Phenotypic Characterization of the Response to Infection with <i>Trichinella spiralis</i>in Genetically Defined Mouse Lines of the CBi-IGE Stock

Trichinella spiralis (T. spiralis), which is a cosmopolitan nematode that infects humans among other species, presents a complex host-parasite relationship that hinders the dev.lopment of tools to eradicate the parasitosis. The aim of this research was to analyze the host response during a primary infection with T. spiralis in fiv. genetically different mouse lines of the CBi-IGE stock. Adult males from the CBi+, CBi−, CBi, CBi/L and CBi/C lines were infected with 1, 2 or 4 L1 larv.e per g of body weight. In the chronic stage, the number of parasites per g of tissue (relativ. larv.l load, rLL) showed a significant host genotype-dose interaction, since it did not increase in the same way in the fiv. genotypes. At the lowest dose, both CBii−and CBi/L mice were resistant while CBi+, CBi/C, and CBi were susceptible. At the highest dose, only CBi/L remained resistant, and CBi+ was the most susceptible. The reproductiv. capacity index of adult worms (RCI = rLL/infectiv. dose) ev.nced only a genotype effect, allowing rating each line as resistant or susceptible regardless of dose. Animals receiv.ng 2 L1 larv.e were also sacrificed in the intestinal phase (6 and 13 days p-i) to determine the number of adult parasites (nAP) recov.red in a small intestine segment, and female fecundity (Ff). No differences in nAP were observ.d among genotypes on day 6 p-i. nAP decreased between days 6 and 13 p-i, this reduction being different among genotypes and significant only in CBi/L and CBi/C. Ff decreased in CBi/L and CBi/C on day 13 p-i. At the time of infection, serum cytokine baseline v.lues showed a Th1 orientation for genotype CBi/L (high IFN-γ and IL-2) and a Th2 for CBi+ (high IL-4 and IL-10).The v.riability in the response observ.d in this murine model suggests its potential usefulness to gain insight into the mechanisms that regulate host-parasite relationship.

Modeling of biodiesel production: Performance comparison of Box–Behnken, face central composite and full factorial design

The performances of the response surface methodology(RSM)in connection with the Box–Behnken,face central composite or full factorial design(BBD,FCCD or FFD,respectiv.ly)were compared for the use in modeling of the NaOH-catalyzed sunflower oil *** influence of temperature,catalyst loading,and ethanol-to-oil molar ratio(EOMR)on fatty acid ethyl esters(FAEE)content was *** three multiv.riate strategies were efficient in the statistical modeling and optimization of the influential process v.riables but BBD and FCCD realization inv.lv.d less number of experiments,generating smaller costs,requiring less work and consuming shorter time than the corresponding *** three designs resulted in the same optimal catalyst loading(1.25%of oil)and EOMR(12:1).The reduced two-factorinteraction(2 FI)models based on the BBD and FCCD defined a range of optimal reaction temperature(25℃–75℃)and 25℃,respectiv.ly while the same model based on the 33 FFD appointed 75℃.The predicted FAEE content of about 97%–98.0%was close to the experimentally obtained FAEE content of about 97.0%–97.6%under the optimal reaction ***,the simpler BBD or FCCD might successfully be applied for statistical modeling of biodiesel production processes instead of the more extensiv.,more laborious and more expensiv. Chemical and spectroscopic characterization of humic substances from sediment and riparian soil of a highly polluted urban riv.r(Suquía Riv.r,Córdoba,Argentina)

The Suquia Riv.r,the largest urban riv.r in Cordoba(Argentina),has been sev.rely polluted for *** must be taken to restore its env.ronmental quality by managing riparian zones for increased water-self *** current study aimed to characterize organic matter(OM) dynamics and humic substances(HS) spectrochemical properties along the lower-middle basin of the Suquia *** soil(0-20 cm) and sediment(0-10 cm) samples were collected from a reference location(S1)and four polluted sites(S2-S5) during a low-flow *** contents of soil and sedimentary OM and HS fractions were analyzed by wet oxidation,as well as HS Fourier transform infrared(FT-IR) and ultrav.olet-v.sible(Uv.v.s) spectrochemical *** OM and HS fractions from riparian soil were high upstream of Cordoba City(S1 and S2,50.2-50.4 g/kg OM) and within a 50 km downstream location(S5,30.9 g/kg OM) owing to a surplus of fresh plant biomass-carbon(C) *** heterogeneous sediment samples did not show any significant differences among sites(P> 0.05).The lowest v.lues of the ratio of absorbances at 465 and 665 nm(E4/E6)(1.78) and the Δ log K(0.15) coefficient(a measure of HS maturity degree) were obtained downstream of Cordoba City,for both riparian soil and sediment,indicating that HS were enriched by more condensed aromatic structures within highly degraded portions of the *** samples exhibited similar IR spectra,implying ov.rlapping recalcitrant-C structures at the functional group lev.l,but with different absorbance *** from the current study constitute a baseline for understanding the chemical nature of HS from sediment and riparian soil along the Suquia Riv.r and can be used as a reference for future studies tracking OM compositional changes ov.r time.

A thrombin-like serine protease, jararassin-I, was isolated from the v.nom of Bothrops jararaca. The protein was obtained in high yield and purity by a single chromatographic step using the affinity resin Benzamidine-Sepharose CL-6B. SDS-PAGE and dynamic light scattering analyses indicated that the molecular mass of the enzyme was about 30 kD. The enzyme possessed fibrinogenolytic and coagulant activ.ties. The jararassin-I degraded the Bβ chain of fibrinogen while theAα chain and γ chain were unchanged. Proteases inhibitors, PMSF and benzamidine inhibited the coagulant activ.ty. These results showed jararassin- I is a serine protease similar to coagulating thrombin-like snake v.nom proteases, but it specifically cleav.s Bβ chain of bov.ne fibrinogen. Single crystals of enzyme were obtained (0.2 mm×0.2 mm×0.2 mm) and used for X-ray diffraction experiments.

Imperfect mimicry of host begging calls by a brood parasitic cuckoo:a cue for nestling rejection by hosts?

Coev.lutionary interactions between av.an brood parasites and their hosts often lead to the ev.lution of discrimination and rejection of parasite eggs or chicks by hosts based on v.sual cues,and the ev.lution of v.sual mimicry of host eggs or chicks by brood *** may also base rejection of brood parasite nestlings on v.cal cues,which would in turn select for mimicry of host begging calls in brood parasite *** cuckoos that exploit multiple hosts with different begging calls,call structure may be plastic,allowing nestlings to modify their calls to match those of their v.rious hosts,or fixed,in which case we would predict either imperfect mimicry or div.rgence of the species into host-specific *** our study of the little bronze-cuckoo(LBC)Chalcites minutillus and its primary host,the large-billed gerygone Gerygone magnirostris,we tested whether:(1)hosts use nestling v.calizations as a cue to discriminate cuckoo chicks;(2)cuckoo nestlings mimic the host begging calls throughout the nestling period;and(3)the cuckoo begging calls are plastic,thereby facilitating mimicry of the calls of different *** found that the begging calls of LBCs are most similar to their gerygone hosts shortly after hatching(when rejection by hosts typically occurs)but become less similar as cuckoo chicks get *** call structure may be used as a cue for rejection by hosts,and these results are consistent with gerygone defenses selecting for age-specific v.cal mimicry in cuckoo *** found no ev.dence that LBC begging calls were plastic.
