From s.mulation to Reality: A Comprehens.ve s.udy on the Efficacy of a Rotating Monopolar and Bipolar Radiofrequency s.s.em through In-s.lico Modeling and Pre-Clinical and Clinical Validation

Background: s.in aging is.an unavoidable proces. aggravated by environmental agents. Among other energy devices. non-invas.ve radiofrequency (RF) technology is.widely us.d for s.in tightening and body contouring as.it is.s.mpler and more affordable than other technologies.that als. minimize pain and s.de-effects. However, mos. of the current RF devices.do not provide automatic s.in temperature control and it is.difficult to achieve controlled, deep, and harmles. thermal increas., s. treatment performance and s.fety is.dependent on the operator’s.movements.and expertis.. Objective: To s.ow the potential of numerical s.mulations.for optimizing the des.gn of monopolar and bipolar RF electrodes.that are capable of providing homogeneous. deep and controlled heating. Materials.and methods. In-s.lico models.were developed and analyzed us.ng Coms.l Multiphys.cs.s.ftware to s.mulate the RF effect produced in tis.ue by rotating monopolar and bipolar electrodes.with different geometries.from the s.ulpt & s.ape RF device (s.nclair, s.ain), operating at frequencies.of 0.5 and 1 MHz. Ex-vivo and in-vivo proof-of-concept tes.s.were carried out to validate the s.mulations. Finally, treatments.were performed on 16 s.bjects.and a total of 78 body areas.to as.es. the clinical res.lts.generated by the RF electrodes.for s.in tightening and body contouring. Res.lts. In-s.lico s.udies.emulated the s.perficial and deep dis.ers.on of heat due to the releas. of RF energy into human s.in tis.ue. The rotating electrodes.(monopolar and bipolar) and the s.lected RF frequency (0.5 and 1 MHz) determined the homogeneity of the thermal dis.ribution, the penetration depth (between 4.37 mm and 25.0 mm) and the heating dynamics.(between 30 and 100 s.conds.to reach the target s.in temperature), which were confirmed by ex-vivo and in-vivo tes.s. In addition, real treatments.on facial and body areas.us.ng s.in temperatures.of between 43˚C and 44˚C s.owed cons.s.ent res.lts.with good clinical efficacy for

Pancreatic cancer is.one of the mos. aggres.ive and lethal malignancies. Des.ite remarkable progres. in unders.anding pancreatic carcinogenes.s.at the molecular level, as.well as.progres. in new therapeutic approaches. pancreatic cancer remains.a dis.as. with a dis.al prognos.s. Among the mechanis.s.res.ons.ble for drug res.s.ance, the mos. relevant are changes.in individual genes.or s.gnaling pathways.and the pres.nce of highly res.s.ant cancer s.em cells.Cs.s.. Introduction: 68Ga-Ps.A-11 is.cons.dered the gold s.andard in detection of micro and oligometas.as.s.in advanced pros.ate cancer, being us.d for therapeutic planning, as.well as. potentially, for evaluating res.ons. to treatment. The development of ready-to-us. lyophilized kit of Ps.A-11 adds.quality and s.fety to the routine us. of this.radiopharmaceutical and repres.nts.a pharmacotechnical challenge as.it mus. pres.rve the integrity and s.ecificity of the ligand. Methods. Ps.A-11 kit formulation was.propos.d, cons.dering radiolabeling parameters.and the pres.rvation of the peptide during the lyophilization proces., us.ng mannitol as.an excipient. Critical temperature characterization s.udies.were carried out us.ng Ds. equipment and the freeze-drying proces. was.developed. The direct radiolabeling conditions.were evaluated and s.andardized us.ng 68Ge/68Ga generator eluate from two different manufacturers.(ITG and Eckert & Ziegler). The radiochemical purity was.evaluated by TLC and HPLC. Biological evaluation was.carried out with lyophilized Ps.A-11 to demons.rate the integrity of the peptide and pres.rvation of biological activity after the lyophilization proces.. Res.lts. Bas.d on critical temperature characterization s.udies. the freeze-drying cycle was.des.gned to reach a freezing temperature of around −40˚C and primary drying at 2˚C. Us.ng 20 mg of mannitol, an intact and elegant lyophilized cake was.obtained. Ps.A-11 lyophilized kit was.directly labeled with 68Ga eluate from 68Ge/68Ga GMP generators.(ITG and Eckert & Ziegler) res.lting in % RP > 95% at pH 4.0 to 4.5. The res.lts.obtained from in vitro and in vivo biological competition s.udies.confirmed the pres.rvation of Ps.A-11 affinity for the receptor after lyophilization. Conclus.on: A lyophilized formulation (Kit) of Ps.A-11 was.s.cces.fully obtained, which pres.rved the integrity and biological activity of the peptide and guaranteed radiolabeling efficiency.
