Aim:To investigate the effects of cisplatin on the human non-small cell lung carcinoma(NCI-H460)cell line regarding cytotoxicity,genotoxicity,and expression of genes associated with apoptosis(BIRC5)and autophagy(BECN1).Methods:Cell cultures were treated with cisplatin concentrations(0.16-33.3μmol/L)for 48 *** and acute and chronic cytotoxicities were assessed using the MTT,clonogenic,and cytokinesis-block micronucleus *** expression of BIRC5 and BECN1 was evaluated by reverse transcription-polymerase chain ***:Cisplatin IC50(0.33μmol/L)increased micronucleus frequency 2.50 *** was also cytotoxic in the 0.6-33.3μmol/L range,with reduced expression of the BIRC5 gene,suggesting induction of *** reducing the expression of the BIRC5 gene,33.3μmol/L cisplatin increased the expression of the BECN1 gene,suggesting that autophagy can be related to cisplatin ***:Cisplatin inhibited NCI-H460 growth,and cisplatin IC50 induced genotoxic *** higher cisplatin concentrations are used,the expression of genes associated with apoptosis and autophagy was *** results points to a further investigation of the role of autophagy in cisplatin resistance.

An Exploratory Behavioral Study in Children of Four Years of Age on the Hypothesis of a Two Streams System in Touch Perception and Language Development

A two streams system seems to exist for tactile *** asked kindergarten children to identify(semantic)and describe the function of grasped unseen instruments of their daily life(fork,knife,spoon,brush,toothbrush,pencil,and scissor),and then,to simulate its use(pragmatic).The capacity to describe the function of the set of instruments was significantly superior to that of identifying them by name,although there was a significant direct association between identification and description of the function in the responses to the set of *** simulation of the instrument function was of two types:(i)use―the child simulated how the instrument is grasped and used;and(ii)incorporation―the child simulated the function of the instrument with her/his own *** results support the hypothesis of a two streams tactile system(semantic and pragmatic),and that kindergarten children have the ability to incorporate functional properties of instruments of their daily life,probably as a support for language development.

Lichen planus affecting a 12-year-old girl

Lichen planus (LP) is a chronic inflammatory mucocutaneous condition which is relatively common in adults but rarely affects children. In addition, few cases with oral lesions affecting children have been reported in the literature. The purpose of this study is to report an unusual case of oral LP involving the tongue and buccal mucosa in a child. A 12-year-old girl complained of painless oral lesions with 18 months of evolution. On clinical evaluation, multiple white lesions in tongue and buccal mucosa were observed with no cutaneous lesions. Diagnosis was made based on clinical examination and histopathological features. No active treatment was necessary. The patient is currently under regular evaluation. Although rarely reported in childhood, lichen planus should be considered in a differential diagnosis of hyperkeratotic and/or reticular lesions of the oral mucosa in children.

Mescaline,among the earliest identified natural hallucinogens,holds great potential in psychotherapy ***,despite the existence of a postulated biosynthetic pathway for more than half a century,the specific enzymes involved in this process are yet to be *** this study,we investigated the cactus Lophophora williamsii(Peyote),the largest known natural producer of the phenethylamine *** employed a multi-faceted approach,combining de novo whole-genome and transcriptome sequencing with comprehensive chemical profiling,enzymatic assays,molecular modeling,and pathway engineering for pathway *** identified four groups of enzymes responsible for the six catalytic steps in the mescaline biosynthetic pathway,and an N-methyltransferase enzyme that N-methylates all phenethylamine intermediates,likely modulating mescaline levels in ***,we reconstructed the mescaline biosynthetic pathway in both Nicotiana benthamiana plants and yeast cells,providing novel insights into several challenges hindering complete heterologous mescaline *** together,our study opens up avenues for exploration of sustainable production approaches and responsible utilization of mescaline,safeguarding this valuable natural resource for future generations.

AIM: To elucidate the mechanism(s) by which S-adenosyl-L-methionine(SAM) decreases hepatitis C virus(HCV) ***: We examined the effects of SAM on viral expression using an HCV subgenomic replicon cell culture system. Huh7 HCV-replicon cells were treated with 1 mmol/L SAM for different times(24-72 h), then total RNA and proteins were isolated. c DNA was synthesized and real time-PCR was achieved to quantify HCV-RNA, superoxide dismutase 1 and 2(SOD-1, SOD-2) catalase, thioredoxin 1, methionine adenosyltransferase 1A and 2A(MAT1A, MAT2A) expression, and GAPDH and RPS18 as endogenous genes. Expression of cellular and viral protein was evaluated by western-blot analysis using antibodies vs HCV-NS5 A, SOD-1, SOD-2, catalase, thioredoxin-1, MAT1 A, MAT2 A, GAPDH and actin. Total glutathione levels were measured at different times by Ellman's recycling method(0-24 h). Reactive oxidative species(ROS) levels were quantified by the dichlorofluorescein assay(0-48 h); Pyrrolidin dithiocarbamate(PDTC) was tested as an antioxidant control and H2O2 as a positive oxidant ***: SAM exposition decreased HCV-RNA levels 50%-70% compared to non-treated controls(24-72 h). SAM induced a synergic antiviral effect with standard IFN treatment but it was independent of IFN signaling. In addition, 1 mmol/L SAM exposition did not modify viral RNA stability, but it needs cellular translation machinery in order to decrease HCV expression. Total glutathione levels increased upon SAM treatment in HCV-replicon cells. Transcriptional antioxidant enzyme expression(SOD-1, SOD-2 and thioredoxin-1) was increased at different times but interestingly, there was no significant change in ROS levels upon SAM treatment, contrary to what was detected with PDTC treatment, where an average 40% reduction was observed in exposed cells. 背景:中风作为一种疾病,中医早在2 000多年前即有诸多经典著作对其进行描述。然而,大部分有关针刺对中风后遗症患者功能恢复的疗效的临床对照试验未能得出针刺具有长期疗效的结论。并且,很多试验以表面肌电图测量患者骨骼肌的电活动以评定针刺刺激的即时疗效的结论也不一致。这些研究结果对针刺在神经肌肉水平的作用提出了质疑。本研究旨在比较徒手针刺影响健康人和中风后肌张力亢进患者的肱二头肌功能及力量的即时效果。方法与设计:本研究拟开展含有4个平行对照组的随机单盲临床试验。分别针刺健康受试者及中风后遗症痉挛性轻瘫患者的曲池(LI11)及天泉(PC2)2个穴位。表面肌电图测量针刺对于肌肉功能的即时疗效及肱二头肌的等长张力作为主要结局指标。次要结局指标包括每组中医证候的出现频率及频率分布。讨论:本研究的临床试验设计在方法学上较之前的类似试验有些改善。预计这一研究能够证实针刺刺激对健康人及中风后遗症患者的神经肌肉的作用。临床试验注册:巴西临床试验注册(***,注册号为RBR-5g7xqh).

Application of Geophysical Logging and Straddle Packers for the Investigation of a Fractured Aquifer in a Contaminated Area by Chlorinated Solvents in Sao Paulo State, Brazil

The investigation of contaminated areas in fractured aquifers represents a great technical challenge, due to the frequent conditions of heterogeneity and anisotropy of these environments, which often make it difficult to identify and predict pathways of subsurface contamination. This work aims to contribute to the development of this subject, through the presentation of the results of an investigation in which geophysical logging tools and straddle packers were used, which allowed the development of a more suitable hydrogeological conceptual model of the study area. Two boreholes were drilled and geophysical logging (gamma, caliper and high resolution acoustic televiewer) were used for the geological-structural characterization of the aquifer. Heatpulse flowmeter and straddle packers were then used to obtain data on hydraulic potentials and flows and for the collection of discrete water samples for chemical analysis. Two types of gneiss rocks were identified below the weathering zone (thickness >30 m), one with predominance of mafic bands, more weathered and with a higher fracture density, up to 65 m, and another deeper one, with the predominance of felsic bands. Seven groups of fractures were defined, being those of Group 1, with low dip angles, the most frequent and important for flow until 65 m, and Group 2 (N to NE-SW with high dips to W and NW) more frequent in deeper felsic gneisses. Downward hydraulic potentials were identified down to 65 m and upward potentials from the bottom up to 65 m. A hydraulic test allowed identifying the occurrence of hydraulic connection between the shallow weathering zone and the underlying fractured aquifer.

The genus Physalis belongs to the family Solanaceae,Physalis peruviana *** the most cultivated *** a newly explored crop in Brazil,little is known about its germination ecology and emergence under local *** aim of the present study was to determine the effect of temperature,photoperiod,planting depth and seed dormancy on the establishment of Physalis *** were conducted on two Physalis biotypes from Sert?o(Rio Grande do Sul state―RS)and Capelinha(Minas Gerais state).Seeds incubated at 27°C and 32°C exhibited high germination percentages of 98%and 96%,*** declined when seeds were incubated at 12,17,22 and 37°*** longer than 8 h are sufficient for seed germination,while the absence of light significantly reduces the germination *** should be planted at a depth of 1-2 cm to ensure better seedling *** 18 months of field work,81%of seeds had germinated and/or been predated prior to assessment,indicating low seed *** such,ideal conditions for Physalis seedling establishment are planting depth between 1 cm and 2 cm,temperatures ranging from 27°C to 32°C and a photoperiod longer than 8 h,with a postharvest seed shelf life of 12 months.
