Patient-specific induced pluripotent stem cells as“disease-in-adish”models for inherited cardiomyopathies and channelopathies–15 years of research

Among inherited cardiac conditions,a special place is kept by cardiomyopathies(CMPs)and channelopathies(CNPs),which pose a substantial healthcare burden due to the complexity of the therapeutic management and cause early *** other inherited cardiac conditions,genetic CMPs and CNPs exhibit incomplete penetrance and variable expressivity even within carriers of the same pathogenic deoxyribonucleic acid variant,challenging our understanding of the underlying pathogenic *** recently,the lack of accurate physiological preclinical models hindered the investigation of fundamental cellular and molecular *** advent of induced pluripotent stem cell(iPSC)technology,along with advances in gene editing,offered unprecedented opportunities to explore hereditary CMPs and *** features of iPSCs include the ability to differentiate into unlimited numbers of cells from any of the three germ layers,genetic identity with the subject from whom they were derived,and ease of gene editing,all of which were used to generate“disease-in-a-dish”models of monogenic cardiac ***,iPSC-derived cardiomyocytes that faithfully recapitulate the patient-specific phenotype,allowed the study of disease mechanisms in an individual-/allele-specific manner,as well as the customization of therapeutic *** review provides a synopsis of the most important iPSC-based models of CMPs and CNPs and the potential use for modeling disease mechanisms,personalized therapy and deoxyribonucleic acid variant functional annotation.

Introduction: 68Ga-PSMA-11 is considered the gold standard in detection of micro and oligometastases in advanced prostate cancer, being used for therapeutic planning, as well as, potentially, for evaluating response to treatment. The development of ready-to-use lyophilized kit of PSMA-11 adds quality and safety to the routine use of this radiopharmaceutical and represents a pharmacotechnical challenge as it must preserve the integrity and specificity of the ligand. Methods: PSMA-11 kit formulation was proposed, considering radiolabeling parameters and the preservation of the peptide during the lyophilization process, using mannitol as an excipient. Critical temperature characterization studies were carried out using DSC equipment and the freeze-drying process was developed. The direct radiolabeling conditions were evaluated and standardized using 68Ge/68Ga generator eluate from two different manufacturers (ITG and Eckert & Ziegler). The radiochemical purity was evaluated by TLC and HPLC. Biological evaluation was carried out with lyophilized PSMA-11 to demonstrate the integrity of the peptide and preservation of biological activity after the lyophilization process. Results: Based on critical temperature characterization studies, the freeze-drying cycle was designed to reach a freezing temperature of around −40˚C and primary drying at 2˚C. Using 20 mg of mannitol, an intact and elegant lyophilized cake was obtained. PSMA-11 lyophilized kit was directly labeled with 68Ga eluate from 68Ge/68Ga GMP generators (ITG and Eckert & Ziegler) resulting in % RP > 95% at pH 4.0 to 4.5. The results obtained from in vitro and in vivo biological competition studies confirmed the preservation of PSMA-11 affinity for the receptor after lyophilization. Conclusion: A lyophilized formulation (Kit) of PSMA-11 was successfully obtained, which preserved the integrity and biological activity of the peptide and guaranteed radiolabeling efficiency.

Despite significant therapeutic progress in recent years,inflammatory bowel disease(IBD),which includes Crohn’s disease and ulcerative colitis,remains a challenge regarding its pathogenesis and long-term *** concepts have emerged in the management of this disease,such as the"treat-totarget"concept,in which mucosal healing plays a key role in the evolution of IBD,the risk of recurrence and the need for *** is essential for the assessment of mucosal inflammation and plays a pivotal role in the analysis of mucosal healing in patients with *** is also essential in the detection of dysplasia and in the identification of the risk of colon *** current surveillance strategy for dysplasia in IBD patients indicates white-light endoscopy with non-targeted *** new chromoendoscopy techniques provide substantial benefits for both clinicians and ***-band imaging(NBI)has similar rates of dysplastic lesion detection as whitelight endoscopy,and it seems that NBI identifies more adenoma-like *** it is used instinctively by many endoscopists,the combination of these two techniques might improve the rate of dysplasia *** spectral imaging color enhancement can help differentiate dysplastic and non-dysplastic lesions and can also predict the risk of recurrence,which allows us to modulate the treatment to gain better control of the *** combination of noninvasive serum and stool biomarkers with endoscopy will improve the monitoring and limit the evolution of IBD because it enables the use of a personalized approach to each patient based on that patient’s history and risk factors.

Since its introduction into clinical practice 15 years ago,capsule endoscopy(CE)has become the first-line investigation procedure in some small bowel pathologies,and more recently,dedicated esophageal and colon CE have expanded the fields of application to include the upper and lower gastrointestinal *** this time,CE has become increasingly popular among gastroenterologists,with more than 2 million capsule examinations performed worldwide,and nearly 3000Pub Med-listed studies on its different aspects *** huge interest in CE may be explained by its noninvasive nature,patient comfort,safety,and access to anatomical regions unattainable via conventional ***,CE has several limitations which impede its wider clinical applications,including the lack of therapeutic capabilities,inability to obtain biopsies and control its *** research groups are currently working to overcome these limitations,while novel devices able to control capsule movement,obtain high quality images,insufflate the gut lumen,perform chromoendoscopy,biopsy of suspect lesions,or even deliver targeted drugs directly to specific sites are under *** current limitations,especially as some of them have already been successfully surmounted,and based on the tremendous progress in technology,it is expected that,by the end of next 15years,CE able to perform both diagnostic and therapeutic procedures will remain the major form of digestive *** review summarizes the literature that prognosticates about the future developments of CE.

Electrochemical Study and Electrodeposition of Copper（I） in Ionic Liquid-reline

The authors made the electrochemical studies on the electrode processes during copper electrodeposition with ionic liquids-reline as an electrolytic medium. Reline is a commercial name for a eutectic mixture of choline chloride and urea（1：2, molar ratio）. This new proposed method of copper deposition is an ecological one because the ionic liquids are recyclable, non-volatile and non-toxic. Cyclic voltammetry investigations were conducted at 343 and 353 K in choline chloride-urea（Reline） deep eutectic solvent with single salt CuCl（generally in 0.05--0.38 mol/L concentration range）. Electrochemical impedance spectroscopy investigations were conducted in choline chloride- urea ionic liquid with CuCI（0.05 mol/L）. We investigated both cathodic and anodic processes of Cu＋ ions at different sweep rates in this new electrolytic system based on choline chloride. We have proposed by this an alternative to the classical copper electrodeposition. Coefficient of diffusion, Do, for the system Reline-CuCl is presented and the value of D0 is 1.22× 10^-8 cm2/s.

In vitro antioxidant,antilipoxygenase and antimicrobial activities of extracts from seven climbing plants belonging to the Bignoniaceae

Objectives： This study aimed to evaluate the in vitro antioxidant capacity, to determine the anti-inflammatory effect due to lipoxygenase inhibition and to test the antimicrobial activity of ethanolic extracts from leaves of seven climbing species belonging to the Bignoniaceae family. These species are Adenocalyrnrna rnarginatum （Cham.） DC., Amphilophium vauthieri DC., Cuspidaria convoluta （VEIL） A. H. Gentry, Dolichandra dentata （K. Schum.） L. G. Lohmann, Fridericia caudigera （S. Moore） L. G. Lohmann, Fridericia chica （Bonpl.） L. G. Lohmann and Tanaecium selloi （Spreng.） L. G. Lohmann. Methods： The antioxidant activity was evaluated using three methods, 2,2＇-azino-bis（3-ethylbenzothiazo line-6-sulfonic acid）（ABTS）, 2,2-diphenyl-l-picrylhydrazyl （DPPH）, and ferric reducing antioxidant power. Lipoxygenase-inhibiting activity was assayed spectrophotometrically; the result was expressed as percent inhibition. The antimicrobial activity was assessed using the agar disk diffusion method. Minimal inhibitory concentration （MIC） and minimal bactericidal]fungicidal concentration were also determined for each extract against 12 pathogenic bacterial strains of Staphylococcus aureus and seven fungal strains of the Candida genus. The identification of the major compounds present in the most promising extract was established by high-performance liquid chromatography-tandem mass spectrometry. Results： C convoluta, F. caudigera, and F. chica exhibited the best antioxidant activity by scavenging DPPH and ABTS~ radicals and reducing Fe3＋ ion. These extracts showed a notable inhibition of lipnxygenase. F. caudigera was found to have the lower MIC value against S. aureus strains and six Candida species. The extracts of F. caudigera and C. convoluta were active even against methicillin-resistant S. aureus. C convoluta had higher total phenol content, better antioxidant activity and superior anti- inflammatory and antimicrobial activity. The main phenolic compounds found in this extract

Open anterior glenohumeral dislocation with associated supraspinatus avulsion: A case report

BACKGROUND Open dislocation of the glenohumeral joint is a very rare complication with only six cases described in the literature. Most cases resulted in severe complications like avascular necrosis of the humeral head or septic arthritis with poor functional results. The "mandatory" treatment of choice is surgery, with accurate debridement and reconstruction of the damaged soft tissues. However, the results in these cases do not approach those seen in classical *** SUMMARY This case report is the first description of an anterior open glenohumeral dislocation without associated fractures, but with complete avulsion of the soft tissue envelope of the proximal humerus. Surgical treatment consisted of copious lavage with saline solution, meticulous debridement of the nonviable soft tissues,reduction of the dislocated humeral head and reconstruction of the soft tissue envelope of the humeral head. The selected surgical approach was an inferior extension of the wound at the level of the delto-pectoral groove, as the best choice to be able to perform an adequate debridement and capsulo-tendinous reinsertion. At 6 mo there were no clinical signs of instability of the glenohumeral joint, the functionality of the joint was restricted to 90o of anterior elevation,internal rotation to L2, with severe limitation of abduction(60o) and external rotation(0o) but without residual pain, with an Oxford shoulder Score of *** Thorough reconstruction of the soft tissues surrounding the shoulder joint and an early rehabilitation program are key points to a good functional result.

Peripheral nerve injuries （PNI） are a major clinical prob- lem. In general, PNI results from motor vehicle accidents, lacerations with sharp objects, penetrating trauma （gunshot wounds） and stretching or crushing trauma and fractures. It is estimated that PNI occur in 2.8% of trauma patients and this number reaches 5% if plexus and root lesions are in- cluded. However, due to lack of recent epidemiological stud- ies, these data probably underestimate the actual number of nerve injuries
