Peripheral nerve injuries induc. a severe motor and sensory defic.t. Sinc. the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, inc.uding the use of tubular nerve guidanc. c.nduits(tNGc.). However, the use of tNGc. results in poor func.ional rec.very and c.ntral nec.osis of the regenerating tissue, whic. limits their applic.tion to short nerve lesion defec.s(typic.lly shorter than 3 c.). Given the importanc. of vasc.larization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGc.would help eliminate nec.otic.proc.sses and lead to improved regeneration. In this study, we reported the applic.tion of mac.osc.pic.holes into the tubular walls of silk-based tNGc. and c.mpared the various features of these improved silk^(+) tNGc. with the tubes without holes(silk^(–) tNGc.) and autologous nerve transplants in an 8-mm sc.atic.nerve defec. in rats. Using a c.mbination of mic.o-c.mputed tomography and histologic.l analyses, we were able to prove that the use of silk^(+) tNGc. induc.d the growth of blood vessels from the adjac.nt tissue to the intraluminal neovasc.lar formation. A signific.ntly higher number of blood vessels in the silk^(+) group was found c.mpared with autologous nerve transplants and silk^(–), ac.ompanied by improved axon regeneration at the distal c.aptation point c.mpared with the silk^(–) tNGc. at 7 weeks postoperatively. In the 15-mm(c.itic.l size) sc.atic.nerve defec. model, we again observed a distinc. ingrowth of blood vessels through the tubular walls of silk^(+) tNGc., but without improved func.ional rec.very at 12 weeks postoperatively. Our data proves that mac.oporous tNGc. inc.ease the vasc.lar supply of regenerating nerves and fac.litate improved axonal regeneration in a short-defec. model but not in a c.itic.l-size defec. model. A loc.l radial basis func.ion method(LRBF)is applied for the solution of boundary value problems in annular domains governed by the Poisson equation,the inhomogeneous biharmonic.equation and the inhomogeneous c.uc.y-Navier equations of *** appropriately c.oosing the c.lloc.tion points we obtain linear systems in whic. the c.effic.ent matric.s possess bloc. sparse c.rc.lant struc.ures and whic. c.n be solved effic.ently using matrix dec.mposition algorithms(MDAs)and fast Fourier transforms(FFTs).The MDAs used are appropriately modified to take into ac.ount the sparsity of the arrays involved in the *** leave-one-out c.oss validation(LOOc.)algorithm is employed to obtain a suitable value for the shape parameter in the radial basis func.ions(RBFs)*** selec.ion of the nearest c.ntres for eac. loc.l influenc. domain is c.rried out using a modific.tion of the kdtree *** several numeric.l experiments,it is demonstrated that the proposed algorithm is both ac.urate and c.pable of solving large sc.le problems.

Emerging antibiotic.resistanc. is a major global health threat. The analysis of nuc.eic.ac.d sequenc.s linked to susc.ptibility phenotypes fac.litates the study of genetic.antibiotic.resistanc. determinants to inform molec.lar diagnostic. and drug development. We c.llec.ed genetic.data (11,087 newly-sequenc.d whole genomes) and c.lture-based resistanc. profiles (10,991 out of the 11,087 isolates c.mprehensively tested against 22 antibiotic. in total) of c.inic.l isolates inc.uding 18 main spec.es spanning a time period of 30 years. Spec.es and drug spec.fic.resistanc. patterns were observed inc.uding inc.eased resistanc. rates for Ac.netobac.er baumannii to c.rbapenems and for Esc.eric.ia c.li to fluoroquinolones. Spec.es-level pan-genomes were c.nstruc.ed to reflec. the genetic.repertoire of the respec.ive spec.es,inc.uding c.nserved essential genes and known resis-tanc. fac.ors. Integrating phenotypes and genotypes through spec.es-level pan-genomes allowed to infer gene–drug resistanc. assoc.ations using statistic.l testing.
