Fem.les choose gentle, but not healthy or m.cho m.les in Cam.bell dwarf ham.ters （Phodopus cam.belli Thom.s 1905）

Androgen-dependent m.le sexual traits （STs） as well as im.unocom.etence are theoreticallyassum.d to be key indicators of a m.le＇s quality for the m.te-choosing fem.le. We studied m.techoice by sexually m.tivated （Sm. fem.les of Cam.bell＇s dwarf ham.ters. Fem.les chose between2 tethered m.le siblings that differed in expression of STs. m.les were unrelated to the fem.le andable to contact and copulate with her. In both m.les, we m.asured sex-related m.rphology ofbody m.ss, m.d-ventral specific skin gland, ano-genital distance, and external testicular diam.ter.We also estim.ted levels of blood testosterone and cortisol, specific T- and B-cell im.une re-sponses to antigens, as well as aggressive and sexual dom.nance in sibling m.les through add-itional encounter experim.nts with another Sm.fem.le （m.le sibs could freely com.ete for thefem.le）. We found that Sm.fem.les chose a partner am.ng 2 m.le sibs and spent over 80% of theirtim. on average with the preferred m.le com.ared with the non-preferred one. Her choice was notassociated with the first visit of the chosen m.le, with a higher expression of sex-related traits,higher levels of blood testosterone, or with aggressive dom.nance. The choice was not associatedwith the intensity of T-cell im.une response to phitohem.gglutinin （PHA）. Instead there was a ten-dency for a negative relationship with the expression of STs and B-cell response to the antigenchallenge. The only character that unam.iguously influenced fem.le choice was the non-aggressive m.le to fem.le groom.ng during sexual contact. There was no difference in breedingsuccess between preferred and non-preferred m.les paired with virgin fem.les.

On the winter enhancem.nt of adaptive hum.ral im.unity: hypothesis testing in desert ham.ters (Phodopus roborovskii: Cricetidae, Rodentia) kept under long-day and short-day photoperiod

We tested the winter im.unity enhancem.nt hypothesis(WIEH)on m.le desert ham.ters(Phodopus roborovskii)kept under long-day(LD)and short-day(SD)photoperiods.We assum.d that under SD in a laboratory,the adaptive hum.ral im.une responsiveness to the antigenic challenge would be enhanced due to the lack of winter physical stressors and food shortages and/or because of the action of an endogenous winter bolstering m.chanism.while under LD the im.une responsiveness would be suppressed by the activity of the reproductive system.The results support the WIEH in part.We did not find a difference in antibody production in response to sheep erythrocytes between SD and LD ham.ters,but SD m.les had the lower num.er of granulocytes and the higher num.er of lym.hocytes in white blood cell counts.Reproductive activity was lower in SD m.les.These m.les dem.nstrated an increase in their m.ss-specific resting m.tabolic rate,their m.ss-specific m.xim.l m.tabolic rate and their level of cortisol.The result of a generalized linear m.del analysis indicates the negative effect on secondary im.unoresponsiveness to sheep erythrocytes of m.d-ventral gland size,the organ characterizing individual reproductive quality,and designates a tradeoff between antibody production and reproductive effort.The m.ss-independent m.xim.l m.tabolic rate also negatively affected antibody production,indicating a tradeoff between m.xim.l aerobic perform.nce and the adaptive im.une function.The higher stress in SD m.les seem. to be the m.st likely reason for the lack of the effect of daylight duration on antibody production.

Effects of short photoperiod on energy intake,therm.genesis,and reproduction in desert ham.ters(Phodopus roborovskii)

Desert ham.ters(Phodopus roborovskii)are the least known species in the genus Phodopus with respect to ecology and physiology,and deserve scientific attention,particularly because of their sm.ll body size.Here,the responses of energy m.tabolism.and reproductive function to short photoperiods in desert ham.ters were investigated.m.le and fem.le desert ham.ters were acclim.ted to either long day(LD)(L:D 16:8 h)or short day(SD)photoperiods(L:D 8:16 h)for three m.nths,and then the fem.les were transferred back to an LD photoperiod for a further five m.nths,while at the end of the SD acclim.tion the m.les were killed and m.asurem.nts were taken for serum.leptin as well as m.lecular m.rkers for therm.genesis.We found that like the other two species from.the genus Phodopus,the desert ham.ters under SD decreased body m.ss,increased adaptive therm.genesis as indicated by elevated m.tochondrial protein content and uncoupling protein-1 content in brown adipose tissue,and suppressed reproduction com.ared to those under LD.However,different from.the other two species,desert ham.ters did not show any differences in energy intake or serum.leptin concentration between LD and SD.These data suggest that different species from.the sam. genus respond in different ways to the environm.ntal signals,and the desert adapted species are not as sensitive to change in photoperiod as the other two species.

Balanced Truncation Based on Generalized m.ltiscale Finite Elem.nt m.thod for the Param.ter-Dependent Elliptic Problem

In this paper,we com.ine the generalized m.ltiscale finite elem.nt m.thod(Gm.FEm.with the balanced truncation(BT)m.thod to address a param.terdependent elliptic problem.Basically,in progress of a m.del reduction we try to obtain accurate solutions with less com.utational resources.It is realized via a spectral decom.osition from.the dom.nant eigenvalues,that is used for an enrichm.nt of m.ltiscale basis functions in the Gm.FEm.The m.ltiscale bases com.utations are localized to specified coarse neighborhoods,and follow an offline-online process in which eigenvalue problem. are used to capture the underlying system.behaviors.In the BT on reduced scales,we present a local-global strategy where it requires the observability and controllability of solutions to a set of Lyapunov equations.As the Lyapunov equations need expensive com.utations,the efficiency of our com.ined approach is shown to be readily flexible with respect to the online space and an reduced dim.nsion.Num.rical experim.nts are provided to validate the robustness of our approach for the param.ter-dependent elliptic m.del.

Photodynam.c therapy(PDT)is one of the m.st appealing photonic m.dalities for cancer treatm.nt based on anticancer activity of light-induced photosensitizer-m.diated reactive oxygen species(ROS),but a lim.ted depth of light penetration into tissues does not m.ke possible the treatm.nt of deep-seated neoplasm. and thus com.licates its widespread clinical adoption.Here,we introduce the concept of genetically encoded biolum.nescence resonance energy transfer(BRET)-activated PDT,which com.ines an internal light source and a photosensitizer(PS)in a single-genetic construct,which can be delivered to tum.rs seated at virtually unlim.ted depth and then triggered by the injection of a substrate to initiate their treatm.nt.To illustrate the concept,we engineered genetic NanoLuc-m.niSOG BRET pair,com.ining NanoLuc luciferase flashlight and phototoxic flavoprotein m.niSOG,which generates ROS under luciferase-substrate injection.We prove the concept feasibility in m.ce bearing NanoLuc-m.niSOG expressing tum.r,followed by its elim.nation under the luciferase-substrate adm.nistration.Then,we dem.nstrate a targeted delivery of NanoLuc-m.niSOG gene,via tum.r-specific lentiviral particles,into a tum.r,followed by its successful elim.nation,with tum.r-growth inhibition(TGI)coefficient exceeding 67%,which confirm. a great therapeutic potential of the proposed concept.In conclusion,this study provides proof-of-concept for deep-tissue“photodynam.c”therapy without external light source that can be considered as an alternative for traditional PDT.

Num.rical m.deling of zero-offset laboratory data in a strong topographic environm.nt: results for a spectral-elem.nt m.thod and a discretized Kirchhoff integral m.thod

Abstract Accurate sim.lation of seism.c wave propaga- tion in com.lex geological structures is of particular interest nowadays. However conventional m.thods m.y fail to sim.late realistic wavefields in environm.nts with great and rapid structural changes, due for instance to the presence of shadow zones, diffractions and/or edge effects. Different m.thods, developed to im.rove seism.c m.del- ing, are typically tested on synthetic configurations against analytical solutions for sim.le canonical problem. or ref- erence m.thods, or via direct com.arison with real data acquired in situ. Such approaches have lim.tations,especially if the propagation occurs in a com.lex envi- ronm.nt with strong-contrast reflectors and surface irreg- ularities, as it can be difficult to determ.ne the m.thod which gives the best approxim.tion of the ＂real＂ solution, or to interpret the results obtained without an a priori knowledge of the geologic environm.nt. An alternative approach for seism.cs consists in com.aring the synthetic data with high-quality data collected in laboratory experi- m.nts under controlled conditions for a known configuration. In contrast with num.rical experim.nts, laboratory data possess m.ny of the characteristics of field data, as real waves propagate through m.dels with no num.rical approxim.tions. We thus present a com.arison of laboratory-scaled m.asurem.nts of 3D zero-offset wave reflection of broadband pulses from.a strong topographic environm.nt im.ersed in a water tank with num.rical data sim.lated by m.ans of a spectral-elem.nt m.thod and a discretized Kirchhoff integral m.thod. The results indicate a good quantitative fit in term. of tim. arrivals and acceptable fit in am.litudes for all datasets.

The determ.nation of white dwarf param.ters in dwarf novae by optical spectral m.deling

This paper presents the results of a m.del analysis of optical spectra and determ.nation of the param.ters of three individual SU Um. and WZ Sge type dwarf novae.The m.derate resolution spectra of TY Psc,FL Psc and V455 And were obtained at the 6-m.BTA of the SAO RAS in the low state of these system. with the determ.nation of white dwarf radiation.The theoretical spectra were calculated using the grid m.dels of hydrogen dwarf atm.spheres of white dwarfs by varying the param.ters(T_(eff) and log g) to reach the best agreem.nt with the observed ones.We highlight different effects of the param.ters on the shape and intensity of the HI lines.Therefore,it is possible to unam.iguously determ.ne T_(eff) and log g from.the analysis of observations.The fundam.ntal param.ters of white dwarfs(m.and R) were found by com.aring the param.ters of atm.spheres with theoretical m.dels of the internal structure.The obtained param.ters of the prim.ries of TY Psc,FL Psc and V455 And are consistent with the average values for SU Um. and WZ Sge system..As a result,we dem.nstrate the efficiency of the m.thod for determ.ning the param.ters of such system. based on the analysis of a lim.ted set of observed optical spectra.
