Long-term and non-invasive in vivo tracking of DiD dye-labeled human hepatic progenitors in chronic liver disease models

BACKGROUND Chronic liver diseases(CLD)are the major public health burden due to the continuous increasing rate of global morbidity and *** inherent limitations of organ transplantation have led to the development of stem cell-based therapy as a supportive and promising therapeutic ***,identifying the fate of transplanted cells in vivo represents a crucial *** To evaluate the potential applicability of DiD dye as a cell labeling agent for longterm,and non-invasive in vivo tracking of transplanted cells in the *** Magnetically sorted,epithelial cell adhesion molecule positive(1×106 cells/mL)fetal hepatic progenitor cells were labeled with DiD dye and transplanted into the livers of CLD-severe combined immunodeficiency(SCID)***-infrared(NIR)imaging was performed for in vivo tracking of the DiD-labeled transplanted cells along with colocalization of hepatic markers for up to 80 *** existence of human cells within mouse livers was identified using Alu polymerase chain reaction and *** NIR fluorescence imaging of CLD-SCID mice showed a positive fluorescence signal of DiD at days 7,15,30,45,60,and 80 ***,positive staining of cytokeratin,c-Met,and albumin colocalizing with DiD fluorescence clearly demonstrated that the fluorescent signal of hepatic markers emerged from the DiD-labeled transplanted *** of liver function was also observed with serum levels of glutamic-oxaloacetic transaminase,glutamate-pyruvate transaminase,and *** detection of human-specific Alu sequence from the transplanted mouse livers provided evidence for the survival of transplanted cells at day *** DiD-labeling is promising for long-term and non-invasive in vivo cell tracking,and understanding the regenerative mechanisms incurred by the transplanted cells.

To study the anti-nociceptive effect of domper-idone and cisapride in mice. METHODS: Initially, the effect of these drugs on motor activity was tested using rotarod. The anti-nociception was tested using chemical and mechanical assay. In the chemical assay, the number of abdominal constrictions either in the saline treated animals or in the domperidone/cisapride (1, 5, or 10 mg/kg either po or ip) treated mice, were recorded for a period of 30 min after acetic acid challenge (10 mLAg, of 0.6 % acetic acid ip). In the tail clip assay, the time taken by the mouse to make attempts to dislodge the bulldog clamp placed at the tail (reaction time) was recorded with a cut off time of 30 s. The role of opioid pathways was examined by pretreating the animals with naloxone (1 mg/kg, ip) 30 min prior to domperidone and cisapride. RESULTS: Domperidone and cisapride, both reduced the number of abdominal constrictions when given orally or intraperitoneally. Domperidone (5 mg/kg) inhibited it to the extent of 57.0 % a

Micropropagation protocol for Salvadora oleoides

Salvadora oleoides Decne. is a pharmaceutically important plant. Owing to poor seed formation, viability and, germination, and to anthropogenic disturbances, this species is on the verge of extinction. A reproducible micropropagation protocol to increase the population through tissue culture has been standardized and the results are reported here. Callus tissues were initiated from young leaves and stem explants. Leaf calluses proliferated with 1.5 mg/L BAP and 0.9 mg/L 2, 4-D with additives and continuous slow proliferation up to 15 weeks on 0.5 mg/L BAP and additives with 200 mg/L activated *** shoot initiation took place from stem node explants after 12 days; 4–5 shoots per node were produced in 30 days. Shoot clumps elongated and grew further on MS media supplemented with 2 mg/L BAP, 0.2 mg/L NAA and additives, which generated 20–23 shoots. The elongated shoots induced tap roots with 4 mg/L NAA and200 mg/L activated charcoal in 12 days. In vitro raised plants produced secondary roots when transferred to pots containing vermiculite maintained at 28–35 ℃. The plantlets successfully acclimatised in pots containing soil in natural conditions.

Compton scattering studies and electronic properties of cerium

Experimental electron momentum density of γ-Ce was measured at an intermediate resolution （0.38 a.u.） using 20 Ci 137Cs Compton spectrometer. The experiment was compared with the first ever theoretical Compton profiles computed using spin polarized relativistic Korringa-Kohn-Rostoker （SPR-KKR） method within atomic sphere approximation; and linear combination of atomic orbitals （LCAO） within generalised gradient approximation （GGA） and recently developed second order generalised gradient approximation （SOGGA）. It was seen that the Compton profile derived from LCAO with GGA and SOGGA schemes gave a reasonable agreement with the experimental Compton line shape. Theoretical anisotropies in Compton profiles derived along [100], [110] and [111] directions were explained in terms of degenerate states in the energy bands.

The generation of high-order harmonics in gases enabled to probe the attosecond electron dynamics in atoms and molecules with unprecedented *** these techniques to solids,which were originally developed for atomic and molecular gases,requires a fundamental understanding of the physics that has been partially addressed ***,we employ timedependent density-functional theory to investigate how the electron dynamics resulting in high-harmonic emission in monolayer hexagonal boron nitride is affected by the presence of *** show how these realistic spin-polarised defects modify the harmonic emission and demonstrate that important differences exist between harmonics from a pristine solid and a defected *** particular,we found that the different spin channels are affected differently by the presence of the spin-polarised point ***,the localisation of the wavefunction,the geometry of the defect,and the electron–electron interaction are all crucial ingredients to describe high-harmonic generation in defected solids.

Loading Detection and Number Estimation of an Electron Plasma in a Penning Trap

A quadrupole Penning trap for spectroscopy and investigations of non-neutral plasmas was designed and built. In this work we provide details of the trap design and a discussion of a simple design and procedure for convenient electron loading from an aligned filament. Electrons from thermionic emission which form a low-energy diffuse beam are trapped in weak magnetic fields. They are detected through a non-destructive electronic detection scheme, the details of which are discussed. The detection signal is diminished when the electron beam energy is increased while the electron flux is kept constant. This is explained by considering the energy shift in the distribution function of electrons emitted from the filament and entering the trap. We present a calculation of the number of trapped electrons from the shape of the detection signal. This calculation, based on a model of a driven damped harmonic oscillator to describe the axial motion of the electrons, compares favourably with the numbers obtained by measurements of the space charge induced shift in the trap potential.

Plant studies in the first Himalayan Biosphere Reserve: a review

Nanda Devi Biosphere Reserve(NDBR) was declared as the first Himalayan Biosphere Reserve owing to its unique biological and cultural wealth. Its core zones, Nanda Devi National Park and Valley of Flowers National Park, are a UNESCO World Natural Heritage Site. In spite of lying at a high altitude, interplay of factors such as unique geographical location, climate, topography and wide altitudinal variations have endowed NDBR with a rich and diverse flora. Proportionately high percentage of endemic and near endemic plants makes NDBR a very important protected area from conservation point of view. However, its floristic wealth is facing unprecedented threats in the form of climate change and growing anthropogenic pressure. Hence, a need was felt to assess the directionality, quality and sufficiency of past and ongoing research for the conservation of floral and ethnobotanical wealth of NDBR in the absence of any such previous attempt. Based on an extensive review of more than 150 plant studies on NDBR, this communication provides a detailed account of the current state of knowledge and information gaps on flora, vegetation ecology, rare, endangered, threatened(RET) and endemic plants and ethnobotany. Priority research areas and management measures are discussed for the conservation of its unique floral wealth. Incomplete floral inventorization, lack of biodiversity monitoring, meagre studies on lower plant groups, population status of medicinal plants, habitat assessment of threatened taxa and geo-spatial analysis of alpine vegetation were identified as areas of immediate concern.
