Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tNGCs). however, the use of tNGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tNGCs and compared the various features of these improved silk^(+) tNGCs with the tubes without holes(silk^(–) tNGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tNGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tNGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tNGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tNGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. We present the design,fabrication and characterization of hydraulically-tunable hyperchromatic lenses for two-dimensional(2D)spectrally-resolved spectral *** hyperchromatic lenses,consisting of a positive diffractive lens and a tunable concave lens,are designed to have a large longitudinal chromatic dispersion and the images of different wavelengths from each other.2D objects of different wavelengths can consequently be imaged using the tunability of the lens *** hyperchromatic lens concepts are demonstrated and their spectral characteristics as well as their functionality in spectral imaging applications are shown.

holocene climate change in the Central Tibetan Plateau inferred by lacustrine sediment geochemical records

Multi-proxies of lacustrine sediments, such as total carbon (TC), total organic carbon (TOC), total inorganic carbon (TIC), total nitrogen (TN), total sulfur (TS), hydrogen index (hI), oxygen index (OI) and stable carbon isotopic composition of organic matter (δ^(13)C_(org)), were analyzed using a 7.3 m core from Zige Tangco. The source of the organic matter in the sediment was mainly from autochthonous phyto-plankton, therefore the significances of proxies can be interpreted as that high TOC, TOC/TS, hI and δ^(13)C_(org) values, low TC, TIC values corresponded to warm and wet climatic condition, and vice versa. The process of climatic development in the Zige Tangco region was hence recovered. During the early and Mid-holocene, the climate was warm and wet and intensive cold events occurred during the periods of 8600 to 8400 cal a BP and 7400 to 7000 cal a BP. In the second half of holocene, the climate became cold and dry gradually. The palaeoclimatic process during holocene in Zige Tangco region matched well with that in Co Ngoin region which is ca 40 km to the south-east. Therefore this palaeoclimatic process represents the holocene climatic feature in the Central Tibetan Plateau which has the same pattern in the Northern Tibetan Plateau, but the time and duration of some climatic events might be different. We can conclude that in holocene solar insolation controlled the climatic pattern on the central Tibetan Plateau.

live-Cell Imaging of Dual-labeled Golgi Stacks in Tobacco BY-2 Cells Reveals Similar Behaviors for Different Cisternae during Movement and Brefeldin A Treatment

In plant cells, the Golgi apparatus consists of numerous stacks that, in turn, are composed of several flattened cisternae with a clear cis-to-trans polarity. During normal functioning within living cells, this unusual organelle displays a wide range of dynamic behaviors such as whole stack motility, constant membrane flux through the cisternae, and Golgi enzyme recycling through the er. In order to further investigate various aspects of Golgi stack dynamics and integrity, we co-expressed pairs of established Golgi markers in tobacco BY-2 cells to distinguish sub-compartments of the Golgi during monensin treatments, movement, and brefeldin A （BFA）-induced disassembly. A combination of cis and trans markers revealed that Golgi stacks remain intact as they move through the cytoplasm. The Golgi stack orientation during these movements showed a slight preference for the cis side moving ahead, but trans cisternae were also found at the leading edge. During BFA treatments, the different sub-compartments of about half of the observed stacks fused with the er sequentially; however, no consistent order could be detected. In contrast, the ionophore monensin resulted in swelling of trans cisternae while medial and particularly cis cisternae were mostly unaffected. Our results thus demonstrate a re- markable equivalence of the different cisternae with respect to movement and BFA-induced fusion with the er. In addi- tion, we propose that a combination of dual-label fluorescence microscopy and drug treatments can provide a simple alternative approach to the determination of protein localization to specific Golgi sub-compartments.

It is well acknowledged that the equation of radiative transfer(erT)provides an accurate prediction of light propagation in biological tissues,while the diffusion approximation(DA)is of limited accuracy for the transport ***,erT-based reconstruction codes require much longer computation times as compared to DA-based reconstruction *** introduce here a computationally efficient algorithm,called a diffusion–transport hybrid solver,that makes use of the DA-or low-order erT-based inverse solution as an initial guess for the full erT-based reconstruction *** evaluate the performance of this hybrid method,we present extensive studies involving numerical tissue phantoms and experimental *** a result,we show that the hybrid method reduces the reconstruction time by a factor of up to 23,depending on the physical character of the problem.

Structural Finite Element Software Coupling Using Adapter Elements

This paper describes a versatile and computationally efficient method for coupling several finite element analysis(FEA)programs together so that the unique modeling and analysis capabilities of each code can be utilized simultaneously to simulate the static or dynamic response of a complete numerical *** arbitrary number of finite element analysis software packages can be coupled by adding two special types of elements,namely generic and adapter elements,to each of the finite element applications using their programming *** elements are inserted at the interfaces between the different sub-domains of the complete system modeled by each finite element analysis software *** of data between the coupled FEA codes is accomplished in a modular and synchronized manner using OpenFresco(Opensource Framework for Experimental Setup and Control).OpenFresco is an objectoriented,environment independent software framework initially developed for hybrid simulation in which certain aspects of a complete structure are simulated numerically and other aspects are simultaneously tested *** important practical advantage of this coupled analysis approach is that all of the connected FEA codes run concurrently and continuously,decreasing analysis time consumption by an order of magnitude or more compared to more traditional approaches that shut down and restart the coupled analysis codes at each integration time *** implementation and accuracy of this approach to FE software coupling are demonstrated using dynamic analyses of three simple structural models from the field of earthquake engineering.
