Influence of fibre and betaine on development of the gastrointestinal tract of broilers between hatch and 14 d of age

An experiment was designed to determine the influence of fibre and betaine on the development of the intestine, liver and pancreas of broilers from hatch to 14 d of age. A total of 250-day-old Cobb 500 male broilers were allocated to 16 cages with 15 broilers each. Treatments were arranged in a 2 x 4 factorial design, consisting of 2 feed formulations(low and high fibre diets) and 4 levels of betaine(0,1, 3 or 5 kg/t). At hatch, 10 birds in total were euthanised, and samples of the liver, pancreas, yolk sac and intestine were collected for reference of the analysed parameters before the start of the *** d 4.9 and 14, 5 birds per cage(10 birds per treatment) were selected, euthanised and treated as the same as the birds at hatch. villus height and width and crypt depth were determined on the duodenum samples, and absorptive area was calculated. The number of enterocytes in mitosis at the villus was determined by a positive reaction to antibody for Ki67 protein, and fused villus was evaluated visually. The relative weight of the yolk sac reduced(P ver reached a maximum(P ve weight of the pancreas and higher villus(P villus width increased(P villus. Betaine inclusion reduced(P villus width, increased(P villus size and absorptive area, and reduced(P ve reaction for the antibody *** inclusion reduced the width and increased the absorptive area and the villus height of the duodenum of birds up to 14 d of age. The higher fibre diet increased feed intake and villus height, yet reduced pancreas relative weight, while not affecting body weight gain. This response was possibly due to a dilution effect of the fibre, reducing nutrient absorption

Coffee is currently the second largest commodity on the world market today,and there is great concern about the quality of the beans exported from producer countries to Europe and *** such as using blends of different species and adding low-cost raw materials,such as chicory,corn,and soybean,impair the sensory and functional characteristics of the drink made from roasted and ground coffee *** is a need to adopt more efficient analytical methods than the microscopy technique currently *** first chromatographic method used to determine fraud was reported in *** method used paper chromatography to differentiate between coffee and chicory based on the free reducing *** of the 1980s,different methods involving high-performance liquid chromatography and gas chromatography were developed in order to demonstrate geographic authenticity,distinction between species,occurrence of adulteration,and the presence of defective beans by determining the monosaccharides,oligosaccharides,tocopherols,fatty acids,volatiles,diterpenes,sterols,and phenolic substances,among *** far as the authors know,there are no papers published in the literature that have compiled such an extensive set of information about these chromatographic methods as *** the last 2 years,there has been a trend to develop analytical methods for ultra-performance liquid chromatography coupled with tandem mass spectrometry to confirm fraud in coffee,due to high sensitivity and selectivity.

Background:Leishmaniasis causes alterations and lesions in the genital system,which leads to azoospermia and testicular atrophy in animals during the chronic phase of the *** aim of this study was to reveal the kinetics of Leishmania chagasi infection in the genital system of male golden hamsters(Mesocricetus auratus).Methods:Animals were intraperitoneally inoculated with amastigotes from *** different time points animals were euthanized and genital organs processed for histo-pathological,qPCR,cytokines and testosterone detection ***:Our results showed a high parasite load in testis,followed by an increase of pro-inflammatory cytokines IL1-β,TNF-αand IFN-γ,and ***,IL-4 expression was upregulated and basal parasite persistence in testis was observed using the experimental ***:Extracellular amastigotes migrated to the epididymis posing as a potential major factor of parasite persistence and venereal transmission of *** infection in hamsters.

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori *** bacterium promotes the production of reactive oxygen species(ROS),which cause DNA damage in gastric epithelial *** this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATM,ATR,and H2AX)and ROS-induced damage repair(APE1)and the expression of some miRNAs(miR-15a,miR-21,miR-24,miR-421 and miR-605)that target genes involved in the DNA damage response(DDR)in 31 fresh tissues of gastric *** v3.1.1 was used to construct the postulated miRNA:mRNA interaction *** performed by real-time quantitative PCR exhibited significantly increased levels of the APE1(RQ=2.55,pvated levels of miR-21,miR-24 and miR-421 were observed in diffuse-type gastric *** analysis reinforced some of the gene:gene(ATM/ATR/H2AX)and miRNA:mRNA relationships obtained also with the interaction ***,our findings show that tumor cells from gastric cancer presents deregulation of genes and miRNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tumor microenvironment.

Temperature Gradient Gel Electrophoresis as a valuable Accessory Tool for Assessment of Dysbiosis in Crohn’s Disease

Escherichia coli and other Proteobacteria are augmented and several other bacteria are diminished in Crohn’s (CD) disease patients’ intestine. This imbalance in bacterial species composition—termed dysbiosis—seems to be determinant of CD manifestation. Since a great part of intestinal bacteria are not cultivable, detection of CD dysbiosis is accomplished by molecular tools, involving sequences analysis of the 16SrRNA gene (16SrDNA) present in the patient’s clinical samples, which can be done by sequencing or electrophoresis in denaturing gels of 16SrDNA amplicons. By analyzing, by temperature gradient gel electrophoresis (TGGE) and next generation sequencing of 16Sv6-v8rDNA amplicons present in gram negative cultures from four distinct clinical samples of a control subject and a CD patient, this study demonstrates that both techniques were able to detect E. coli overgrowth and reduction in species richness in CD and that TGGE can discriminate sequences collectively labeled as “unclassified” in 16SrDNA databases. Although TGGE per se does not identify the sequences, the discriminatory power that it confers represents valuable accessory information to next generation DNA sequencing (NGS), and as such must be used as a NGS complementary tool.

The disposal of mining tailings has increasingly focused on the use of dry *** structures offer more security since they use filtered and compacted *** of the construction method and the heights achieved,the material that compounds the structure can be subjected to different stress paths along the failure *** theoretical framework considered in the design of these structures generally is the critical state soil mechanics(CSSM).However,the data in the literature concerning the uniqueness of critical state line(CSL)is still controversial as the soil is subjected to different stress *** respect to tailings,this question is even more *** paper studies two tailings with different gradings due to the beneficial processes over extension and compression paths.A series of drained and undrained triaxial tests was conducted over a range of initial densities and stress *** the q-p'plane,different critical stress ratio(M)values were obtained for compression and extension stress ***,the critical state friction angle is very similar with a slightly higher critical state friction angle for extension *** stress path dependent CSLs were obtained in the n-lnp0 plane with the extension tests below the CSL defined in *** the fines content,the studied tailings presented very similar M and critical state friction angle ***,the fines content af-fects the volumetric behavior of the studied tailings and the CSLs on the n-lnp0 plane shift downwards with the increasing fines content for compression and extension *** relation to dilatancy analysis,the fines content did not present an evident influence on the dilatancy of the ***,different values of mean stress ratio N were obtained between compression and extension tests and can corroborate the existence of non-unique CSLs for these materials.

Detection and quantitation of host cell proteins in monoclonal antibody drug products using automated sample preparation and data-independent acquisition LC-MS/MS

With the size of the biopharmaceutical market exponentially increasing,there is an aligned growth in the importance of data-rich analyses,not only to assess drug product safety but also to assist drug development driven by the deeper understanding of structure/function *** monoclonal antibodies,many functions are regulated by N-glycans present in the constant region of the heavy chains and their mechanisms of action are not completely *** importance of their function focuses analytical research efforts on the development of robust,accurate and fast methods to support drug development and quality *** N-glycan analysis is considered as the gold standard for glycosylation characterisation;however,it is not the only method for quantitative analysis of glycoform *** this study,ten different analytical workflows for N-glycan analysis were compared using four monoclonal *** observing good comparability between the quantitative results generated,it was possible to appreciate the advantages and disadvantages of each technique and to summarise all the observations to guide the choice of the most appropriate analytical workflow according to application and the desired depth of data generated.

Correction of Acidity of a Brazilian Cerrado Oxisol with Limestone and Wood Ash on the Initial Growth of Cowpea

Among Brazilian soils orders, there are some of it classified as acid soils, which need correction to guarantee crop productivity. Currently, limestone is the most used soil corrective and wood ash has been a sustainable alternative to this process. Thus, the objective was to evaluate the effect of two correctives of soil acidity on an Oxisol collected in the Brazilian Cerrado area using limestone and wood ash and the effect of soil correction on initial growth of cowpea. Two greenhouse experiments were carried out: 1) with limestone, using base saturation levels (v%) of 0, 20, 40, 60 and 80;and 2) wood ash, with doses of 0, 8, 16, 24 and 32 g⋅dm−3, both in five randomized blocks. At 40 days after sowing, it was verified that pH values were within the range considered ideal for soil (pH of 5 to 7), according to the increase of base saturation levels and wood ash doses, but limestone provided faster results. Initial growth of cowpea was positively influenced by soil correction with use of both correctives. Wood ash and limestone increased soil pH to adequate values and resulted in better initial crop development.
