Pharmacokinetics/Pharmacodynamics study of Fixtral SB as compared to supra bioavailable itraconazole and conventional itraconazole

BACKGROUND Itraconazole is a broad-spectrum triazole antifungal inhibiting fungal growth by inhibiting ergosterol synthesis and exhibits a nonlinear pharmacokinetic profile.Erratic absorption pattern with wide fluctuations in blood levels causes inconsistent and unpredictable clinical behaviour of this drug despite its low minimum inhibitory concentration(MIC)as compared to other antifungal agents.AIM To compare the oral bioavailability and bioequivalence of Fixtral SB(supra bioavailable itraconazole)with reference product R2(supra bioavailable 2×50 mg itraconazole).METHODS The study population consisted of 54 healthy volunteers,aged between 18-45 years and randomized to receive a single oral dose of either test[T;Fixtral SB(supra bioavailable itraconazole)100 mg]or reference product(R1;Sporanox 100 mg×2 capsules and R2;Lozanoc capsules 50 mg×2 capsules).Blood samples were taken pre-dose and post-dose up to 96 h.The study evaluated bioequivalence by comparing the oral bioavailability of the test product with reference product R2.The pharmacodynamic characteristics of the drug were evaluated by comparing the test product with reference product R1.Pharmacokinetics(PK)-PD comparative analysis[area under the concentration-time curve(AUC)/minimum inhibitory concentration(MIC)>25]was performed for conventional itraconazole 100 mg and supra bioavailable itraconazole 50 mg.Adverse events(AEs)assessments were performed in each study period and post-study evaluation.RESULTS Statistical analysis of primary PK variables revealed bioequivalence,with confidence intervals being completely inside the acceptance criteria of 80%-125%.The peak concentration levels of itraconazole were achieved at 10 h(T)and 8.5 h(R2),respectively.Pharmacodynamic parameter assessment showed that AUC/MIC for R1 are comparable to Fixtral SB 100mg for MIC levels up to 16mcg/mL(P>0.05 and observed P=0.3196).Six AEs were observed that were mild to moderate in severity and resolved.No severe AE was reported.CONCLUSION Test product itraconazole Capsule 100

Systemic hypertension is an established risk factor for coronary artery disease and cerebrovascular accident and control of blood pressure reduces the risk of a major cardiovascular event.Both non-pharmacological and pharmacological treatment options are available to treat hypertension.Yoga,recently received more attention as a treatment modality for various lifestyle disorders,even though practiced in India since ancient times.In this review,we are analyzing the role of yoga in the treatment of systemic hypertension.

2,5-Dimercapto-1,3,4-Thiadiazole(DMCT)-Based Polymers for Rechargeable Metal-Sulfur Batteries

Organosulfur materials are a sustainable alternative to the present-day layered oxide cathodes in lithium-based batteries.One such organosulfur material that was intensely explored from the 1990s to early 2010s is 2,5-dimercapto-1,3,4-thiadiazole(DMCT).However,research interest declined as the electrode reactions with DMCT were assumed to be too sluggish to be practical.Armed with the advances in metal-sulfur batteries,we revisit DMCT-based materials in the form of poly[tetrathio-2,5-(1,3,4-thiadiazole)],referred to as pDMCT-S.With an appropriate choice of electrode design and electrolyte,pDMCT-S cathode paired with a Li-metal anode shows a capacity of 715 mA h g^(-1)and a Coulombic efficiency of 97.7%at a C/10 rate,thus quelling the concerns of sluggish reactions.Surprisingly,pDMCT-S shows significantly improved long-term cyclability compared to a sulfur cathode.Investigations into the origin of the stability reveals that the discharge product Li-DMCT in its mesomeric form can strongly bind to polysulfides,preventing their dissolution into the electrolyte and shuttling.This unique mechanism solves a critical problem faced by sulfur cathodes.Encouragingly,this mechanism results in a stable performance of pDMCT-S with Na-metal cells as well.This study opens the potential for exploring other organic materials that have inherent polysulfide sequestering capabilities,enabling long-life metal-sulfur batteries.

Statistical optimization and formulation of microalga cultivation medium for improved omega 3 fatty acid production

Microalgae are considered a rich source of high-value metabolites with an array of nutraceutical and pharmaceutical applications.Diferent strategies have been developed for cultivating microalgae at large-scale photobioreactors but high cost and low productivity are the major hurdles.Optimizing the composition of media for the cultivation of microalgae to induce biomass production and high-value metabolite accumulation has been considered as an important factor for sustainable product development.In this study,the efect of plant growth regulators together with basal microalgal cultivation medium on biomass,total lipid,and EPA production was studied using the Plackett–Burman model and Response surface methodology.The traditional one-factor-at-a-time optimization approach is laborious,time-consuming,and requires more experiments which makes the process and analysis more difcult.The Designed PB model was found to be signifcant for biomass(396 mg/L),lipid(254 mg/L),and EPA(5.6%)production with a P value<0.05.The major objective of this study is to formulate a medium for EPA production without compromising the growth properties.Further,we had formulated a new media using RSM to achieve the goal and the signifcant variables selected were NaNO_(3),NaH_(2)PO_(4),and IAA and was found to be signifcant with 16.72%EPA production with a biomass production of 893 mg/L with a P value<0.05.The formulated medium can be used in large-scale cultivation systems which can enhance biomass production as well as the omega 3 fatty acid production in marine microalgae Nannochloropsis oceanica.

Secured Vehicle Life Cycle Tracking Using Blockchain and Smart Contract

Life Cycle Tracking(LCT)involves continuous monitoring and analy-sis of various activities associated with a vehicle.The crucial factor in the LCT is to ensure the validity of gathered data as numerous supply chain phases are involved and the data is assessed by multiple stakeholders.Frauds and swindling activities can be prevented if the history of the vehicles is made available to the interested parties.Blockchain provides a way of enforcing trustworthiness to the supply chain participants and the data associated with the various actions per-formed.Machine learning techniques when combined decentralized nature of blockchains can be used to develop a robust Vehicle LCT model.In the proposed work,Harmonic Optimized Gradient Descent andŁukasiewicz Fuzzy(HOGD-LF)Vehicle Life Cycle Tracking in Cloud Environment is proposed and it involves three stages.First,the Progressive Harmonic Optimized User Registra-tion and Authentication model is designed for computationally efﬁcient registra-tion and authentication.Next,for the authentic user,the Gradient Descent Blockchain-based SVM Data Encryption model is designed with minimum CPU utilization.Finally,Łukasiewicz Fuzzy Smart Contract Veriﬁcation is per-formed with encrypted data to ensure accurate and precise fraudulent activity deduction.The experimental analysis shows that the proposed method achieves signiﬁcant performance in terms of life cycle’s prediction time,overhead,and accuracy for a different number of users.

Dynamic Ensemble Multivariate Time Series Forecasting Model for PM2.5

In forecasting real time environmental factors,large data is needed to analyse the pattern behind the data values.Air pollution is a major threat towards developing countries and it is proliferating every year.Many methods in time ser-ies prediction and deep learning models to estimate the severity of air pollution.Each independent variable contributing towards pollution is necessary to analyse the trend behind the air pollution in that particular locality.This approach selects multivariate time series and coalesce a real time updatable autoregressive model to forecast Particulate matter(PM)PM2.5.To perform experimental analysis the data from the Central Pollution Control Board(CPCB)is used.Prediction is car-ried out for Chennai with seven locations and estimated PM’s using the weighted ensemble method.Proposed method for air pollution prediction unveiled effective and moored performance in long term prediction.Dynamic budge with high weighted k-models are used simultaneously and devising an ensemble helps to achieve stable forecasting.Computational time of ensemble decreases with paral-lel processing in each sub model.Weighted ensemble model shows high perfor-mance in long term prediction when compared to the traditional time series models like Vector Auto-Regression(VAR),Autoregressive Integrated with Mov-ing Average(ARIMA),Autoregressive Moving Average with Extended terms(ARMEX).Evaluation metrics like Root Mean Square Error(RMSE),Mean Absolute Error(MAE)and the time to achieve the time series are compared.

Disseminated tuberculosis presenting as meningitis and spondylodiscitis in an immunocompetent adult

Rationale:Disseminated tuberculosis involves the central nervous system in up to a third of cases.However,meningitis and spondylodiscitis due to miliary tuberculosis rarely occur together,particularly in the immuno-competent population.Patient concerns:A 37-year-old immunocompetent male presented with altered level of consciousness for one week and lower back pain with evening pyrexia for one month.Examination revealed spastic paraplegia and left hemiparesis.Diagnosis:Disseminated tuberculosis presenting with meningitis and spondylodiscitis.Interventions:Category I anti-tuberculous therapy with a tapering regimen of intravenous dexamethasone was administered.Outcomes:There was clinical improvement after nine months of treatment.Lessons:Tuberculosis may present with atypical clinical manifestations.Contrast enhanced computed tomography scan or magnetic resonance imaging combined with histopathological features,a high index of suspicion and clinical improvement with anti-tuberculous treatment can confirm the diagnosis in the absence of microbiological evidence,especially in extrapulmonary tuberculosis.

Seasonal Variability of Biofouling Community Structure in the Gulf of Mannar,Southeast Coast of India:A Multivariate Approach

In this research,an Underwater Biofouling Panel(UWBFP)system was erected for the qualitative and quantitative estimation of macro fouling organisms in the Gulf of Mannar.Forty-four biofoulers were identified from four types of selected test panels.Among these biofoulers,Amphibalanus amphitrite(Darwin,1854)was the dominant one.The concrete panel encouraged the highest barnacle density compared to the other panels.Three series of test panels were used to assess the seasonal density of biofouling communities.The overall variation in barnacle count in the seaward and shoreward sides of all these three series were tested.They were found to be significantly different from each other.The greater variations in the barnacle density observed in this study in A-series of test panels could be due to the lack of or absence of other foulers to compete within the fortnight.The Shannon-Wiener species diversity index showed the highest diversity in wood substratum among the three series with greater accumulation of different types of fouling organisms.Multivariate analyses were also performed to understand the seasonal variation as well as the settlement pattern on the different directions of test panels based on validated data.PCA showed a strong variability(PC1 between 70.8%and 98.6%variance)between the directions of the panels in connection with barnacle density.The shade plot and CAP analysis segregated the short-term A-series test panels from the long-term(B-and C-series)test panels.Hence,the output was helpful in understanding the recruitment status of various faunal resources involved in the biofouling processes.

Mesoporous titanium-aluminosilicate as an efficient catalyst for selective oxidation of cyclohexene at mild reaction conditions

Mesoporous titanium containing alumino-silicate materials with various titanium/silicon(Ti/Si) ratio(AlSi-Ti(n);n = Ti/Si mole ratio) have been successfully synthesized by a novel single-step sodium(Na)-free method, for the first time. The obtained characterization results of the prepared materials reveal that in-situ addition of Ti into AlSi shows ordered mesoporous structure along with uniformly dispersed Ti species in +4 and +3 oxidation states suitable for selective oxidation of allylic C—H bond. The prepared mesoporouse Ti-AlSi(n) samples exhibited excellent activity in the oxidation of cyclohexene with 100%conversion and 100% selectivity to ketone-alcohol(KA) oil(cyclohex-2-en-1-ol and 2-cyclohexen-1-one) at low temperature and reaction time(35℃ and 30 min reaction time). This study suggests that AlSi-Ti(0.05) material can be a promising catalyst for the selective oxidation of cyclohexene under mild reaction conditions.

Simulation and Experimental Design of Load Adaptive Braking System on Two Wheeler

The braking quality is considered the main execution of the adaptive control framework that impacts the vehicle safety and rides solace astoundingly notably the stopping distance.This research work aims to create a pattern and design of an electromechanically adjusted lever that multiplies the applied braking force depending on the inputs given by the sensors to reduce the stopping distance of the vehicle.It is carried out using two main parts of the two-wheeler vehicle:thefirst part deals with the detection of load acting on the vehicle and identifying the required braking force to be applied,and the second part deals with the micro-controller which activates the stepper motor for varying the mechanical leverage ratio from various loads on the vehicle using two actively movable wedges.The electromechanically operated variable braking force system is developed to actuate the braking system based on the load on the motorcycle.The MATLAB simulation and experimental work are carried out for various loading(driver and pillion)conditions on a two-wheeler.The results indicate that the proposed electronically operated braking system is more effective than the conventional braking system for various loads and vehicle speeds.Specifically,the stopping distance of the vehicle is decreased significantly by about 4.9%between the con-ventional braking system and the simulated proposed system.Further,the experi-mental results show that the stopping distance is condensed by about 4.1%.The validation between simulated and experimental results revealed a great deal with the least error percentage of about 0.8%.
