The transforming growth factor-β(TGF-β)family controls embryogenesis,stem cell differentiation,and tissue ***,how post-translation modifications contribute to fine-tuning of TGF-βfamily signaling responses is not well ***(I)-Smads can antagonize TGF-β/Smad signaling by recruiting Smurf E3 ubiquitin ligases to target the active TGF-βreceptor for proteasomal degradation.A proteomic interaction screen identified Vpr binding protein(VprBP)as novel binding partner of ***-expression studies revealed that VprBP negatively controls Smad2 phosphorylation,Smad2–Smad4 interaction,as well as TGF-βtarget gene *** was found to promote Smad7–Smurf1–TβRI complex formation and induce proteasomal degradation of TGF-βtype I receptor(TβRI).Moreover,VprBP appears to stabilize Smurf1 by suppressing Smurf1 *** multiple adult and mouse embryonic stem cells,depletion of VprBP promotes TGF-βor Activin-induced *** the mouse embryo VprBP expression negatively correlates with mesoderm marker expression,and VprBP attenuated mesoderm induction during zebrafish *** findings thereby uncover a novel regulatory mechanism by which Smurf1 controls the TGF-βand Activin cascade and identify VprBP as a critical determinant of embryonic mesoderm induction.

Convolutional neural network-assisted recognition of nanoscale L1_(2) ordered structures in face-centred cubic alloys

Nanoscale L12-type ordered structures are widely used in face-centered cubic(FCC)alloys to exploit their hardening capacity and thereby improve mechanical *** fine-scale particles are typically fully coherent with matrix with the same atomic configuration disregarding chemical species,which makes them challenging to be *** distribution maps(SDMs)are used to probe local order by interrogating the three-dimensional(3D)distribution of atoms within reconstructed atom probe tomography(APT)***,it is almost impossible to manually analyze the complete point cloud(>10 million)in search for the partial crystallographic information retained within the ***,we proposed an intelligent L1_(2)-ordered structure recognition method based on convolutional neural networks(CNNs).The SDMs of a simulated L1_(2)-ordered structure and the FCC matrix were firstly *** simulated images combined with a small amount of experimental data were used to train a CNN-based L1_(2)-ordered structure recognition ***,the approach was successfully applied to reveal the 3D distribution of L1_(2)–typeδ′–Al3(LiMg)nanoparticles with an average radius of 2.54 nm in a FCC Al-Li-Mg *** minimum radius of detectable nanodomain is even down to 5Å.The proposed CNN-APT method is promising to be extended to recognize other nanoscale ordered structures and even more-challenging short-range ordered phenomena in the near future.

Importance of LV Venting after ECMO Implantation in Post-Cardiotomy Syndrome: A Case Report

Prolonged cardiorespiratory support can be achieved with ECMO that may provide time for myocardial recovery, prevent multiorgan dysfunction and reduce mortality. Left ventricle (LV) distension can worsen already distended and hypocontractile heart. Early recognition and aggressive management of LV distension are essential for the treatment of patients with low cardiac output. The case report presented intends to show advantages of left ventricular venting on ECMO after post-cardiotomy shock. With direct flow measurements on bypass-grafts before and after the vent implantation, it was possible to clearly demonstrate the importance of venting for myocardial perfusion.

Mineral and bone disorder(MBD)in chronic kidney disease(CKD)is tightly linked to cardiovascular disease(CVD).In this study,we aimed to compare the prognostic value of nine MBD biomarkers to determine those associated best with adverse cardiovascular(CV)outcomes and *** 5217 participants of the German CKD(GCKD)study enrolled with an estimated glomerular filtration rate(eG FR)between 30–60 mL·min-1 per 1.73 m2 or overt proteinuria,serum osteoprotegerin(OPG),C-terminal fibroblast growth factor-23(FGF23),intact parathyroid hormone(iP TH),bone alkaline phosphatase(BAP),cross-linked C-telopeptide of type 1 collagen(CTX1),procollagen 1intact N-terminal propeptide(P1NP),phosphate,calcium,and 25-OH vitamin D were measured at *** with missing values among these parameters(n=971)were excluded,leaving a total of 4246 participants for *** a median follow-up of6.5 years,387 non-CV deaths,173 CV deaths,645 nonfatal major adverse CV events(MACEs)and 368 hospitalizations for congestive heart failure(CHF)were *** and FGF23 were associated with all outcomes,with the highest hazard ratios(HRs)for *** the final Cox regression model,adjusted for CV risk factors,including kidney function and all other investigated biomarkers,each standard deviation increase in OPG was associated with non-CV death(HR 1.76,95%CI:1.35–2.30),CV death(HR 2.18,95%CI:1.50–3.16),MACE(HR 1.38,95%CI:1.12–1.71)and hospitalization for CHF(HR 2.05,95%CI:1.56–2.69).Out of the nine biomarkers examined,stratification based on serum OPG best identified the CKD patients who were at the highest risk for any adverse CV outcome and mortality.
