Effects of serum containing natural cerebrolysin on glucose-regulated protein 78 and ccAAT enhancer-binding protein homologous protein expression in neuronal Pc12 cells following tunicamycin-induced endoplasmic reticulum stress

BAcKGRoUND： Glucose-regulated protein 78 （GRP78）, a marker of endoplasmic reticulum stress, can prolong cell survival. Alternatively, ccAAT enhancer-binding protein homologous protein （cHoP）, a transcription factor specific for endoplasmic reticulum stress, can cause cell cycle arrest and cell apoptosis. oBjEcTIVE： To study the protective effects of serum containing natural cerebrolysin on endoplasmic reticulum stress in tunicamycin-induced neuronal Pc12 cells, and analyze the influence on GRP78 and cHoP expressions. DESIGN, TIME AND SETTING： A parallel controlled study was performed at the Institute of Integrated Western and Traditional chinese Medicine, Shenzhen Hospital, Southern Medical University, between March 2006 and August 2008. MATERIALS： Adult Sprague-Dawley rats were perfused with natural cerebrolysin aqueous extract （0.185 g/kg/d） to produce serum containing natural cerebrolysin. Physiological saline was used to produce blank serum. Pc12 cell line was provided by Shanghai Institute of cell Biology, chinese Academy of Science. Tunicamycin was provided by Sigma （St. Louis, USA）, and natural cerebrolysin, containing ginseng, rhizoma gastrodiae, and gingko leaf （1：2：2）, by Shengzhen Institute of Integrated Western and Traditional chinese Medicine. METHoDS： Pc12 cells were treated with DMEM culture media containing 10% blank serum （normal control group）, tunicamycin （1 μg/mL; model group）, and 5%, 10%, and 15% serum containing natural cerebrolysin and tunicamycin （1 μ g/mL; low-, moderate-, and high-dose serum containing natural cerebrotysin groups）, for 2 hours. MAIN oUTcoME MEASURES： Pc12 cells were treated with tunicamycin for 48 hours after which apoptosis was measured using the TUNEL method to calculate apoptotic index. GRP78 expression was detected using immunocytochemistry. After 24 hours of treatment with tunicamycin, GRP78 and cHoP mRNA expressions were measured using RT-PcR. Investigation of HcAR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving the herd. In vitro administration of beta-hydroxybutyric acid(BHBA;the primary blood ketone) has inhibitory effects on the function of bovine leukocytes. BHBA is a ligand of HcAR2 and the activation of these receptors promotes an anti-inflammatory response which may be related with immunosuppression observed in transition dairy cattle. The objective of this study was to identify and test antagonists for HcAR2 in bovine immune cells cultured with *** We observed expression of HcAR2 at the protein level within lymphocytes, monocytes, and granulocytes. The proportion of cells expressing HcAR2 tended to be greater in mid-lactation compared to early lactation cows;the increase was a result of increased proportion of T and B cells expressing HcAR2. Stimulation of HcAR2 with niacin or BHBA promoted ca^(2+) mobilization in neutrophils and mononuclear cells. Mononuclear cells treated with BHBA had diminished intracellular ca^(2+) responses when HcAR2 was knocked down by si RNA silencing, indicating ca^(2+) mobilization was mediated by HcAR2 signaling. Two candidate antagonists for HcAR2, synthesized from niacin(NA-1 and NA-5), were tested;monocytes and neutrophils pre-treated with NA-1 and NA-5 had reduced ca^(2+) mobilization after incubation with BHBA. Furthermore, NA-5 but not NA-1 prevented BHBA-associated reductions in cyclic *** We demonstrated that HcAR2 is present on bovine leukocytes and has greater expression later in lactation. We confirmed that BHBA and niacin derived HcAR2 antagonists alter bovine leukocyte activity. our results demonstrate that both BHBA and niacin affect bovine leukocyte ca^(2+) mobilization in a HcAR2-dependent manner.

Laser 3D printing,also known as laser additive manufacturing(LAM),is favored for its ability to form bulk metallic glass(BMG)and its composite materials(BMGcs)with freeform *** this work,two different kinds of Fe_(41)co_(7)cr_(15)Mo_(14)c_(15)B_(6)Y_(2)amorphous coatings(A and B)were prepared by using LAM technology under air-and water-cooled conditions,respectively;meanwhile,to reduce the cracks generated due to the residual thermal stresses,coating c obtained by air-sweep annealing of B with a low energy-density *** morphology and amorphous content and microstructure of the coatings were investigated,the results show many cracks in coating B deposited under water-cooled conditions,and its microstructure shows an amorphous-crystal-nanocrystalline mixed *** was suppressed in coating c,obtained by air-sweep annealing based on coating B,but the amorphous content was reduced from 32.6 to 13.4%.And the hardness and corrosion resistance of the coating will increase with the increase in the amorphous ***,the internal friction behavior of a BMGcs was prepared on the basis of the process of sample c is compared with that of as-cast amorphous *** results show that the low temperature internal friction behavior of BMGcs is affected by the defects produced during printing,and the high temperature internal friction behavior is affected by the precipitated hard phase.

Granger causal representation learning for groups of time series

Discovering causality from multivariate time series is an important but challenging *** existing methods focus on estimating the Granger causal structures among multivariate time series,while ignoring the prior knowledge of these time series, e.g., the group of the time series. Focusing on discovering the Granger causal structures among groups of time series, we propose a Granger causal representation learning method to solve this problem. First, we use the multiset canonical correlation analysis method to learn the Granger causal representation of each group of time series. Then, we model the Granger causal relationships among the learned Granger causal representations using a recurrent neural network with temporal information. Finally, we formulate the above two stages into one unified optimization problem, which is efficiently solved using the augmented Lagrangian method. We conduct extensive experiments on synthetic and real-world datasets to validate the correctness and effectiveness of the proposed method.

BAcKGRoUND cheilectomy of the 1^(st)metatarsophalangeal joint(MTPj)is one of the most common procedures for the management of hallux ***,there is no consensus regarding outcomes following minimally invasive dorsal cheilectomy(MIDc)for the management of hallux *** To evaluate outcomes following MIDc for the management of hallux *** During November 2023,the PubMed,EMBASE and cochrane Library databases were systematically reviewed to identify clinical studies examining outcomes following MIDc for the management of hallux *** Six studies were *** total,348 patients(370 feet)underwent MIDc for hallux rigidus at a weighted mean follow-up of 37.9±16.5 *** distribution of patients by coughlin and Shurna's classification was recorded in 4 studies as follows:Ⅰ(58 patients,27.1%),Ⅱ(112 patients,52.3%),Ⅲ(44 patients,20.6%).Three studies performed an additional 1^(st)MTPj arthroscopy and debridement following *** intra-articular bone debris was observed in 100%of patients in 1 *** weighted mean American orthopedic foot and ankle society score improved from a preoperative score of 68.9±3.2 to a postoperative score of *** complication rate was 8.4%,the most common of which was persistent joint pain and ***-two failures(8.7%)were ***-three secondary procedures(8.9%)were performed at a weighted mean time of 8.6±3.2 months following the index *** This systematic review demonstrated improvements in subjective clinical outcomes together with a moderate complication rate following MIDc for the management of hallux rigidus at short-term follow-up.A moderate reoperation rate at short-term follow-up was *** marked heterogeneity between included studies and paucity of high quality comparative studies limits the generation of any robust conclusions.

Laminar Flame Structure-Dependent Exergy Destruction Behavior at Auto-Ignition Time Scale:A case Study of Dimethyl Ether(DME)

Hybrid deflagration/auto-ignition flame structures coexist in the combustion of advanced *** exergy destruction caused by different irreversible processes under varied flame regimes is thus important for understanding engine *** this study,the flame propagation modes for the premixed DME/air mixtures are numerically investigated under engine-relevant *** entropy generation and exergy destruction characteristics are compared under different flame *** reveal that as the typical premixed flame transition towards auto-ignition front,the exergy destruction from heat conduction and species mass diffusion gradually vanish and are dominated by chemical *** distributions of temperature and species mole fraction in the flame domain are analyzed to clarify the exergy destruction behaviors caused by heat conduction and mass ***,by dividing the DME oxidation process into four stages,the main reaction channels that contribute to the increase in exergy destruction from chemical reaction have been *** is found that the production and consumption of cH_(2)o and Ho_(2) radical dominate the exergy destruction behavior during DME *** this basis,the kinetic mechanism of low-temperature chemistry causing greater exergy destruction is ***,low-temperature chemistry leads to significant exergy destruction due to(a)the large irreversibility of itself and(b)(mainly)the enhancement of H_(2)o_(2)loop reactions by low-temperature reaction *** the reduction of combustion irreversibility is promising to be achieved by reasonably regulating the fuel oxidation path.

Slits have been widely used in laser-plasma interactions as plasma optical components for generating high-harmonic light and controlling laser-driven particle ***,we propose and demonstrate that periodic thin slits can be regarded as a new breed of optical elements for efficient focusing and guiding of intense laser *** fundamental physics of intense laser interaction with thin slits is studied,and it is revealed that relativistic effects can lead to enhanced laser focusing far beyond the pure diffractive focusing *** addition,the interaction of an intense laser pulse with periodic thin slits makes it feasible to achieve multifold enhancement in both laser intensity and energy transfer efficiency compared with conventional *** results provide a novel method for manipulating ultra-intense laser pulses and should be of interest for many laser-based applications.

In situ direct reprogramming of astrocytes to neurons via polypyrimidine tract-binding protein 1 knockdown in a mouse model of ischemic stroke

In situ direct reprogramming technology can directly convert endogenous glial cells into functional neurons in vivo for central nervous system repair. Polypyrimidine tract-binding protein 1(PTB) knockdown has been shown to reprogram astrocytes to functional neurons in situ. In this study, we used AAV-PHP.e B-GFAP-sh PTB to knockdown PTB in a mouse model of ischemic stroke induced by endothelin-1, and investigated the effects of GFAP-sh PTB-mediated direct reprogramming to neurons. our results showed that in the mouse model of ischemic stroke, PTB knockdown effectively reprogrammed GFAP-positive cells to neurons in ischemic foci, restored neural tissue structure, reduced inflammatory response, and improved behavioral function. These findings validate the effectiveness of in situ transdifferentiation of astrocytes, and suggest that the approach may be a promising strategy for stroke treatment.
