Deployment of Pre-Industrial Autonomous Microbe Sensor in Saudi Arabia’s Injection Seawater System

Microbial growth in water injection systems can lead to many problems, including biofouling, water quality deterioration, injectivity loss, microbial corrosion, and reservoir formation damage. Monitoring of microbial activities is required in any mitigation strategy, enabling operators to apply and adjust countermeasures properly and in due time. In this study, the pre-industrial autonomous microbe sensor (AMS) was constructed with technical improvements from the prototype for increased sensitivity, durability, robustness, and maintainability. The pre-industrial AMS was lab validated, field proven, and deployed at critical locations of seawater injection network for automated detection of microorganisms under the Saudi Arabia’s harsh environment. An excellent correlation between AMS measurement data (fluorescence count) and actual count of microbial cell number under microscope was established (coefficient of determination, R2 > 0.99) for converting AMS fluorescence count to cell numbers (cell mL-1) in the injection seawater. The pre-industrial AMS only required monthly maintenance with solutions refill, and was able to cope with hot summer months even without protection in an air-conditioned shelter. The study team recommended wider deployment of the online AMS for real-time monitoring of bacteria numbers in the various strategic locations in Saudi Aramco’s complex seawater injection network, as an integral component of pipeline corrosion and leak mitigation program.

50 years of scanning electron microscopy of bone——a comprehensive overview of the important discoveries made and insights gained into bone material properties in health,disease,and taphonomy

Bone is an architecturally complex system that constantly undergoes structural and functional optimisation through renewal and repair.The scanning electron microscope (SEM) is among the most frequently used instruments for examining bone.It offers the key advantage of very high spatial resolution coupled with a large depth of field and wide field of view.Interactions between incident electrons and atoms on the sample surface generate backscattered electrons,secondary electrons,and various other signals including X-rays that relay compositional and topographical information.Through selective removal or preservation of specific tissue components (organic,inorganic,cellular,vascular),their individual contribution(s) to the overall functional competence can be elucidated.With few restrictions on sample geometry and a variety of applicable sample-processing routes,a given sample may be conveniently adapted for multiple analytical methods.While a conventional SEM operates at high vacuum conditions that demand clean,dry,and electrically conductive samples,non-conductive materials (e.g.,bone) can be imaged without significant modification from the natural state using an environmental scanning electron microscope.This review highlights important insights gained into bone microstructure and pathophysiology,bone response to implanted biomaterials,elemental analysis,SEM in paleoarchaeology,3D imaging using focused ion beam techniques,correlative microscopy and in situ experiments.The capacity to image seamlessly across multiple length scales within the meso-micro-nano-continuum,the SEM lends itself to many unique and diverse applications,which attest to the versatility and user-friendly nature of this instrument for studying bone.Significant technological developments are anticipated for analysing bone using the SEM.

High-angle deflection of metagrating-integrated laser emission for high-contrast microscopy

Flat metaoptics components are looking to replace classical optics elements and could lead to extremely compact biophotonics devices if integrated with on-chip light sources and detectors.However,using metasurfaces to shape light into wide angular range wavefronts with high efficiency,as is typically required in high-contrast microscopy applications,remains a challenge.Here we demonstrate curved GaAs metagratings integrated on vertical-cavity surface-emitting lasers(VCSELs)that enable on-chip illumination in total internal reflection and dark field microscopy.Based on an unconventional design that circumvents the aspect ratio dependent etching problems in monolithic integration,we demonstrate off-axis emission centred at 60°in air and 63°in glass with>90%and>70%relative deflection efficiency,respectively.The resulting laser beam is collimated out-of-plane but maintains Gaussian divergence in-plane,resulting in a long and narrow illumination area.We show that metagrating-integrated VCSELs of different kinds can be combined to enable rapid switching between dark-field and total internal reflection illumination.Our approach provides a versatile illumination solution for high-contrast imaging that is compatible with conventional microscopy setups and can be integrated with biophotonics devices,such as portable microscopy,NIR-II range bioimaging,and lab-on-a-chip devices.

Calcium phosphates(CaP)represent an important class of osteoconductive and osteoinductive biomaterials.As proof-of-concept,we show how a multi-component CaP formulation(monetite,beta-tricalcium phosphate,and calcium pyrophosphate)guides osteogenesis beyond the physiological envelope.In a sheep model,hollow dome-shaped constructs were placed directly over the occipital bone.At 12 months,large amounts of bone(~75%)occupy the hollow space with strong evidence of ongoing remodelling.Features of both compact bone(osteonal/osteon-like arrangements)and spongy bone(trabeculae separated by marrow cavities)reveal insights into function/need-driven microstructural adaptation.Pores within the CaP also contain both woven bone and vascularised lamellar bone.Osteoclasts actively contribute to CaP degradation/removal.Of the constituent phases,only calcium pyrophosphate persists within osseous(cutting cones)and non-osseous(macrophages)sites.From a translational perspective,this multi-component CaP opens up exciting new avenues for osteotomy-free and minimally-invasive repair of large bone defects and augmentation of the dental alveolar ridge.

Luminescent properties of praseodymium doped Y_2O_3 and LaAlO_3 nanocrystallites and polymer composites

Luminescent properties of Y2O3 and LaAlO3 nanopowders doped with Pr^3＋ ions and PMMA-based composite materials doped with these powders were reported. The set of active nanopowders differing in praseodymium ions concentration was prepared using sol-gel method and carefully characterized with respect to emission properties in the visible spectral range. In particular, the excitation and emission spectra were measured together with fluorescence decay profiles and the differences between optical properties of these materials were discussed and compared with data available for bulk materials. Finally, the PMMA-based composite materials doped with Pr^3＋：Y2O3 and Pr^3＋：LaAlO3 nanopowders were manufactured and characterized. The obtained results showed that polymer composites doped with active nanopowders in general tended to keep the luminescent properties of the original nanopowders.

Antimicrobial peptides(AMPs)are potentially powerful alternatives to conven-tional antibiotics in combating multidrug resistance,given their broad spectrum of activity.They mainly interact with cell membranes through surface electrostatic potentials and the formation of secondary structures,resulting in permeability and destruction of target microorganism membranes.Our earlier work showed that two leading AMPs,MSI-78(4–20)and pardaxin(1–22),had potent antimicrobial activ-ity against a range of bacteria.It is known that the attachment of moderate-length lipid carbon chains to cationic peptides can further improve the functionality of these peptides through enhanced interactions with the membrane lipid bilayer,inducing membrane curvature,destabilization,and potential leakage.Thus,in this work,we aimed to investigate the antimicrobial activity,oligomerization propensity,and lipid-membrane binding interactions of a range of N-terminal lipidated analogs of MSI-78(4–20)and pardaxin(1–22).Molecular modeling results suggest that aggregation of the N-lipidated AMPs may impart greater structural stability to the peptides in solu-tion and a greater depth of lipid bilayer insertion for the N-lipidated AMPs over the parental peptide.Our experimental and computationalfindings provide insights into how N-terminal lipidation of AMPs may alter their conformations,with subsequent effects on their functional properties in regard to their self-aggregation behavior,membrane interactions,and antimicrobial activity.

Semiconductor quantum dots(QDs)have been demonstrated viable for efficient light emission applications,in particular for the emission of single photons on demand.However,the preparation of QDs emitting photons with predefined and deterministic polarization vectors has proven arduous.Access to linearly polarized photons is essential for various applications.In this report,a novel concept to directly generate linearly-polarized photons is presented.This concept is based on InGaN QDs grown on top of elongated GaN hexagonal pyramids,by which the predefined elongation determines the polarization vectors of the emitted photons from the QDs.This growth scheme should allow fabrication of ultracompact arrays of photon emitters,with a controlled polarization direction for each individual emitter.

Treatment of Budd-Chiari syndrome with a focus on transjugular intrahepatic portosystemic shunt

AIM:To evaluate long-term complications and survival in patients with Budd-Chiari syndrome (BCS) referred to a Danish transjugular intrahepatic portosystemic shunt (TIPS) centre.METHODS:Twenty-one consecutive patients from 1997-2008 were retrospectively included [15 women and 6 men,median age 40 years (range 17-66 years)].Eighteen Danish patients came from the 1.8 million catchment population of Aarhus University Hospital and three patients were referred from Scandinavian hospitals.Management consisted of tests for underlying haematological,endocrinological,or hypercoagulative disorders parallel to initiation of specific treatment of BCS.RESULTS:BCS was mainly caused by thrombophilic (33%) or myeloproliferative (19%) disorders.Fortythree percents had symptoms for less than one week with ascites as the most prevalent finding.Fourteen (67%) were treated with TIPS and 7 (33%) were manageable with treatment of the underlying condition and diuretics.The median follow-up time for the TIPS-treated patients was 50 mo (range 15-117 mo),and none required subsequent liver transplantation.Ascites control was achieved in all TIPS patients with a marked reduction in the dose of diuretics.A total of 14 TIPS revisions were needed,mostly of uncovered stents.Two died during follow-up:One non-TIPS patient worsened after 6 mo and died in relation to transplantation,and one TIPS patient died 4 years after the TIPS-procedure,unrelated to BCS.CONCLUSION:In our BCS cohort TIPS-treated patients have near-complete survival,reduced need for diuretics and compared to historical data a reduced need for liver transplantation.
