease

AIM:To investigate the contribution of polymorphisms in nuclear receptors to risk of inflammatory bowel disease(IBD) . METHODS:Genotypes of nuclear factor(NF) -κB(NFKB1) NFκB-94ins/del(rs28362491) ;peroxisome proliferatoractivated receptor(PPAR) -γ(PPARγ) PPARγPro12Ala(rs1801282) and C1431T(rs 3856806) ;pregnane X receptor(PXR) (NR1I2) PXR A-24381C(rs1523127) ,C8055T(2276707) ,and A7635G(rs 6785049) ;and liver X receptor(LXR) (NR1H2) LXR T-rs1405655-C and T-rs2695121-C were assessed in a Danish case-control study of 327 Crohn's disease patients,495 ulcerative colitis(UC) patients,and 779 healthy *** ratio(OR) and 95%CI were estimated by logistic regression models. RESULTS:The PXR A7635G variant,the PPARγPro12Ala and LXR T-rs2695121-C homozygous variant genotypes were associated with risk of UC(OR:1.31,95% CI:1.03-1.66,P=0.03,OR:2.30,95%CI:1.04-5.08,P=0.04,and OR:1.41,95%CI:1.00-1.98,P=0.05,respectively) compared to the corresponding homozygous wild-type *** never smokers,PXR A7635G and the LXR T-rs1405655-C and T-rs2695121-C variant genotypes were associated with risk of IBD(OR:1.41,95%CI:1.05-1.91,P=0.02,OR:1.63,95%CI:1.21-2.20,P=0.001,and OR:2.02,95%CI:1.36-2.99,P=0.0005,respectively) compared to the respective homozygous variant *** A7635G(rs6785049) variant genotype was associated with a higher risk of UC diagnosis before the age of 40 years and with a higher risk of extensive disease(OR:1.34,95%CI:1.03-1.75 and OR:2.49,95%CI:1.24-5.03,respectively) . CONCLUSION:Common PXR and LXR polymorphisms may contribute to risk of IBD,especially among never smokers.
