Induction of deletion mutation on ompR gene of Salmonella enterica serovar Typhi isolates from asymptomatic typhoid carriers to evolve attenuated strains for vaccine development

Objective:To develop allenualed slrains of Salmonella enterica serorar Typhi(***) for the candidate vaccine by osmolar ***:*** SS3 and SS5 strains were isolated from asymptomatic typhoid carriers in Mamakkal,Tamil *** strains were grown in LB(Luria Bertani) medium supplemented with various concentration of NaCl(0.1- 0.7M) *** effecl of osmolar stress was determined at molecular level by PCR using MCR 06 and MCR07 primers corresponding to ompR with chromosomal DNA of *** SS3 and SS5 *** by osmolar stress results in deletion mutation of *** slrains was determined by agglutination assays,precipitation *** PAGE analysis and by animal ***:The 799 bp amplified ompR gene product from wild type *** SS3 and SS5 illustrate the presence of virulent ***,there was only a 282 bp amplified product from *** SS3 and SS5 grown in the presence of 0.***.0.6 and 0.7 M *** illustrates the occurrence of deletion mutation in ompR gene al high concentration of ***,both the wildtype and mutant *** outer membrane *** reveals the differences in the expression of *** and ompA *** mice,wild type and mutant strains lethal dose(LD_(50)) were *** mice died within 72 h when both the wild type strains were injected intraperitoneally with 3 log CFU-mL^(-1).When the mice were injected with the mutants in same dosage,no clinical symptoms were observed;whereas the serum antibodv litre was elicited within two weeks indicated that the mutants have the ability to induce protective humoral immune *** results suggest that *** SS3 and SS5 may bo used as good candidate strains for the development of live attenuated vaccine against ***:This study demonstrates that the *** strains were allenualed and could be good vaccine candidates in future.

Antipyretic,anti-inflammatory and analgesic properties of nilavembu kudineer choornam:a classical preparation used in the treatment of chikungunya fever

Objective:To investigate the efficacy of ethanolic extract of nilavembu kudineer choornam (EENkC) in inflammation,pain and fever using animal models to support its ***: Acute toxicity study of EENkC was performed in mice to fix the effective *** antipyretic, anti-inflammatory and analgesic activity of EENkC was evaluated in brewer’s yeast induced pyrexia in rats,carrageenan-induced inflammation in rats and acetic-acid induced writhing in mice ***:Acute toxicity revealed that EENkC didn’t show death and toxic signs up to 2 000 mg/*** brewer’s yeast induced pyrexia and carrageenan-induced inflammation EENkC at the doses of 200 and 400 mg/kg inhibited fever and inflammation significantly(PkC than control animals. EENkC 200 mg/kg inhibits inflammation higher level in carrageenan-induced paw edema,but there is no significant difference when compared to indomethacin 10 mg/***:The present findings revealed that EENkC possesses antipyretic,anti-inflammatory and analgesic activity which supports nilavembu kudineer choornam efficacy in chikungunya fever.

在高海拔喜马拉雅山地带，滑坡是非常普遍的。喜马拉雅山地带的主要公路，由于受滑坡灾害的严重影响而频繁堵塞，且长期被迫中断。需要对这些滑坡采取长期防治措施以保持公路畅通。位于甘托克（印度锡金邦首都）北部71．2km处的Lanta khota滑坡，是锡金北部公路上最古老的滑坡之一，自1975年起该滑坡开始活动。滑坡两侧的岩石类型是不同的（东部为透镜状片麻岩，西部为长石英质片岩），我们认为，主中央冲断层（MCT）穿过滑坡带。由于滑坡总是在强降雨过后变得异常活跃，因此，查明滑坡面之下含水介质结构，了解滑坡活动的诱发因素湿得非常重要。这仅能通过地球物理调查完成。然而，必须选择适宜这种地形勘查的地球物理技术。为了查明地下地层结构，在Lanta khola滑坡区域开展了甚低频（VLF）电磁法测量。虽然全球仅有少数可利用的VLF发射台，但可以从一些VLF发射台获取VLF信号，即便是在高海拔的山区。垂直地质岩层走向布置了5个VLF测量剖面，并根据VLF测量结果圈定出低阻区。这种低阻区与根据梯度电阻率测深剖面圈定的低电阻带有关。这些异常证实，在Lanta khola滑坡区域范围内，即使在与片麻岩-片岩地质接触面平行的地下滑动面中也存在水饱和带（潮湿带）。这表明，传导特征与弱水饱和的岩屑层有关，这些岩屑层位于滑坡体沿线并与地质接触面平行。为此，滑坡区两侧的电性结构特征可能与稳定地层有关。为了把滑坡活动次数降至最低，必需排放潮湿带中的水，这是因为潮湿带中的水可渗入滑坡体。

Structural behavior of intermediate length cold-formed steel rack columns with C-stitches

This article presents an experimental and numerical investigation on the strength and performance of intermediate length rack column sections with C-stitches under axial compression. The test program consisted of 10 axial concentric compression tests on columns with and without C-stitches under pin end conditions for two different geometric lengths. Finite element (FE) models were developed using commercial FE package ABAQUS considering material and geometric nonlinearities as well as initial geometric imperfections. The elastic buckling properties of the section were calculated using readily available linear elastic buckling analysis tools based on Generalized Beam Theory (GBT) and Finite Strip Method (FSM). Obtained FE results were compared with those obtained experimentally, and once verified the developed FE modeling technique was used to carry out a parametric study to examine changes in structural response due to variations in length, depth and spacing of C-stitches. Observed influences of C-stitches on the behavior and resistance of the considered columns were carefully analyzed, and key design aspects are presented herein.

Indoor versus outdoor scene recognition for navigation of a micro aerial vehicle using spatial color gist wavelet descriptors

In the context of improved navigation for micro aerial vehicles,a new scene recognition visual descriptor,called spatial color gist wavelet descriptor(SCGWD),is *** was developed by combining proposed Ohta color-GIST wavelet descriptors with census transform histogram(CENTRIST)spatial pyramid representation descriptors for categorizing indoor versus outdoor scenes.A binary and multiclass support vector machine(SVM)classifier with linear and non-linear kernels was used to classify indoor versus outdoor scenes and indoor scenes,*** this paper,we have also discussed the feature extraction methodology of several,state-of-the-art visual descriptors,and four proposed visual descriptors(Ohta color-GIST descriptors,Ohta color-GIST wavelet descriptors,enhanced Ohta color histogram descriptors,and SCGWDs),in terms of experimental *** proposed enhanced Ohta color histogram descriptors,Ohta color-GIST descriptors,Ohta color-GIST wavelet descriptors,SCGWD,and state-of-the-art visual descriptors were evaluated,using the Indian Institute of Technology Madras Scene Classification Image Database two,an Indoor-Outdoor Dataset,and the Massachusetts Institute of Technology indoor scene classification dataset[(MIT)-67].Experimental results showed that the indoor versus outdoor scene recognition algorithm,employing SVM with SCGWDs,produced the highest classification rates(CRs)—95.48%and 99.82%using radial basis function kernel(RBF)kernel and 95.29%and 99.45%using linear kernel for the IITM SCID2 and Indoor-Outdoor datasets,*** lowest CRs—2.08%and 4.92%,respectively—were obtained when RBF and linear kernels were used with the MIT-67 *** addition,higher CRs,precision,recall,and area under the receiver operating characteristic curve values were obtained for the proposed SCGWDs,in comparison with state-of-the-art visual descriptors.

Protective Role of C-Phycocyanin Against Secondary Changes During Sodium Selenite Mediated Cataractogenesis

Age related cataract is the leading cause of blindness associated with accumulation of oxidative stress in the eye *** present investigation reveals the rational of the beneficial effects of the natural compound C-phycocyanin(CPC)is beneficial when administered to rat pups to protect against the secondary effects of sodium selenite induced cataractogenesis.A single subcutaneous dose of sodium selenite(19 lmol/kg body weight)on the 10th day of postpartum is adequate to induce cataract in rat *** biochemical parameters,such as the level of electrolytes,mean activities of anti-oxidant enzymes *** dismutase,catalase and reduced glutathione were observed to be significantly altered during selenite induced cataractogenic *** examination revealed signs of degradation of normal cell architecture in the liver,kidney and eye ***,the deleterious effects of sodium selenite toxicity were restored with the simultaneous treatment with *** results suggest that an administration of 200 mg/kg body weight of C-PC has the ability to prevent/alter the secondary changes reflected in the serum biochemical and histological modifications in rats exposed to sodium *** results complement the beneficial role of C-PC of cyanobacterial origin as a efficacious anti-cataractogenic agent against sodium selenite toxicity.

Protective effect of Pisonia aculeata on thioacetamide induced hepatotoxicity in rats

Objective:To evaluate the protective effect of Pisonia aculeata(***) on thioacetamide induced hepatotoxicity in ***:Male Wistar rats were administered 250 or 500 mg/kg *** *** extract for 21 days and simultaneously administered thioacetamide(TAA) 50 mg/kg bw s.c.1 h alter the respective assigned treatments every 72 *** the end of all experimental methods,all the animals were sacrificed by cervical *** samples were *** was separated and analyzed for various biochemical ***:TAA induced a significant rise in aspartate amino transferase(AST),alanine amino transferase(ALT),alkaline phosphatase(ALP),total bilirubin,gamma glutamate transpeptidase(GGTP),lipid peroxidase(LPO)with a reduction of total protein,superoxide dismutase(SOD),catalase,glutathione peroxidase(GPx) and glutathione S-transferase(GST).Treatment of rats with different does of plant extract(250 and 500 mg/kg) significantly(Pker enzymes and antioxidant levels to near normal against TAA treated *** activity of the extract at a dose of 300 tug/kg was comparable to the standard drug,silymarin(50 mg/kg.p.o.).Conclusions:It can be concluded that *** extract possesses a remarkable hepaloprolective and antioxidant activity against TAA induced *** research is required lo derive an optimal therapeutic dose.

Potential applications of lactic acid bacteria and bacteriocins in antimycobacterial therapy

Tuberculosis(TB) is a communicable disease caused by Mycobacterium tuberculosis(M. tuberculosis). WHO estimated that 10.4 million new(incident) TB cases worldwide in year 2016. The increased prevalence of drug resistant strains and side effects associated with the current anti-tubercular drugs make the treatment options more complicated. Hence, there are necessities to identify new drug candidates to fight against various sub-populations of M. tuberculosis with less or no toxicity/side effects and shorter treatment duration. Bacteriocins produced by lactic acid bacteria(LAB) attract attention of researchers because of its "Generally recognized as safe" status. LAB and its bacteriocins possess an effective antimicrobial activity against various bacteria and fungi. Interestingly bacteriocins such as nisin and lacticin 3147 have shown antimycobacterial activity in vitro. As probiotics, LAB plays a vital role in promoting various health benefits including ability to modulate immune response against various infectious diseases. LAB and its metabolic products activate immune system and thereby limiting the M. tuberculosis pathogenesis. The protein and peptide engineering techniques paved the ways to obtain hybrid bacteriocin derivatives from the known peptide sequence of existing bacteriocin. In this review, we focus on the antimycobacterial property and immunomodulatory role of LAB and its metabolic products. Techniques for large scale synthesis of potential bacteriocin with multifunctional activity and enhanced stability are also discussed.

Mutation in angiotensin II type 1 receptor disrupts its binding to angiotensin II leading to hypotension： An insight into hydrogen bonding patterns

To understand the role of angiotensin II type 1 receptor gene （AGTR1） gene products in relation to hypotension we have analyzed the single nucleotide polymorphisms （SNPs） associated with this gene. This can help us to understand the genetic variations that can alter the function of the gene products. In this present study, we report the polymorphic variant associated with AGTR1 and its weak interaction with angiotensin II （AngII） which leads to hypotension. Out of 1318 SNPs, six are found to be non-synonymous, of which rs1064533 shows significant damaging effect. A missense mutation （T1255G）, i.e., from thymine to guanine for rs1064533 in AGTR1 gene results in amino acid substitution from cysteine （Cys） to tryptophan （Trp） in the receptor protein. A strong hydrogen bond exists between Cys289 of native AGTR1 protein and glutamine 167 of AngII. Interestingly, it is replaced by a weak hydrogen bond in the mutant protein between Trp289 （mutant residue） and serine 340. Such a substitution from small, hydrophilic to bulky, hydrophobic residue in AGTR1 protein results in reduced binding affinity of the receptor protein with AngII, leading to hypotension. The results presented from this in silico study will open up new prospect for genetic analysis of AGTR1 gene and will be beneficial to the researchers for understanding the role played by AGTR1 gene in hypotension disease.
