Influence of fibre and betaine on development of the gastrointestinal tract of broilers between hatch and 14 d of age

An experiment was designed to determine the influence of fibre and betaine on the development of the intestine, liver and pancreas of broilers from hatch to 14 d of age. A total of 250-day-old Cobb 500 male broilers were allocated to 16 cages with 15 broilers each. Treatments were arranged in a 2 x 4 factorial design, consisting of 2 feed formulations(low and high fibre diets) and 4 levels of betaine(0,1, 3 or 5 kg/t). At hatch, 10 birds in total were euthanised, and samples of the liver, pancreas, yolk sac and intestine were collected for reference of the analysed parameters before the start of the trial.On d 4.9 and 14, 5 birds per cage(10 birds per treatment) were selected, euthanised and treated as the same as the birds at hatch. villus height and width and crypt depth were determined on the duodenum samples, and absorptive area was calculated. The number of enterocytes in mitosis at the villus was determined by a positive reaction to antibody for Ki67 protein, and fused villus was evaluated visually. The relative weight of the yolk sac reduced(P ver reached a maximum(P ve weight of the pancreas and higher villus(P villus width increased(P villus. Betaine inclusion reduced(P villus width, increased(P villus size and absorptive area, and reduced(P ve reaction for the antibody Ki-67.Betaine inclusion reduced the width and increased the absorptive area and the villus height of the duodenum of birds up to 14 d of age. The higher fibre diet increased feed intake and villus height, yet reduced pancreas relative weight, while not affecting body weight gain. The disposal of mining tailings has increasingly focused on the use of dry stacks.These structures offer more security since they use filtered and compacted material.Because of the construction method and the heights achieved,the material that compounds the structure can be subjected to different stress paths along the failure plane.The theoretical framework considered in the design of these structures generally is the critical state soil mechanics(CSSM).However,the data in the literature concerning the uniqueness of critical state line(CSL)is still controversial as the soil is subjected to different stress paths.With respect to tailings,this question is even more restricted.This paper studies two tailings with different gradings due to the beneficial processes over extension and compression paths.A series of drained and undrained triaxial tests was conducted over a range of initial densities and stress levels.In the q-p'plane,different critical stress ratio(M)values were obtained for compression and extension stress paths.However,the critical state friction angle is very similar with a slightly higher critical state friction angle for extension tests.Curved stress path dependent CSLs were obtained in the n-lnp0 plane with the extension tests below the CSL defined in compression.Regarding the fines content,the studied tailings presented very similar M and critical state friction angle values.However,the fines content af-fects the volumetric behavior of the studied tailings and the CSLs on the n-lnp0 plane shift downwards with the increasing fines content for compression and extension tests.In relation to dilatancy analysis,the fines content did not present an evident influence on the dilatancy of the materials.However,different values of mean stress ratio N were obtained between compression and extension tests and can corroborate the existence of non-unique CSLs for these materials.

【目的】研究中国咖啡主栽区咖啡种质资源圃中由专性寄生病原真菌咖啡驼孢锈菌(Hemileia vastatrix Berk.et Br.)引起的咖啡黄锈病(coffee leaf rust,CLR)致病类型,对保护中国咖啡种质资源以及指导主栽区抗性品种的布局和选育具有重要意义。【方法】收集分布在中国咖啡主栽区4个具有代表性的咖啡种质资源圃CLR病样,在超感病基因型寄主上进行扩繁及单孢分离,在特定基因型寄主上对咖啡驼孢锈菌的毒力基因进行鉴定,并推导出相应的致病类型。【结果】57份样品中,有42份样品(占73.68%)被成功扩繁,有27份样品(占64.29%)被保存。根据样品信息录入23个CIFC条目,共鉴定出3种致病类型:XXXⅦ(v_(2,5,6,7,9)),占86.96%,为优势致病类型;New race(v_(2,5,6,7)),占8.69%,为在中国种质资源圃CLR调查的首次报道;XXXIv(v_(2,5,7,9)),占4.35%。【结论】中国咖啡主栽区咖啡驼孢锈菌群体遗传结构正在发生改变,原有致病类型已渗入位于主栽区内的种质资源圃,并在基因型相近的寄主上加速进化;新的致病类型在主栽区内的咖啡基地和种质资源圃之间随机流动,并对当前主栽品种的抗性造成威胁。研究结果将为中国咖啡种质资源圃的科学管理提供重要依据,为CLR防控提供理论支持。

Objective::Prenatal infection with the Zika virus (ZIKv) can lead to congenital Zika syndrome (CZS), characterized by microcephaly and brain injury. However, there are questions regarding the prevalence of microcephaly/CZS after the ZIKv outbreak in defined geographic areas. This study aimed to identify adverse outcomes in live births of fetuses exposed in utero to the ZIKv, compared to unexposed births, as well as maternal sociodemographic, delivery, and birth characteristics.Methods::Here, we conducted a cross-sectional observational study to investigate the characteristics of all live births in the city of Tangará da Serra, Mato Grosso, Brazil, in 2016, after the outbreak of ZIKv infection in late 2015. All live births of children to women residing in the municipality of Tangará da Serra between January 1 and December 31, 2016, were evaluated, and head circumference was measured at birth and after 24 hours. Children born with microcephaly or a maternal history of confirmed or suspected prenatal ZIKv infection were evaluated by a multidisciplinary team. The outcomes of the exposed and non-exposed children were compared. Prevalence ratios and their respective 95% confidence intervals were calculated for sociodemographic, delivery, and live birth characteristics.Results::Of 1,441 live births, 106 (7.3%) were from mothers with confirmed or highly probable exposure to ZIKv. The prevalence of severe congenital microcephaly (41.7/10,000) in Tangará da Serra in 2016 was ten-fold higher than that in Latin America before 2015.Conclusion::This study may serve as a model to investigate possible outbreaks of infections in a defined geographical space in the future.

Upregulation of the APE1 and H2AX genes and miRNAs involved in DNA damage response and repair in gastric cancer

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori infection.This bacterium promotes the production of reactive oxygen species(ROS),which cause DNA damage in gastric epithelial cells.In this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATM,ATR,and H2AX)and ROS-induced damage repair(APE1)and the expression of some miRNAs(miR-15a,miR-21,miR-24,miR-421 and miR-605)that target genes involved in the DNA damage response(DDR)in 31 fresh tissues of gastric cancer.Cytoscape v3.1.1 was used to construct the postulated miRNA:mRNA interaction network.analysis performed by real-time quantitative PCR exhibited significantly increased levels of the APE1(RQ=2.55,pvated levels of miR-21,miR-24 and miR-421 were observed in diffuse-type gastric cancer.Correlation analysis reinforced some of the gene:gene(ATM/ATR/H2AX)and miRNA:mRNA relationships obtained also with the interaction network.Thus,our findings show that tumor cells from gastric cancer presents deregulation of genes and miRNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tumor microenvironment.

Background:Leishmaniasis causes alterations and lesions in the genital system,which leads to azoospermia and testicular atrophy in animals during the chronic phase of the infection.The aim of this study was to reveal the kinetics of Leishmania chagasi infection in the genital system of male golden hamsters(Mesocricetus auratus).Methods:Animals were intraperitoneally inoculated with amastigotes from L.chagasi.At different time points animals were euthanized and genital organs processed for histo-pathological,qPCR,cytokines and testosterone detection assays.Results:Our results showed a high parasite load in testis,followed by an increase of pro-inflammatory cytokines IL1-β,TNF-αand IFN-γ,and testosterone.Subsequently,IL-4 expression was upregulated and basal parasite persistence in testis was observed using the experimental approach.Conclusion:Extracellular amastigotes migrated to the epididymis posing as a potential major factor of parasite persistence and venereal transmission of L.chagasi infection in hamsters.

Coffee is currently the second largest commodity on the world market today,and there is great concern about the quality of the beans exported from producer countries to Europe and USA.Practices such as using blends of different species and adding low-cost raw materials,such as chicory,corn,and soybean,impair the sensory and functional characteristics of the drink made from roasted and ground coffee beans.There is a need to adopt more efficient analytical methods than the microscopy technique currently used.The first chromatographic method used to determine fraud was reported in 1958.This method used paper chromatography to differentiate between coffee and chicory based on the free reducing sugars.As of the 1980s,different methods involving high-performance liquid chromatography and gas chromatography were developed in order to demonstrate geographic authenticity,distinction between species,occurrence of adulteration,and the presence of defective beans by determining the monosaccharides,oligosaccharides,tocopherols,fatty acids,volatiles,diterpenes,sterols,and phenolic substances,among others.As far as the authors know,there are no papers published in the literature that have compiled such an extensive set of information about these chromatographic methods as here.Over the last 2 years,there has been a trend to develop analytical methods for ultra-performance liquid chromatography coupled with tandem mass spectrometry to confirm fraud in coffee,due to high sensitivity and selectivity.

Temperature Gradient Gel Electrophoresis as a valuable Accessory Tool for Assessment of Dysbiosis in Crohn’s Disease

Escherichia coli and other Proteobacteria are augmented and several other bacteria are diminished in Crohn’s (CD) disease patients’ intestine. This imbalance in bacterial species composition—termed dysbiosis—seems to be determinant of CD manifestation. Since a great part of intestinal bacteria are not cultivable, detection of CD dysbiosis is accomplished by molecular tools, involving sequences analysis of the 16SrRNA gene (16SrDNA) present in the patient’s clinical samples, which can be done by sequencing or electrophoresis in denaturing gels of 16SrDNA amplicons. By analyzing, by temperature gradient gel electrophoresis (TGGE) and next generation sequencing of 16Sv6-v8rDNA amplicons present in gram negative cultures from four distinct clinical samples of a control subject and a CD patient, this study demonstrates that both techniques were able to detect E. coli overgrowth and reduction in species richness in CD and that TGGE can discriminate sequences collectively labeled as “unclassified” in 16SrDNA databases. Although TGGE per se does not identify the sequences, the discriminatory power that it confers represents valuable accessory information to next generation DNA sequencing (NGS), and as such must be used as a NGS complementary tool.

Up-regulation of tumor necrosis factor-a pathway survival genes and of the receptor TNFR2 in gastric cancer

BACKGROUND Gastric carcinogenesis can be induced by chronic inflammation triggered by Helicobacter pylori(H. pylori) infection. Tumor necrosis factor(TNF)-α and its receptors(TNFR1 and TNFR2) regulate important cellular processes, such as apoptosis and cell survival, and the disruption of which can lead to cancer. This signaling pathway is also modulated by microRNAs(miRNAs), altering gene expression.AIM To evaluate the mRNA and miRNAs expression involved in the TNF-α signaling pathway in gastric cancer(GC) tissues and its relationship.METHODS Quantitative polymerase chain reaction(qPCR) by TaqMan? assay was used to quantify the RNA transcript levels of TNF-α signaling pathway(TNF, TNFR1,TNFR2, TRADD, TRAF2, CFLIP, NFKB1, NFKB2, CASP8, CASP3) and miRNAs that targets genes from this pathway(miR-19 a, miR-34 a, miR-103 a, miR-130 a,miR-181 c) in 30 GC fresh tissue samples. Molecular diagnosis of H. pylori was performed by nested PCR for gene HSP60. A miRNA:mRNA interaction network was construct using Cytoscape v3.1.1 from the in silico analysis performed using public databases.RESULTS Up-regulation of cellular survival genes as TNF, TNFR2, TRADD, TRAF2, CFLIP,and NFKB2, besides CASP8 and miR-34 a was observed in GC tissues, whereas mediators of apoptosis such as TNFR1 and CASP3 were down-regulated. When the samples were stratified by histological type, the expression of miR-103 a and miR-130 a was significantly increased in the diffuse-type of GC compared to the intestinal-type. However, no influence of H. pylori infection was observed on the expression levels of mRNA and miRNAs analyzed. Moreover, the miRNA:mRNA interaction network showed several interrelations between the miRNAs and their target genes, highlighting miR-19 a and miR-103 a, which has as predicted or validated target a large number of genes in the TNF-α pathway,including TNF, TNFR1, TNFR2, CFLIP, TRADD, CASP3 and CASP8.CONCLUSION Our findings show that cell survival genes mediated by TNF/TNFR2 binding is up-regulated in GC favoring its pro-tumoral e

Ensuring the removal of host cell proteins(HCPs) during downstream processing of recombinant proteins such as monoclonal antibodies(m Abs) remains a challenge.Since residual HCPs might affect product stability or safety,constant monitoring is required to demonstrate their removal to be below the regulatory accepted level of 100 ng/mg.The current standard analytical approach for this procedure is based on ELISA;however,this approach only measures the overall HCP content.Therefore,the use of orthogonal methods,such as liquid chromatography-mass spectrometry(LC-MS),has been established,as it facilitates the quantitation of total HCPs as well as the identification and quantitation of the individual HCPs present.In the present study,a workflow for HCP detection and quantitation using an automated magnetic bead-based sample preparation,in combination with a data-independent acquisition(DIA) LC-MS analysis,was established.Employing the same instrumental setup commonly used for peptide mapping analysis of m Abs allows for its quick and easy implementation into pre-existing workflows,avoiding the need for dedicated instrumentation or personnel.Thereby,quantitation of HCPs over a broad dynamic range was enabled to allow monitoring of problematic HCPs or to track changes upon altered bioprocessing conditions.
