Development of a sample preparation approach to measure the size of nanoparticle aggregates by electron microscopy

Electron microscopy (EM) is widely used for nanoparticle (NP) sizing. Following an initial assessment of two sample preparation protocols described in the current literature as"unperturbed", we found that neither could accurately measure the size of NPs featuring a broad size distribution, e.g., aggregates. Because many real-world NP samples consist of aggregates, this finding was of considerable concern. The data showed that the protocols introduced errors into the measurement by either inducing agglomeration artefacts or providing a skewed size distribution towards small particles (skewing artefact). The focus of this work was to develop and apply a mathematical refinement to correct the skewing artefact. This refinement provided a much improved agreement between EM and a reference methodology, when applied to the measurement of synthetic amorphous silica NPs. Further investigation, highlighted the influence of NP chemistry on the refinement. This study emphasised the urgent need for greater and more detailed consideration regarding the sample preparation of NP aggregates to routinely achieve accurate measurements by EM. This study also provided a novel refinement solution applicable to the size characterisation of silica and c让rate-coated gold NPs featuring broad size distributions. With further research, this approach could be extended to other NP types.

The color of watermelon flesh is an important trait determined by a series of ***,we used cream of Saskatchewan(pale yellow flesh)and PI 186490(white flesh)as parental materials for an F2 segregation and initial mapping using the bulked segregant analysis sequencing(BSA-seq)*** BSA results revealed a flesh colorrelated QTL that spans approximately 2.45 Mb on chromosome *** region was preliminarily positioned in a 382-kb segment,and then narrowed down into a 66.8-kb segment with 1260 F2 individuals.A total of nine candidate genes were in the fine mapping interval,but only cla007528(encoding chlorophyllase)had non-synonymous mutations and was significantly expressed between the parental materials throughout flesh *** also checked the expression patterns of the carotenoid metabolic pathway genes based on RNA-seq data and qRT-PcR *** genes in the xanthophyll cycle(clcHYB,clNcED-1 and clNcED-7)exhibited differential expression patterns between the two parental lines at different flesh color formation ***1,clPDS,clZDS,clcHXE,clcRTISO and clLcYB also exhibited clearly different expression patterns accompanied by carotenoid accumulation.

We report amphibian and reptile distribution records based on recent biodiversity surveys conducted at the Angat Watershed Reservation, Bulacan Province, Luzon Island, Philippines. This watershed constitutes the principal water source for Manila, the Philippines＇ largest metropolitan area. As virtually nothing is known of the herpetological diversity of the immediate area and the surrounding Bulacan Province, all species recorded as part of our surveys constitute major geographical records and/or significant range extensions. Our data result in a total of 63 new records of amphibian （19 frogs） and reptile （22 lizards, 2 turtles, and 20 snakes） species for this protected area （and immediate vicinity） that serves as a watershed for the major metropolitan area of Manila and surrounding cities. Together with the few previous literature records, our new records bring the total number of amphibian and reptile species for Bulacan Province to 68. We discuss several strategies for future survey work （focusing on habitat type, seasonal variation, and elevational variability） that we anticipate will result in increased knowledge of diversity within the Angat Watershed Reserve. The impressive level of herpetological diversity within such a small area, so close to Metro Manila, emphasizes that the diversity and distribution patterns of amphibians and reptiles from Luzon are still poorly known and in need of further study.

Biomass responses to intraspecific competition differ between wild species and cO_(2)

Aims How growth of wild and crop species responds to global environmental perturbations has both ecological and agricultural significance in a changing *** primary aim of this synthesis was to quantitatively assess the interactive effects of intraspecific competition and elevated cO_(2) on biomass production in herbaceous *** Using meta-analytical techniques,we synthesized data from publications before 2006 that reported biomass responses to elevated cO_(2) in 321 herbaceous species grown in isolation or in competition with con-specific *** findings Intraspecific competition differentially modified biomass responses to elevated cO_(2) in wild and crop *** example,competition reduced cO_(2) stimulation of total biomass(WT)from 27 to 23%in wild species,but by a much greater magnitude,i.e.,from 43 to 32%in *** had no effect on responses of either above-(WAG)or below-ground(WBG)biomass to elevated cO_(2) in wild species,but significantly diminished cO_(2) enhancement of WAG,although not of WBG,in *** variations were found among functional groups in the modification of growth responses to elevated cO_(2) by intraspecific competition,which exerted greater depression on cO_(2) enhancement in c3 than in c4 species and in non-legumes than in *** cO_(2) affected leaf and stem growth of individually grown c3 graminoids and forbs similarly,but increased leaf growth only in c4 graminoids and stem growth only in c4 *** from this synthesis demonstrated that intraspecific competition differentially affected growth responses to elevated cO_(2) in wild and crop *** wildcrop species differences will have important implications for understanding primary production by herbaceous species in both natural and agricultural ecosystems in the future when atmospheric cO_(2) is significantly higher than the current level.

BRcA mutation rate and characteristics of prostate tumor in breast and ovarian cancer families:analysis of 6,591 Italian pedigrees

Objective:As prostate cancer(Pr c)shows a BRcA mutation rate as high as 30%,it becomes crucial to find the optimal selection criteria for genetic *** primary objective of this study was to evaluate the BRcA mutation rate in families with Pr c associated with breast and/or ovarian cancers;secondary aims were to compare the characteristics of families and BRcA-related Pr c outcome among BRcA1 and BRcA2 ***:Following the Modena criteria for the BRcA test,we evaluated the mutation rate in families with breast and/or ovarian cancer with a Gleason score≥7 Pr cs,by testing breast or ovarian cases and inferring the mutation in the prostate *** characteristics of families and BRcA-related Pr c outcomes were measured using the chi-square(χ^(2))test and Kaplan–Meier methods,***:Among 6,591 families,580(8.8%)with a Gleason score≥7 Pr cs were identified,of which 332(57.2%)met the Modena selection criteria for BRcA ***,215 breast or ovarian cancer probands(64.8%)were tested,of which 41 resulted positive for BRcA and one for cHEK2 genes(19.5%).No statistically significant differences were found in BRcA-related Pr c prognosis and in the characteristics of families among BRcA1,BRcA2 and non-tested *** of 23(44%)mutations in the BRcA2 gene fell in the prostate cancer cluster region(PccR)at the 3′terminal of the 7914 ***:It appears the Modena criteria are very useful for BRcA testing selection in families with breast and/or ovarian cancer and Pr c.A trend toward a worse prognosis has been found in BRcA2 carriers.

Lessons from the past:isotopes of an endangered rail as indicators of underlying change to tidal marsh habitats

Introduction:Tidal marsh systems along the Pacific coast of the United States have experienced substantial stress and loss of area and ecosystem function,which we examined by using the endangered california Ridgway’s Rail,Rallus obsoletus obsoletus(‘rail’)as an indicator of its tidal marsh habitat in the San Francisco *** organized a collection of historical(1885-1940)and modern(2005-2014)rail feathers and analyzed the feather isotope means for delta carbon(δ^(13)c),sulfur(δ^(34)S),and nitrogen(δ^(15)N)by region and time ***:Feather isotopes represented the primary foraging habitat during historical then modern time *** individual nor regional rail feather isotopes suggested freshwater or terrestrial foraging by the *** regions with both historic and modern feather isotopes revealed non-uniform spatial shifts in isotope levels consistent with a marine based food web and significantδ^(15)N ***:Our results supported the rail’s status as a generalist forager and obligate tidal marsh species throughout the historic *** variable isoscape trends generated from feather isotope means illustrated a modern loss of the isotopic homogeneity between regions of historical tidal marsh,which correlated with spatially-explicit habitat alterations such as increasing biological invasions and sewage effluent over ***:These findings have reinforced the importance of tidal marsh conservation in the face of ongoing underlying changes to these important ecosystems.
