Optimization of.Nitrogen, Phosphorus and Salt f.r Lipid Accumulation of.Microalgae: Towards the Viability of.Microalgae Biodiesel

In recent years, microalgae biodiesel has attracted expressive attention and investment, once it was considered a potential resource f.r energy. Although the wide use of.microalgae biodiesel is restricted by its high production cost. f.r cost-ef.icient and sustainable production of.biodiesel f.om microalgae, a proper understanding of.the variables and their impacts on physiology of.the strains is required. In this study, a simple f.ctorial design 23 was used to f.nd optimal conditions f.r the cultivation of.Ankistrodesmus sp. and Chlamydomonas sp. in batch culture. The three components considered were nitrate, phosphate and sodium chloride, used to assess the metabolic versatility of.the strains in brackish conditions. The results showed that culture medium with 0.04 g·L?1 nitrate, 0.01 g·L?1 phosphate and 5.0 g·L?1 sodium chloride resulted to be the most ef.ective condition to growth and f.tty acids accumulation. Using this optimal condition, Ankistrodesmus sp. and Chlamydomonas sp. increased in 2.1 and 2.4 f.lds their f.tty acids yield, respectively. Importantly, this protocol reduced 75% of.the nitrate and phosphate concentrations of.the original medium (ASM-1). Additionally, f.tty acids analysis f.und that these strains were mainly constituted of.C16-C18, in accordance with the requirements f.r biodiesel production. The simple f.ctorial design applied here proved to be an important tool towards a better understanding of.synergistic ef.ects of.tested f.ctors on microalgae metabolism, and the resulting inf.rmation could be used ef.ectively to improve microalgae cultivation.

Non-Protein-Coding RNAs and their Interacting RNA-Binding Proteins in the Plant Cell Nucleus

The complex responses of.eukaryotic cells to external f.ctors are governed by several transcriptional and post-transcriptional processes. Several of.them occur in the nucleus and have been linked to the action of.non-proteincoding RNAs （or npcRNAs）, both long and small npcRNAs, that recently emerged as major regulators of.gene expression. Regulatory npcRNAs acting in the nucleus include silencing-related RNAs, intergenic npcRNAs, natural antisense RNAs, and other aberrant RNAs resulting f.om the interplay between global transcription and RNA processing activities （such as Dicers and RNA-dependent polymerases）. Generally, the resulting npcRNAs exert their regulatory ef.ects through interactions with RNA-binding proteins （or RBPs） within ribonucleoprotein particles （or RNPs）. A large group of.RBPs are implicated in the silencing machinery through small interf.ring RNAs （siRNAs） and their localization suggests that several act in the nucleus to trigger epigenetic and chromatin changes at a whole-genome scale. Other nuclear RBPs interact with npcRNAs and change their localization. In the f.ssion yeast, the RNA-binding Mei2p protein, playing pivotal roles in meiosis, interact with a meiotic npcRNA involved in its nuclear re-localization. Related processes have been identif.ed in plants and the ENOD40 npcRNA was shown to re-localize a nuclear-speckle RBP f.om the nucleus to the cytoplasm in Medicago truncatula. Plant RBPs have been also implicated in RNA-mediated chromatin silencing in the f.C locus through interaction with specif.c antisense transcripts. In this review, we discuss the interactions between RBPs and npcRNAs in the context of.nuclear-related processes and their implication in plant development and stress responses. We propose that these interactions may add a regulatory layer that modulates the interactions between the nuclear genome and the environment and, consequently, control plant developmental plasticity.

Adsorption Behavior of.Reducing End-Modif.ed Cellulose Nanocrystals:A Kinetic Study Using Quartz Crystal Microbalance

In this work,we studied the adsorption of.modif.ed cellulose nanocrystals onto solid surf.ces by quartz crystal microbalance with dissipation monitoring(QCM-D).Cellulose nanocrystals obtained f.om tunicate(CNC)were modif.ed at reducing end by amidation *** dif.erent f.nctionalities were investigated:a polyamine dendrimer(CNC-NH_(2)),which interacts with gold surf.ce by the amine groups;and a biotin moiety(CNC-Biot),which has a strong af.inity f.r the protein streptavidin(SAV).QCM-D results revealed dif.erent adsorption behaviors between modif.ed and unmodif.ed ***,unmodif.ed CNCs covered almost all the surf.ce f.rming a rigid and f.at layer whereas reducing end modif.ed CNCs remained rather upright f.rming a hydrated and viscoelastic layer with lower surf.ce *** analysis of.adsorption kinetics allowed the calculation of.an apparent collision rate f.ctor,which resulted 10-f.ld higher f.r unmodif.ed CNCs compared to reducing end modif.ed CNCs,theref.re,demonstrating the dif.erent adsorption behavior.

DNA damage levels in systemic lupus erythematosus patients with low disease activity:An evaluation by comet assay

Oxidative stress has been implicated in the inf.ammatory process of.Systemic Lupus Erythematosus (SLE), particularly by the f.rmation of.anti-DNA autoantibodies, which can lead to DNA damage. The aim of.this study was to investigate, through comet assay, whether the level of.DNA damage in SLE patients is dif.erent f.om that of.healthy subjects. Twenty-f.ve adult SLE patients with SLEDAI up to ten, and 25 healthy subjects were paired according to age, gender and Body Mass Index (BMI). Other anthropometric variables were also assessed. Comet assay was assessed as the marker of.oxidative stress described as DNA Damage (DD) percentage. Waist Circumf.rence (WC), Hip Circumf.rence (HC) and BMI were also perf.rmed. Exclusion criteria f.r patients and controls comprised smoking and other chronic disorders. Level of.damage index was remarkably higher in SLE patients than in controls, and no signif.cant dif.erences between the groups were f.und f.r age, BMI, WC and HC. No stratif.cation concerning gender was perf.rmed, since there were just two males per group. No correlation was observed between BMI and DD (%). DD increased in SLE, which ref.ects the oxidant/antioxidant imbalance in these patients. These f.ndings support an association between oxidative stress and SLE. This stronger correlation observed in patients with low disease activity may be usef.l in elucidating the mechanisms of.disease pathogenesis.

Cof.ee is currently the second largest commodity on the world market today,and there is great concern about the quality of.the beans exported f.om producer countries to Europe and *** such as using blends of.dif.erent species and adding low-cost raw materials,such as chicory,corn,and soybean,impair the sensory and f.nctional characteristics of.the drink made f.om roasted and ground cof.ee *** is a need to adopt more ef.icient analytical methods than the microscopy technique currently *** f.rst chromatographic method used to determine f.aud was reported in *** method used paper chromatography to dif.erentiate between cof.ee and chicory based on the f.ee reducing *** of.the 1980s,dif.erent methods involving high-perf.rmance liquid chromatography and gas chromatography were developed in order to demonstrate geographic authenticity,distinction between species,occurrence of.adulteration,and the presence of.def.ctive beans by determining the monosaccharides,oligosaccharides,tocopherols,f.tty acids,volatiles,diterpenes,sterols,and phenolic substances,among *** f.r as the authors know,there are no papers published in the literature that have compiled such an extensive set of.inf.rmation about these chromatographic methods as *** the last 2 years,there has been a trend to develop analytical methods f.r ultra-perf.rmance liquid chromatography coupled with tandem mass spectrometry to conf.rm f.aud in cof.ee,due to high sensitivity and selectivity.
