Early tacrolimus exposure does not impact long-term outcomes after liver transplantation

The Early Evolutionary History and Adaptive Radiation of the Pterosauria

Phylogenetic analyses of early pterosaur species are relatively new and contradictory. However, they imply a similar evolutionary history for early pterosaurs： a large divergence of all major early groups of pterosaurs before the first named pterosaur species appeared in the fossil record. This large, unsampled divergence may be a record of the initial adaptive radiation of the pterosaurs, but a recent comprehensive phylogenetic analysis reconstructs named Triassic pterosaur species grouped together in a monophyletic clade with all other major groups diverging later. Ten patterns that characterize adaptive radiations put forward by Gavrilets and Losos （2009） are assessed for congruence with the comprehensive phylogeny and two previous phylogenies to determine which are consistent with our notions of an adaptive radiation. Of the eight patterns that can be applied to pterosaurs, only six vary between pterosaur phylogenies. Of these six patterns, all apply to the three phylogenies with varying amounts of support. For many patterns, however, the comprehensive phylogeny has a greater magnitude, shorter time span, or is the only topology within which they can be assessed. Pterosaurs are characterized by an adaptive radiation no matter which phylogeny is considered, but congruence with the fossil record increases the support and information content of a phylogeny by reducing the time and area within which to reconstruct evolutionary history.

Phylogenetic framework for coevolutionary studies： a compass for exploring jungles of tangled trees

Phylogenetics is used to detect past evolutionary events, from how species originated to how their ecological interactions with other species arose, which can mirror cophylogenetic patterns. Cophylogenetic reconstructions uncover past ecological relationships between taxa through inferred coevolutionary events on trees, for example, codivergence, duplication, host-switching, and loss. These events can be detected by cophylogenetic analyses based on nodes and the length and branching pattern of the phylogenetic trees of symbiotic associations, for example, host-parasite. In the past 2 decades, algorithms have been developed for cophylogetenic analyses and implemented in different software, for ex ample, statistical congruence index and event-based methods. Based on the combination of these approaches, it is possible to integrate temporal information into cophylogenetical inference, such as es- timates of lineage divergence times between 2 taxa, for example, hosts and parasites. Additionally, the advances in phylogenetic biogeography applying methods based on parametric process models and combined Bayesian approaches, can be useful for interpreting coevolutionary histories in a scenario of biogeographical area connectivity through time. This article briefly reviews the basics of parasitology and provides an overview of software packages in cophylogenetic methods. Thus, the objective here is to present a phylogenetic framework for coevolutionary studies, with special emphasis on groups of parasitic organisms. Researchers wishing to undertake phylogeny-based coevolutionary studies can use this review as a ＂compass＂ when ＂walking＂ through jungles of tangled phylogenetic trees.

Ascites, refractory ascites and hyponatremia in cirrhosis

Ascites is the most common complication related to cirrhosis and is associated with increased morbidity and *** is a consequence of the loss of compensatory mechanisms to maintain the overall effective arterial blood volume due to worsening splanchnic arterial vasodilation as a result of clinically significant portal *** order to maintain effective arterial blood volume,vasoconstrictor and antinatriuretic pathways are activated,which increase overall sodiumand fluid *** a result of progressive splanchnic arterial vasodilation,intestinal capillary pressure increases and results in the formation of protein-poor fluid within the abdominal cavity due to increased capillary permeability from the hepatic sinusoidal *** some patients,the fluid can translocate across diaphragmatic fenestrations into the pleural space,leading to hepatic *** addition,infectious complications such as spontaneous bacterial peritonitis can ***,as the liver disease progresses related to higher portal pressures,loss of a compensatory cardiac output and further splanchnic vasodilation,kidney function becomes compromised fromworsening renal vasoconstriction as well as the development of impaired solute-free water excretion and severe sodium *** then translate into significant clinical complications,such as refractory ascites,hepatorenal syndrome and hyponatremia,and all are linked to increased ***,liver transplantation is the only curative option for this spectrumof clinical manifestations but ongoing research has led to further insight on alternative *** review will further explore the current understanding on the pathophysiology andmanagement of ascites as well as expand on two advanced clinical consequences of advanced liver disease,refractory ascites and hyponatremia.

Endotoxin-induced alterations of adipose tissue function:a pathway to bovine metabolic stress

During the periparturient period, dairy cows exhibit negative energy balance due to limited appetite and increased energy requirements for lactogenesis. The delicate equilibrium between energy availability and expenditure puts cows in a state of metabolic stress characterized by excessive lipolysis in white adipose tissues(AT), increased production of reactive oxygen species, and immune cell dysfunction. Metabolic stress, especially in AT, increases the risk for metabolic and inflammatory diseases. Around parturition, cows are also susceptible to endotoxemia. Bacterial-derived toxins cause endotoxemia by promoting inflammatory processes and immune cell infiltration in different organs and systems while impacting metabolic function by altering lipolysis, mitochondrial activity, and insulin sensitivity. In dairy cows, endotoxins enter the bloodstream after overcoming the defense mechanisms of the epithelial barriers, particularly during common periparturient conditions such as mastitis, metritis, and pneumonia, or after abrupt changes in the gut microbiome. In the bovine AT, endotoxins induce a pro-inflammatory response and stimulate lipolysis in AT, leading to the release of free fatty acids into the bloodstream. When excessive and protracted, endotoxin-induced lipolysis can impair adipocyte's insulin signaling pathways and lipid synthesis. Endotoxin exposure can also induce oxidative stress in AT through the production of reactive oxygen species by inflammatory cells and other cellular components. This review provides insights into endotoxins' impact on AT function, highlighting the gaps in our knowledge of the mechanisms underlying AT dysfunction, its connection with periparturient cows' disease risk, and the need to develop effective interventions to prevent and treat endotoxemia-related inflammatory conditions in dairy cattle.
