cellular radiosensitivity is directly correlated with the mechanism of DNA repair, in which p53 protein plays a major role. In this context, this study correlated cell death with p53 expression in lymphocytes irradiated in vitro with different doses of gammaradiation. For this, peripheral blood samples were collected from 10 healthy subjects. Each sample was divided in aliquots and, separately, irradiated with doses of 0,5;2 and 4 Gy. After this, peripheral blood mononuclear cells (PBMcs) were isolated and cultivated during 72 hours in 5% cO2 at 37oc without mitogen stimulation. The expression of p53 protein was evaluated by flow cytometry. In parallel, cell viability was determined by trypan blue staining. Statistical analysis was performed us-ing analysis of variance (ANOVA), differences were considered as statistically significant when p crease of p53 expression with the absorbed dose, which was proportional to cell death, suggesting that p53 can be used as bioindicator of individual radiosensitivity.

Objective: To evaluate the role of regulatory T cells (Tregs) at late stages of stroke. Methods:Anti-cD25 antibody (or PBS as a control) was injected to reduce the pool of Tregs in Wistar rats;then, ischemia was induced transiently by middle cerebral artery occlusion during 60 min and reperfusion was allowed for 7 d. Then, Treg frequency was analyzed in peripheral blood, spleen and lymph nodes. Neurological score (0-6) and infarct volume were also determined. Results: Nine days after injection, the cD4+cD25+ T cells were reduced by 70.4%, 44.8% and 57.9% in peripheral blood, spleen and lymph nodes, respectively compared to PBS-treated rats. In contrast, the reduction of cD4+FOXP3+ T cells was lower in the same compartments (38.6%, 12.5%, and 29.5%, respectively). The strongest reduction of cD25+cD4+ T cells was observed in those FOXP3-negative cells in blood, spleen and lymph nodes (77.8%, 52.8%, and 60.7%, respectively), most likely corresponding to activated T cells. Anti-cD25-treated transient middle cerebral artery occlusion rats had a lower neurological deficit and did not develop tissue damage compared with PBS-treated animals. conclusions: These findings suggest that treatment with anti-cD25 in our model preferentially reduce the T cell population with an activated phenotype, rather than the Treg population, leading to neuroprotection by suppressing the pathogenic response of effector T cells.

Long-Term Efficacy of Repositioning Maneuvers in Benign Paroxysmal Positional Vertigo

To determine the characteristics of 12 patients with a diagnosis of benign paroxysmal positional vertigo (BPPV) treated by the repositioning maneuver during a 24-month follow-up period after the initial discharge a longitudinal clinical study was performed in which a questionnaire for the assessment of discomfort caused by dizziness was applied at the first visit, at the last visit after treatment and 2 years after the initial discharge. Repositioning treatment was again offered to patients who suffered relapses. In 10 cases (83.34%) there was no recurrence of symptoms and 2 patients (16.66%) presented symptoms exactly 2 years after the original discharge showing that treatment of BPPV by repositioning manuevers is effective in the long term, but new studies in larger populations are needed to determine the pattern of the rates of recurrence of BPPV.

The microbiological, chemical and sensory characteristics of licensed and unlicensed commercial Serrano cheese, a traditional product from the highlands of South Brazil, were studied. The average bacteria counts (log cFU/g) were 8.8, 3.4, 3.4, 5.1, and 8.8, for mesophilic bacteria, fecal coliforms, staphylococci, yeasts and moulds, and lactic acid bacteria (LAB), respectively. Differences in microbiological quality were detected between licensed and unlicensed cheeses. However, the presence of potential gastroentheric bacteria in both kind of cheeses is disturbing and point out the necessity of alternative manufacturing process and good manufacturing practices to minimize consumers’ health risks. The most prevalent LAB were Lactobacillus (91%), followed Lactococcus and Enterococcus. Lactobacillus isolates comprised L. plantarum (57.7%), L. paracasei (19.2%), L. rhamnosus (7.7%), L. acidophilus (7.7%), and 3.8% of both L. curvatus and L. fermentum. Non-significant physico-chemical differences were detected between licensed and unlicensed cheeses. cheeses that received the highest sensorial scores exhibited the presence of both L. plantarum and L. paracasei, indicating that mixed populations of these species positively contributed to flavor development.

The Human Papillomavirus (HPV) is responsible for almost 99% of cervical cancers. cervical cancer is a public health problem, being the fourth most common type of tumor among women worldwide and the third most common cancer among women in Brazil. It is known that cervical cancer is one of the most easily preventable forms of cancer, as there is a highly effective vaccine against HPV, however, morbidity and mortality rates related to human papillomavirus are still frightening, especially in less developed countries. Low vaccination adherence rates, especially the second dose, are probably a reflection of the lack of knowledge about this virus, its clinical repercussions, and the vaccine. A clinical trial was carried out in a higher education institution in Alagoas (Brazil), with students of the medical course to whom a validated questionnaire was applied on HPV in the virtual environment (google forms). Data collection was carried out throughout the month of May of the year 2023. It was shown that the knowledge of medical students about the human papillomavirus was less than what would be desired for future doctors. The alert for the acute need to assess health illiteracy is clear, especially regarding cancer that can be prevented by a highly effective vaccine.

The platelet indices Mean Platelet Volume (MPV), Platelet Distribution Width (PDW) and Plate-letcrit (PcT) provided by automated hematology analysis are rarely used in clinical practice. Platelets play a central role in the pathogenesis of acute coronary syndromes (AcS) and high MPV has been associated to more reactive platelets and regarded as an independent risk factor for myocardial infarction. In this study, platelet indices were evaluated in 39 patients with AcS presenting two altered biochemical parameters, c reactive protein (cRP) and creatine kinase fraction MB (cK-MB). The results obtained showed significantly higher MPV and PDW values in the group with AcS in comparison to the control group. Also observed was a weak but significant positive correlation between MPV and cK-MB. In view of findings of previous studies, which have associated macroplatelets with higher thrombotic potential our results suggest that the use of the MPV and PDW indices as additional and complementary markers may contribute to the investigation and follow-up of thrombotic risk in patients with AcS.

To identify risk factors for A. baumannii infection in patients hospitalized in an Intensive care Unit (IcU) of a tertiary public hospital in Paraná, Brazil, a retrospective paired case-control study (ratio 1:2) was conducted from January 2018 to December 2020. Patients in the case group were hospitalized in the IcU with A. baumanni (n = 68 cases) and were compared with patients in the control group, without infection by A. baumannii (n = 136). Both were matched by age (±10 years), sex, and IcU stay (±5 days). conditional multiple logistic regression was used to determine statistically significant risk factors based on the results of bivariate analyses. Mortality was higher in infected (cases) than in non-infected patients (51.5% vs. 39.7%). The incidence and bacterial resistance increased annually. At bivariate analysis, cases had longer hospital stays (median 35 vs. 22 days, p A. baumannii and antimicrobial resistance. There is need for surveillance, and constant evaluation of control actions. Risk factors were colonization, previous hospitalization, and hospitalization time. This is essential for the decision-making of professionals and optimization of prevention, control, and therapeutic management actions.

Difficulties Faced by Physicians in the Diagnosis of Endometriosis in the Brazilian Public Health System

Endometriosis is a clinical and recurrent condition characterized by the presence of functional endometrial tissue outside the uterine cavity and myometrium. It is estimated that 6% to 10% of women of reproductive age, 50% to 60% of adolescents and adults with pelvic pain, and up to 50% of women with infertility are pregnant from the condition. However, this proportion tends to be even greater due to the difficulty in diagnosing it: because it presents different symptoms, lack of information from health professionals, and a high-cost diagnostic test. Another aggravating factor is the average time from the onset of symptoms to the medical diagnosis, which even today ranges from five to ten years;this delay has consequences for disease progression, as it prevents early treatment, which is important for improving pain levels and physical and psychological well-being. Aware of these mishaps, it is therefore important to assess the difficulties faced by doctors in the diagnosis of endometriosis in the public health system, in Brazil, the Unified Health System (SUS). With this purpose, a cross sectional was carried out, developed in the virtual environment using traffic in Google forms, with the difficulties quantified in the Likert scale. Participating physicians pointed out a medium difficulty for patients with endometriosis to access the consultation (58%) and moderate difficulty (52%) to perform an ultrasound examination;also signaled great difficulty in performing magnetic resonance imaging (77%) and laparotomy (61%) in these patients. It is hoped that knowledge about the difficulties faced by physicians in the diagnosis of endometriosis in the SUS will give these physicians a voice and corroborate, even if minimally, so that there is more research that provokes the search for these difficulties, in order to improving the quality of life of these women.

This study investigated the efficacy of calcium hydroxide and chlorhexidine gel for the elimination of intratubular candida albicans (***).Human single-rooted teeth contaminated with *** were treated with calcium hydroxide,2%chlorhexidine gel, calcium hydroxide plus 2%chlorhexidine gel,or saline(0.9%sodium chloride) as a positive *** samples obtained at depths of 0-100 and 100-200μm from the root canal system were analyzed for *** load by counting the number of colony forming units and for the percentage of viable *** using fluorescence ***,the antimicrobial activity of calcium hydroxide and the 2%chlorhexidine gel was evaluated by counting the number of colony forming *** 14 days of intracanal medication,there was a significant decrease in the number of *** colony forming units at a depth of 0-100μm with chlorhexidine treatment either with or without calcium hydroxide compared with the calcium hydroxide only ***,there were no differences in the number of colony forming units at the 100-200μm depth for any of the medications *** viability was also evaluated by vital staining techniques and fluorescence microscopy *** activity against *** significantly increased at both depths in the chlorhexidine groups with and without calcium hydroxide compared with the groups treated with calcium hydroxide *** with only chlorhexidine or chlorhexidine in combination with calcium hydroxide were effective for elimination of ***.

A Promising Strategy for the Treatment of Onychomycosis with curcumin and Photodynamic Therapy

Onychomycosis is a disease of high incidence in the nail plate and responsible for approximately half of the cases of nail infections. conventionally, dermatologists prescribe antibiotics and antifungals for long periods for its treatment. The high incidence of this type of infection and the increase of microbial strains resistant to the available drugs have justified the importance of the development of new technologies and treatments. This paper presents the photodynamic therapy as an alternative treatment of onychomycosis. New strategies for the use of curcumin as a photosensitizer and its therapeutic response were investigated in different formulations (gel and emulsion). Photodynamic therapy is a promising technique by which microorganisms are eliminated by a photosensitizing compound, light and oxygen. It was evaluated in two patients who had developed lesions in the fingernails caused by onychomycosis for approximately 10 years. The lesions were treated by photodynamic therapy with curcumin. The therapeutic efficacy was observed after a maximum of six photodynamic therapy sessions without any other adjuvant therapy. curcumin has become more promising than the therapeutic standard as it is a natural and versatible drug for incorporation in different formulations, at low cost and low probability of side effects.
