Endotoxins(also known as lipopolysaccharides(LPS)) are undesirable by-products of recombinant proteins,purified from Escherichia coli.LPS can be considered stable under a wide range of temperature and pH,making their removal one of the most difficult tasks in downstream processes during protein purification.The inherent toxicity of LPS makes their removal an important step for the application of these proteins in several biological assays and for a safe parenteral administration.Immobilized metal affinity chromatography(IMAC) enables the affinity interactions between the metal ions(immobilized on the support through the chelating compound) and the target molecules,thus enabling high-efficiency separation of the target molecules from other components present in a mixture.Affinity chromatography is applied with Ca2+-iminodiacetic acid(IDA) to remove most of the LPS contaminants from the end product(more than90%).In this study,the adsorption of LPS on an IDA-Ca2+ was investigated.The adsorption Freundlich isotherm of LPS-IDA-Ca2+ provides a theoretical basis for LPS removal.It was found that LPS is bound mainly by interactions between the phosphate group in LPS and Ca2+ ligands on the beads.The factors such as pH(4.0 or 5.5) and ionic strength(1.0 mol/L) are essential to obtain effective removal of LPS for contaminant levels between endotoxin' concentration values less than100 EU/mL and 100 000 EU/mL.This new protocol represents a substantial advantage in time,effort,and production costs.

Induced pluripotent stem cells, a giant leap for mankind therapeutic applications

Induced pluripotent stem cells(iPSC)technology has propelled the field of stem cells biology,providing new cells to explore the molecular mechanisms of pluripotency,cancer biology and aging.A major advantage of human iPSC,compared to the pluripotent embryonic stem cells,is that they can be generated from virtually any embryonic or adult somatic cell type without destruction of human blastocysts.In addition,iPSC can be generated from somatic cells harvested from normal individuals or patients,and used as a cellular tool to unravel mechanisms of human development and to model diseases in a manner not possible before.Besides these fundamental aspects of human biology and physiology that are revealed using iPSC or iPSC-derived cells,these cells hold an immense potential for cell-based therapies,and for the discovery of new or personalized pharmacological treatments for many disorders.Here,we review some of the current challenges and concerns about iPSC technology.We introduce the potential held by iPSC for research and development of novel health-related applications.We briefly present the efforts made by the scientific and clinical communities to create the necessary guidelines and regulations to achieve the highest quality standards in the procedures for iPSC generation,characterization and long-term preservation.Finally,we present some of the audacious and pioneer clinical trials in progress with iPSC-derived cells.

Biliary stone causing afferent loop syndrome and pancreatitis

We report the case of an 84-year-old female who had a partial gastrectomy with Billroth-II anastomosis 24 years ago for a benign peptic ulcer who now presented an acute pancreatitis secondary to an afferent loop syndrome. The syndrome was caused by a gallstone that migrated through a cholecystoenteric fi stula. This is the fi rst description in the literature of a biliary stone causing afferent loop syndrome.

Innate immunity and hepatocarcinoma:Can toll-likereceptors open the door to oncogenesis?

Hepatocarcinoma(HCC) is a highly prevalent cancer worldwide and its inflammatory background was established long ago.Recent studies have shown that innate immunity is closely related to the HCC carcinogenesis.An effective innate immunity response relies on the tolllike receptors(TLR) found in several different liver cells which,through different ligands and many signaling pathways can elicit,not only a pro-inflammatory but also an oncogenic or anti-oncogenic response.Our aim was to study the role of TLRs in the liver oncogenesis and as a consequence their value as potential therapeutic targets.We performed a systematic review of PubMed searching for original articles studying the relationship between HCC and TLRs until March 2015.TLR2 appears to be a fundamental stress-sensor as its absence reveals an augmented tendency to accumulate DNAdamages and to cell survival.However,pathways are still not fully understood as TLR2 up-regulation was also associated to enhanced tumorigenesis.TLR3 has a wellknown protective role influencing crucial processes like angiogenesis,cell growth or proliferation.TLR4 works as an interesting epithelial-mesenchymal transition's inducer and a promoter of cell survival probably inducing HCC carcinogenesis even though an anti-cancer role has already been observed.TLR9's influence on carcinogenesis is also controversial and despite a potential anticancer capacity,a pro-tumorigenic role is more likely.Genetic polymorphisms in some TLRs have been found and its influence on the risk of HCC has been reported.As therapeutic targets,TLRs are already in use and have a great potential.In conclusion,TLRs have been shown to be an interesting influence on the HCCs microenvironment,with TLR3 clearly determining an antitumour influence.TLR4 and TLR9 are considered to have a positive relationship with tumour development even though,in each of them anti-tumorigenic signals have been described.TLR2 presents a more ambiguous role,possibly depending on the stage of the inflammationHCC axis.

Influence of fire on woody vegetation of savanna and forest formations in the Cerrado biome

Wildfires have environmental,economic,and social impacts,and can shape the landscape and benefit ecosystems such as the Cerrado.This study evaluated the diversity,similarity,and floristic and structural differences of woody savanna and forest formations of the Cerrado,when affected and not affected by fire.Twenty-eight 25-400m2 plots were randomly allocated and divided into Burnt Cerrado and Unburnt Cerrado,Burnt Forest,and Unburnt Forest,and divided into three levels of inclusion according to diameter class(smaller than 2 cm,2-5 cm and larger than5 cm).Species were identified,DBH and height measured,and phyto sociological parameters such as volume,diversity,and floristic similarity evaluated.Burnt Cerrado had lower diversity,density,and dominance at all inclusion levels compared to the Unburnt Cerrado,and showed similarities between treatments at the inclusion levels.Burnt Forest had smaller differences in diversity,density,and dominance than Unburnt Forest.Forest formation was similar at level 1 of inclusion and in the total area.However,it was dissimilar at lower levels.The lower density and diversity of species at the lowest levels of inclusion was associated with mortality from fire.

Investigating local corrosion processes of magnesium alloys with scanning probe electrochemical techniques: A review

The study of corrosion of magnesium and its alloys has emerged a hot topic in the applications of lightweight structural materials. The inherently high electrochemical activity of bare magnesium surfaces still lacks a convincing mechanism to describe the observed experimental characteristics, and it has prompted the development of various types of protective coatings with the aim of slowing metal dissolution. In recent years, new instruments and techniques have been developed to study with spatial resolution the local corrosion processes that occur in metallic materials in general, and for magnesium and its alloys in particular, both for bare surfaces and coated. Scanning microelectrochemical techniques, such as local electrochemical impedance spectroscopy(LEIS), scanning electrochemical microscopy(SECM), scanning vibrating electrode technique(SVET), scanning ion-selective electrode technique(SIET) and scanning Kelvin probe(SKP) can provide information about the local electrochemical activity of metallic surfaces. In the present work, the applications of these techniques in corrosion studies of magnesium and its alloys are reviewed. Assessment of corrosion mechanisms, barrier properties of conventional coatings and active corrosion behavior of self-healing coatings are examined. Limitations and future developments in this area are discussed.

Objective:To investigate the morphological structure of ovarian follicular cells and biochemical parameters of both ovaries and fat bodies(sites of vitellogenesis)from Rhodnius(R.)prolixus infected with Trypanosoma(T.)rangeli.Methods:Adult virgin females of R.prolixus were fed upon a membrane apparatus containing heat-inactivated citrated rabbit blood and a suspension of T.rangeli epimastigotes(Macias strain).Females from the control group and all the males received parasite-free blood.Transmission electron microscopy was used to reveal the morphological aspects of ovarian follicle cells in both control and parasite-infected groups.Protein profile,proteolytic activities and Western blotting analyses were performed in either ovary or fat body samples of control and parasite-infected groups.Results:According to the ultrastructural data,T.rangeli infection elicited a degeneration process in the ovarian follicular cells of R.prolixus.Proteolytic assays indicated a reduction in the activity of aspartic peptidases in the ovary and fat body from parasite-infected group,while a significant increase in the cysteine peptidase activity was measured in both insect organs.Additionally,immunoblotting revealed that vitellogenin was overexpressed in the ovary of parasite-infected insects.Conclusions:T.rangeli infection seems to elicit an early programmed cell death in the ovarian follicle cells as well as induces the modulation on the activities of different peptidase classes in either ovaries or fat bodies and the overexpression of the vitellogenin in the ovary of R.prolixus.

The aim of this review was to verify the risks of drug-nutrient interactions in the elderly. This is an integrative literature review, with a descriptive approach, carried out through research in indexed databases, legislation and pharmacopoeias. Data collection took place in the year 2022, through the analysis of literature collected of the last 10 years collected. The data showed that many of the continuous drugs use by the elderly can lead to interactions of different orders with nutrients, increasing the risk of plasmatic fluctuation as grapefruit, orange, lemon and lime interact with systemic arterial hypertension, increasing the risk of drug intoxication in the elderly. The action of the health team is essential, through the careful evaluation of the administered drugs, diet therapy and the interaction between them, to benefit the elderly with better use of the therapy and improvement of nutritional conditions.

Histological Study of the Male Internal Genital Tract of Dwarf Caiman (Paleosuchus palpebrosus Cuvier, 1807)

Brazil has six species of crocodiles belonging to the subfamily Caimaninae,one of them being Paleosuchus palpebrosus(Cuvier,1807)(dwarf caiman).It is considered one of the smallest crocodilian species,and is sensitive to environmental changes.The anatomical and histological descriptions of the reproductive tract of reptiles are scarce,so it is important to study the morphology of the genital tract of caimans for a better understanding of the reproduction of these species.We performed the histological analysis of the testis,epididymis,and deferent duct of the dwarf caiman Paleosuchus palpebrosus.Structures of the genital tract were collected from euthanized animals.Testes,epididymis,and deferent ducts were separated and prepared according to routine histological techniques for morphological characterization.The testis is covered by a capsule of fibromuscular connective tissue.Spermatogonia are observed as spherical cells with round nuclei and loose chromatin,and primary spermatocytes show dark cytoplasm,round nuclei,and pale chromatin.Spermatids are seen as cylindrical or filamentous cells with nuclei that may be highly condensed,ranging from round to oval.The epididymis is observed as a coiled duct lined by a tall cuboidal epithelium with stereocilia;the deferent duct shows ciliated pseudostratified cylindrical epithelium.The adrenal gland is found in the dorsomedial portion of the testis.The histological structure of the reproductive tract of Paleosuchus palpebrosus is similar to that of other reptiles’species.
