＜正＞A Parallel Expression system has been developed in our laboratory based in part on Gateway technology. The new system allows for the independent expression of multiple genes in the same bacterial cell. At the current stage of development this system allows for the co-expression of up to four different genes or gene constructs. The system has the potential of parallel expression of 8 or more genes if desired.

目的探讨VERGE蛋白在小鼠大脑中动脉梗死模型(McAO)中蛋白表达的变化及其机制。方法c57BL/6小鼠30只,随机分为2组:30 min McAO模型组、90 min McAO模型组,制备McAO模型,应用激光多普勒血流仪进行术中脑血流监测,应用Western印迹法观察各组梗死侧及非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达的变化。结果30 min、90 min McAO模型组梗死侧VERGE、p-JNK2的蛋白表达明显增高,p-ERK1/2、P-38蛋白表达无变化;两组McAO模型非梗死侧的VERGE、p-JNK2、p-ERK1/2、P-38蛋白表达均无变化。结论VERGE蛋白表达在小鼠McAO的早期明显升高可能与JNK2通路有关而与ERK、P38通路无关。

AIM: To determine whether IFN-α is the agent that turns a slightly effective treatment (radiochemotherapy) into a potent therapy, we tested IFN-α for its synergistic ***: Eight pancreatic carcinoma cell lines were treated with the single agents and combinations of *** role of IFN-α regarding a) direct inhibitory effects; b)radio and chemosensitizing effects; c) anti-angiogenic properties and d) enhancement of immunogenicity was ***: Our results show that IFN-α has direct inhibitory properties and some synergistic influence as determined by AnnexinV/PI stain and cell count. IFN-α is also able to prevent the increase in proliferation rate and VEGF secretion of cDDP resistant cells. Having taken the results from immunogenicity experiments together, we found cells that can be influenced by IFN-α but were less susceptible against T cells. Furthermore, high expression of MHc molecules, cD118, EGF-R and Fas was predictive for a good ***: In conclusion, IFN-α has direct cytotoxic effects, acts as a radiosensitizer and circumvents tumor cell-regrowth after cDDP treatment. These mechanisms may be responsible for the good clinical outcome of capRI.

中国家庭普惠金融的发展及影响

本文利用2017年中国家庭普惠金融调查数据构建了一个全面的普惠金融指标体系,并合成了家庭普惠金融指数,在此基础上从微观家庭视角展示了中国家庭普惠金融发展的总体状况及其在不同群体间的差异。研究结果显示,2017年中国家庭普惠金融总指数平均为0.575,家庭普惠金融发展总体处于中上等水平。从需求层面看,中国社会医疗保险和养老保险发展成效可观,传统银行账户服务和数字金融服务都还有较大推广空间,家庭正规信贷市场参与度明显较低,金融服务质量的提升任重道远。从供给层面看,社区层面的金融基础设施覆盖大致处于中等水平,依然需要重视提高金融服务可及性。农户、贫困家庭、低收入家庭、年老家庭尤其是夹心层家庭,享受到的普惠金融水平明显较低。基于世界银行和国际货币基金组织数据的国际比较发现,中国普惠金融发展领先于大部分金砖国家,但是和欧美发达国家相比存在较大差距。影响方面,地区和家庭层面的证据均显示出普惠金融与经济发展的正向关系。家庭层面研究表明,普惠金融对家庭人均收入的异质性具有较强的解释力度,并且更有利于提高低收入家庭的收入水平。本文的研究成果可为发展普惠金融的政策制定提供重要参考。

Objective:To describe and molecularly characterize an outbreak of dengue virus(DENV)infection in cajamarca,an Andean region in ***:A total of 359 serum samples from patients with acute febrile illness were assessed for the presence of DENV via RT-PcR,ELISA NS1,Ig M and Ig G in cajamarca,Peru from January 2017 to June *** evaluation of the different diagnostic tests and their applicability was ***:Dengue virus was detected in 24.7%of samples by ***,serological analysis detected 30.3%positive cases via ELISA NS1 antigen,16.7%via ELISA Ig G and 9.7%via ELISA Ig *** of the cases corresponded to DENV-3(77.5%).The use of RT-PcR performed better in primary infections(Pction of ELISA Ig M performed better in secondary infections(Pcombination of NS1 and Ig M performed better than the other assays in detecting primary(92.5%)and secondary infections(96.6%).The most frequent symptoms associated with fever were headaches,myalgias,and arthralgias across all ***:We report an important outbreak of dengue infection caused by DENV-3 in cajamarca,*** findings encourage the use of NS1 antigen and Ig M *** findings demonstrate an increasing expansion of DENV-3 in Peru and highlight the importance of molecular diagnosis and serotype characterization among the clinically defined dengue cases to strengthen the Peruvian epidemiological surveillance.

The current distribution of forest tree species is a result of natural or human mediated historical and contemporary processes. Knowledge of the spatial distribution of the diversity and divergence of populations is crucial for managing and conserving genetic resources in forest tree species. By combining tools from population genetics, landscape ecology and spatial statistics, landscape genetics thus represents a powerful method for evaluating the geographic patterns of genetic resources at the population level. In this study, we explore the possibility of combining genetic diversity data, spatial statistic tools and GIS technologies to map the genetic divergence and diversity of 31 castanea sativa populations collected in Spain, Italy, Greece, and Turkey. The IDW technique was used to interpolate the diversity values and divergence indices as expected hetereozygosity (He), allelic richness (Rs), private allelic richness (PRs), and membership values (Q) of each population to different clusters. Genetic diversity maps and a synthetic map of the spatial genetic structure of European chestnut populations were produced. Spatial coincidences between landscape elements and statistically significant genetic discontinuities between populations were investigated. Evidence is provided of the significance of cartographic outputs produced in the study and on their usefulness in managing genetic resources.

Soybean Seed Protein, Oil, Fatty Acids, and Isoflavones Altered by Potassium Fertilizer Rates in the Midsouth

Previous research has shown that the effect of potassium fertilizer on soybean ([Glycine max (L.) Merr.] seed composition (protein, oil, fatty acids, and isoflavones) is still largely unknown. Therefore, the objective of this research was to investigate the effects of potassium application on seed protein, oil, fatty acids, and isoflavones under Midsouth environmental conditions. A three-year experiment was conducted in two locations (Milan, TN and Jackson, TN). Potassium (K) rates were applied in the form of K2O at a rate of 0 (control, c), 45 (T1), 90 (T2), 134 (T3), and 179 (T4) kg·ha-1 in a randomized complete block design. The results showed that increasing the K application rate did not result in consistent effects on yield. However, increasing K application rate did increase protein, oleic acid, and individual and total isoflavone concentrations at both locations in 2008 and 2009. In Jackson in 2010, the increase of K rate did not change oleic acid, but resulted in an increase in glycitein and genistein isoflavone concentrations. In 2010, increasing K application rate increased protein concentrations, decreased individual and total isoflavones, and did not change oleic acid concentration at Milan. At the highest rate of K, 179 kg·ha-1, yield and some seed composition constituents were negatively impacted. Generally, K concentration in leaves at V5, R1, R3, and seed at harvest maturity stage (R8) increased with the increase of K rate applications. The research demonstrated that K application can alter seed composition, but this alteration depended on location, environmental stress factors, mainly heat and drought, K level in soil, and K application rate. Higher rates of K application may negatively impact seed composition constituents.

众所周知,在美国低收入地区长大的孩子面临着很多挑战。虽然有许多孩子表现出了惊人的天赋,甚至超过了富裕社区长大的孩子,但无数生活在低收人地区的孩子在发展上仍然是落后的。长期生活在贫困、住房不足和诸多社会问题地区的儿童,面临一系列负面成长发展的风险。在认知发展领域,他们多数学习成绩不佳,甚至会留级。他们也不太可能从高中毕业。未毕业的年轻人又面临更大的青少年犯罪风险和少女怀孕风险。在社会和情感发展方面,低收人地区长大的孩子更具有攻击性、冷漠、抑郁等人格障碍问题。
