AIM: To analyze the relationship between perisinusoidal stellate cell (PSC) activation and the dietary fat quantity and composition in the treatment of hepatic ***: Using an experimental rat model of steatosis based on the intake of a hyperlipidic diet (14% fat as olive oil or sunflower oil, HL-O and HL-S, respectively), we analyzed the liver's capability of recovery after the treatment with a normal-lipidic diet (5% fat as olive oil or sunflower oil, NL-O and NL-S, respectively) by immunocytochemical and Western blot analysis of glial fibrillary acidic protein (GFAP) expression in PSCs, collagen quantification and serum aminotransferase ***: The fatty infiltration in the steatotic livers decreased after the treatment with both NL diets, indicating liver recovery. This decrease was accompanied with a lower collagen deposition and aminotransferase level as well as changes in the PSC population that increased the GFAP expression. The above-mentioned effects were more pronounced in animals fed on NL-O based diet. CONCLUSION: Treatment with a balanced dietenriched in olive oil contributes to the liver recovery from a stea totic process. The PSC phenotype is a marker of this hepatic-recovery model.

Epilepsy is a common,chronic neurological disorder that has been associated with impaired neurodevelopment and *** chemokine receptor CXCR5 is involved in seizures via an unknown ***,we first determined the expression pattern and distribution of the CXCR5 gene in the mouse brain during different stages of development and the brain tissue of patients with ***,we found that the knockdown of CXCR5 increased the susceptibility of mice to pentylenetetrazol-and kainic acid-induced seizures,whereas CXCR5 overexpression had the opposite ***5 knockdown in mouse embryos via viral vector electrotransfer negatively influenced the motility and multipolar-to-bipolar transition of migratory *** a human-derived induced an in vitro multipotential stem cell neurodevelopmental model,we determined that CXCR5 regulates neuronal migration and polarization by stabilizing the actin cytoskeleton during various stages of *** experiments demonstrated that the knockdown of CXCR5 induced neuronal hyperexcitability,resulting in an increased number of ***,our results suggested that CXCR5 deficiency triggers seizure-related electrical activity through a previously unknown mechanism,namely,the disruption of neuronal polarity.

Exosomes derived from mesenchymal stem cells(MSC-Exos)are nano-sized extracellular vesicles enriched with bioactive molecules,such as microRNAs,enzymes,cytokines,chemokines,immunomodulatory,trophic,and growth *** molecules regulate the survival,phenotype,and function of malignant and tumor-infiltrated immune *** to their nano-size and bilayer lipid envelope,MSC-Exos can easily bypass biological barriers and may serve as drug carriers to deliver chemotherapeutics directly into the tumor ***,we summarize current knowledge regarding molecular mechanisms responsible for MSC-Exos-dependent modulation of tumor progression and discuss insights regarding the therapeutic potential of MSC-Exos in the treatment of malignant diseases.

Detrimental immune response has a crucially important role in the development and progression of inflammatory eye *** mediators and proteolytic enzymes released by activated immune cells induce serious injury of corneal epithelial cells and retinal ganglion cell which may result in the vision *** stem cells(MSCs)are regulatory cells which produce various immunosuppressive factors that modulate phenotype and function of inflammatory immune ***,several safety issues,including undesired differentiation and emboli formation,limit clinical use of ***-derived exosomes(MSC-Exos)are nano-sized extracellular vesicles which contain all MSC-derived immunoregulatory *** administration of MSCExos efficiently attenuated eye inflammation and significantly improved visual acuity in experimental animals without causing any severe side *** cell-free product,MSC-Exos addressed all safety issues related to the transplantation of ***,MSC-Exos could be considered as potentially new remedy for the treatment of inflammatory eye diseases which efficacy should be explored in up-coming clinical trials.

Spent mushroom substrate(SMS)is an abundant residue generated by the mushroom industry,which is mostly *** this study,the SMS of four cultivated mushrooms were evaluated as potential sources of ligninolytic *** ostreatus,***,*** and Ganoderma lucidum were cultivated in two different substrates to determine growth,mushroom yields and ligninolytic activity of laccase(Lac)and manganese peroxidase(MnP).Maximum activities in the SMS were 3.731 U/g Lac and 0.191 U/g MnP,both for *** mushroom substrates were also analyzed,and higher activities were found in almost all samples.A correlation test between mushroom yields and ligninolytic activities of each culture was made,and we found that high enzymatic activity in the substrate did not result in high mushroom productivity or vice *** SMS was a good source of ligninolytic *** activity depended on the mushroom species and its affinity to the substrate,and it was not influenced by the mushroom *** of mushrooms in diverse lignocellulosic substrates is gaining interest,and ob-taining ligninolytic enzymes from residual SMS is an excellent alternative in the circular economy concept.

The transient receptor potential melastatin 2 is a calcium-permeable cation channel member of the TRP family. Also known as an oxidative stress-activated channel, the transient receptor potential melastatin 2 gating mechanism is dependent on reactive oxygen species. In pathological conditions, transient receptor potential melastatin 2 is overactivated, leading to a Ca~(2+) influx that alters cell homeostasis and promotes cell death. The role of transient receptor potential melastatin 2 in neurodegenerative diseases, including Alzheimer's disease and ischemia, has already been described and reviewed. However, data on transient receptor potential melastatin 2 involvement in Parkinson's disease pathology has emerged only in recent years and the issue lacks review studies that focus specifically on this topic. The present review aims to elucidate the role of the transient receptor potential melastatin 2 channel in Parkinson's disease by reviewing, summarizing, and discussing the in vitro, in vivo, and human studies published until August 2022. Here we describe fourteen studies that evaluated the transient receptor potential melastatin 2 channel in Parkinson's disease. The Parkinson's disease model used, transient receptor potential melastatin 2 antagonist and genetic approaches, and the main outcomes reported were discussed. The studies described transient receptor potential melastatin 2 activation and enhanced expression in different Parkinson's disease models. They also evidenced protective and restorative effects when using transient receptor potential melastatin 2 antagonists, knockout, or silencing. This review provides a literature overview and suggests where there is a need for more research. As a perspective point, this review shows evidence that supports transient receptor potential melastatin 2 as a pharmacological target for Parkinson's disease in the future.

In recent decades,life expectancy has been increasing *** this scenario,health interventions are ne-cessary to improve prognosis and quality of life of elderly with cardiovascular risk factors and cardiovascular ***,the number of elderly patients included in clinical trials is low,thus current clinical practice guidelines do not include specific *** document aims to review prevention recommendations focused in patients≥75 years with high or very high cardiovascular risk,regarding objectives,medical treatment options and also including physical exercise and their inclusion in cardiac rehabilitation ***,we will show why geriatric syndromes such as frailty,dependence,cognitive impair-ment,and nutritional status,as well as comorbidities,ought to be considered in this population regarding their important pro-gnostic impact.
